Effects of the oral probiotic candidate Lactobacillus crispatus YIT 12319 on Candida albicans biofilm formation
Full text link研究成果の概要（和文）： Candida albicans ATCC 18804、Streptococcus mutans ATCC 25175およびStreptococcus sobrinus ATCC 33478に対するプロバイオティクス候補菌Lactobacillus crispatus YIT 12319のプロバイオティクス効果を調べた結果、L.crispatus YIT 12319は、C. albicansおよびS. mutansの増殖とC.albicans、S. mutansおよびS. sobrinusのバイオフィルム形成を抑制し、これらの微生物に対してプロバイオティクス効果を示すことが示唆された。研究成果の概要（英文）：Objective: The purpose of this study was to investigate the probiotic effect of Lactobacillus crispatus on Candida albicans and mutans streptococci. Design: Competition assays were employed to examine the growth suppression effects of the probiotic candidate L. crispatus YIT 12319 against C. albicans ATCC 18804, Candida glabrata ATCC 2001, Streptococcus mutans ATCC 25175, and Streptococcus sobrinus ATCC 33478. Furthermore, the formation of the biofilm composed of L. crispatus and the other microbe was monitored using the real time cell analyzer xCELLigence. Results: L. crispatus showed a strong and moderate inhibitory effect on the growth of C. albicans and S. mutans, respectively. L. crispatus inhibited the formation and maturation of biofilm of C. albicans, S. mutans or S. sobrinus. Conclusions: L. crispatus YIT 12319 was suggested to demonstrate the probiotic effect on C. albicans, S. mutans, and S. sobrinus.・2022(令和4)年度 科学研究費補助金 若手研究 研究成果報告書・研究期間 (年度)：2020-04-01 – 2023-03-31・研究分野：口腔細菌学・研究成果の学術的意義や社会的意義 : 近年、欧米諸国や日本を含む先進国諸国では医療の発展に伴い強い感染力を有する細菌由来の感染症は減少した。一方で高齢者や周術期の患者など免疫力が減弱した宿主への日和見感染症が問題になっている。世界に目を向ければ、COVID-19 患者の日和見感染が増加しているとの報告が増えている。重度COVID-19 患者の合併症として真菌感染症が増加しており、真菌のカンジダ属に関する基礎と臨床研究が再燃している。本研究ではプロバイオティクス候補菌を用いてカンジダ属やう蝕原性細菌のバイオフィルム形成と増殖を抑制する事に成功している。これを応用することで薬に頼らない治療法や予防の一助となる事が期待できる。研究課題/領域番号 : 20K18815research repor

Development of a digital workflow for fabrication of prosthetic devices using optimal structural design and laser sintering additive manufacturing
Full text link研究成果の概要（和文）： 可撤性有床義歯のフルデジタル製作を目指し，まずは義歯構成要素の中でも特に製作が困難なクラスプと義歯床に関して，臨床応用が可能となる加工条件と実際の維持力や適合精度を確認した．次に，高強度素材を用いた挙上副子と一般的な金属床パーシャルデンチャーのフルデジタル製作を試み，合理的な技工プロセスを探求しながら臨床応用の可能性を検証した．さらに本研究の最終目標であるフレームワークのトポロジー最適化に関しては，最もシンプルな加力計画の基に，CADとCAEを繰り返すことにより最適構造設計を行い，実際にCo-Cr合金粉末を用いてフレームワークを積層造形後，義歯として完成し口腔内に装着した．研究成果の概要（英文）：The purpose of this study was to develop the full digital fabrication method for removable dentures. Firstly, basic experiments were conducted on the processing accuracy of clasps and denture bases, which are difficulty fabrication in the denture components. The retentive force and fitting accuracy were confirmed. Next, full-digital fabrications of an occlusal splint using high-strength materials and a removable partial denture were attempted, and the possibility of clinical application was verified while exploring rational laboratory processes. As final goal of this study, the topology optimization of the framework, namely, the optimal structural design was performed by repeating CAD and CAE based on the simplest loading situation on the denture. After the framework was additively manufactured according to the topology 3D data using the Co-Cr alloy powder, overdenture was completed and delivered to the partially edentulous patient.・2022(令和4)年度 科学研究費補助金 基盤研究(C) 研究成果報告書・研究期間 (年度)：2020-04-01 – 2023-03-31・研究分野：歯科補綴学・研究成果の学術的意義や社会的意義 : 義歯の製作をフルデジタルで製作するにあたり，個々の義歯構成要素をデジタル製作による基礎的研究を実施し，最適な製作方法を模索した．その結果，支台装置と義歯床のデジタル製作に関する研究成果を報告した．デジタル技術を用いた臨床応用に対しては，まず通法では製作困難なグラスファイバー強化型レジンを用いた咬合挙上副子のデジタル製作を行った．また，デジタルワークフローで製作したパーシャルデンチャーの装着とトポロジー最適化によりデジタル製作したオーバーデンチャーの装着を試みた．この結果より，ジェネラティヴデザインとレーザー焼結積層造形を組み合わせたデジタルワークフローの臨床応用への道が開けた．研究課題/領域番号 : 20K10081research repor
