Analysis of Antibody Drug Conjugate on Cellular Senescent Human Glioblastoma
Background: Glioblastoma Multiforme (GBM) is an aggressive and malignant brain tumor known for its rapid progression and unfavorable prognosis. The standard treatment options primarily involve radiation and temozolomide, however recurrence is inevitable.
B7H3/CD276, an immunomodulatory transmembrane glycoprotein, demonstrates tumor-specific expression, with limited presence in normal healthy tissue, making it a promising and attractive treatment target in GBM. AbbV155 is an antibody drug conjugate that inhibits BCL-XL and specifically targets B7H3 (CD276) on GBM cells with the goal of selectively eliminating viable tumor cells.
Objectives: First, we sought to confirm the presence of B7H3/CD276 in our selected cell line via ELISA testing. Once the protein was successfully identified we assessed ABBV-155’s senolytic effect on radiation induced senescent GBM cells compared to non-radiated.
Methods: GBM 6 patient derived cells were cultured in vivo. Cells were allowed to grow until \u3e50% confluency was obtained before undergoing radiation at 15 gray. Cells were maintained in culture and treatment was started 9 days post-radiation with ABBV155. A dosage response curve was calculated using a 5-step 10-fold dilution. Cells were then maintained in drug treated media for 96 hours and a Keyence Microscope was used to quantify the number of viable cells.
Results: The dosage administered did not demonstrate significant efficacy between treated and non-treated cell lines.
Conclusions: Future experiments will seek to utilize an expanded dose-response curve. Undoubtedly, the contrast in B7H3/CD276 expression between tumors and healthy tissue remains a compelling prospect as a biomarker and therapeutic target. Notably, antibody-drug conjugates such as ABBV155 persists as encouraging treatment modalities in this regard
A Pilot Study Assessing the Efficacy of Amniotic Bladder Therapy in Spinal Cord Injury Patients with Female Sexual Dysfunction
Introduction and Objectives: Female sexual dysfunction (FSD) following spinal cord injury (SCI) presents with abnormalities in sexual desire, arousal, orgasm, pain, distress, and dissatisfaction. FSD negatively impacts quality of life and is often refractory to available treatment options. Previously, we reported preliminary benefits of intra-detrusor micronized amniotic membrane (AM) on detrusor dysfunction following SCI. In this study, we evaluated the effect of intra-detrusor micronized AM injection on FSD symptoms in SCI patients using the multi-domain female sexual function index (FSFI) and pain visual analog scale (VAS).
Methods: Five SCI females (mean age: 32 ± 12 years) with FSD recalcitrant to multiple therapies received intra-detrusor injections under general anesthesia of reconstituted 100mg micronized AM. The FSFI questionnaire and pain VAS were obtained preoperatively and at 4, 8, 12, 24, and 36-weeks post-injection.
Results: All five patients exhibited an improvement from baseline FSFI at 12 weeks. All patients relapsed at 24 to 36 weeks. Pain (VAS) associated with intercourse decreased from 8.4±0.9 at baseline to 7.1±0.8 at 4 weeks, 6.7±1.0 at 8 weeks, 6.3±1.2 at 12 weeks, 6.2±0.6 at 24 weeks and then increased to 7.9±0.6 at 36 weeks. No adverse events arose during the study.
Conclusion: ABT therapy in SCI females with severe FSD symptoms holds potential promise, although all patients experience symptomatic relapse at 24 to 36 weeks post-injection. More research is needed to better understand the mechanism through which ABT treats these complex sexual disorders
Assessing Mentor and Coordinator Professional Development in the Context of Medical School Learning Communities
Background Learning communities (LCs) create a bridge for senior students to engage with peers to provide advice and support. Anecdotally, LC leaders gain confidence in communication, empathy, and leadership - well received traits for residency. We aim to describe and quantify the growth of LC leaders over time.
Methods This study aims to elucidate an objective measure of student leader growth and development. We will evaluate opinions on the LC’s ability to foster professional and personal growth, and we will assess areas for improvement. Through Qualtrics self-rated queries using a Likert scale, we will survey 32 Coordinators from classes 2023-2026 and 24 M2 student mentors at various time-points in the year.
Results From the first two iterations of this research in 2022 and 2023, we found that most peer mentors acknowledged growth in leadership, empathy, and communication. Mentors prioritized building mentees’ confidence and mentee goal achievement but individually lacked confidence in facilitating these outcomes, possibly due to a lack of perceived support. We also assessed areas of improvement in the program, such as the perceived benefit of an educational intervention. Students responded that an optional curriculum would be most beneficial to allow them to develop skills they felt weak in.
ConclusionThis research presents a quality-assessment tool that will be repeated year-over-year to build longitudinal findings. We plan to incorporate optional educational interventions next year based on feedback from this year. These findings help us internally improve our LC program and validate the benefits of near-peer mentorship in medical education
Biological sex is a predictor of pretibial subcutaneous tissue depth for intraosseous catheter insertion
Introduction
Intraosseous (IO) vascular access is most commonly used when critical patients need rapid establishment of vascular access. They have shown high rates of successful placement, with the proximal tibia showing the highest first-attempt success rates. Proper establishment of vascular access requires a needle properly sized to enter the bony cortex and stay there. In this study, we analyzed demographic associations with pre-tibial subcutaneous tissue depth (PTSTD).
Methods
The PTSTD was calculated using computed tomography (CT) images of adult (³ 18 years old) patients. Variables including side, age, sex, height, weight, BMI, hypertension, diabetes mellitus, atherosclerosis, coronary artery disease, and osteoarthritis were analyzed statistically.
Results
368 patients were included in the final data analysis. Patient body mass index, height and weight showed a statistically significant impact on PTSTD overall, and betweenx \u3e 20 mm and \u3c 20 mm and \u3e 40 mm groups. Only height displayed a statistically significant effect between 40 mm \u3e x \u3e 20 mm and \u3e 40 mm group. Sex displayed a statistically significant effect on PTSTD.
Conclusions
Female sex and higher BMI appear to be related to increased soft tissue thickness in this patient population. Longer catheters may be needed for some obese patients, especially females

Tendon Rupture and Tendonitis in Low-Profile Dorsal versus Volar Plating for Distal Radius Fractures: A Systematic Review and Meta-Analysis
Background: Dorsal plating of distal radius fractures has historically been associated with high rates of hardware removal, tendonitis, and tendon rupture. Newer generation low-profile dorsal plates are thinner (\u3c1.6mm thick) with improved characteristics. We examine whether low-profile dorsal plates still have higher rates of tendon complications than volar locking plates.
Methods: We searched Ovid MEDLINE, Web of Science, and EMBASE for published literature describing tendon complications in association with plating of distal radius fractures. Inclusion criteria encompasses any primary study of low-profile dorsal plates that included data on tendon complications. Studies that included both low-profile dorsal and volar plating arms were included in the meta-analysis.
Results: Nine studies were selected for inclusion. All studies were retrospective cohorts or case series with a total of 446 low-profile dorsal plates. Five studies were included in the meta-analysis with a total of 806 subjects; 584 received volar plates and 222 received low-profile dorsal plates. Meta-analysis showed no significant difference in rates of tendonitis or tendon rupture.
Discussion: The included studies were all of level III or IV evidence. To our knowledge, this review provides the largest comparison of low-profile dorsal and volar locked distal radius plates to date. Pooled analysis results provide further comparison of low-profile dorsal versus volar plating

A Short-Term Longitudinal Study on the Moderating Effect of Sibling Trust on Internalizing Difficulties in Early Adolescence
Adolescents’ perceptions of their sibling relationship are important to their wellbeing, yet there is little work on the role of trust as a predictor of their adjustment. The present study investigated the effects of domain-specific facets of sibling trust on adolescent internalized adjustment. Participants included 169 early adolescents (51% girls) 10–13 years old with an older or younger sibling, who rated their perceptions of trust in that sibling and their depressed affect and intolerance of uncertainty across a 2-month period. Findings showed a positive association between reliability trust and honesty trust at T1. Results also indicated that reliability trust and honesty trust differentially moderated the stability of depressed affect. Specifically, higher levels of reliability trust attenuated the stability of depressed affect, whereas higher honesty trust appeared to strengthen it. Results emphasize the influence that sibling relationships, and trust between siblings, has on adolescent well-being. Findings are discussed in relation to theory and application to future interventions
