Optimization Protocol for Extraction and Purification of Recombinant Protein Modeled with Taq Polymerase
No full textThis experiment explores recombinant protein drug protocols and how to better optimize and purify large scale recombinant proteins expressed in bacterial cells. Using Escherichia coli bacteria, the gene Taq will be cloned into the bacterial plasmid, grown, induced, and incubated . Taq proteins will then be transcribed, lysed out of cells, and purified using an ion exchange chromatography column. By altering elements of this current process — varying the column flow rate and using different salt gradient concentrations for column purification — more concentrated yields were achieved with greater purity. It was found that lower flow rates of 100 mL/hour at normal salt gradient concentration of 25 to 700 mM produced the most optimal protein fractions. While Taq polymerase is not a recombinant protein drug, it can serve as a model to represent therapeutic protein drugs and purification optimizations that would work well for both. Utilizing these results, they can potentially provide the pharmaceutical industry novel ways to increase their production while lowering costs. Drug manufacturers would be able to produce these drugs at higher rates and more affordable prices, while saving on resources and time. Patients purchasing recombinant protein drugs would benefit from this as drugs become more widely accessible and reasonably priced. Educational Leadership In Independent Schools
No full textIndependent schools provide a unique environment from other educational systems. By circumventing state and federal funding, they are able to cultivate autonomy for curriculum choices, management decisions, and independence not known in public schools. The educational leaders of independent schools may require different leadership skills than other school settings due to these additional possibilities and responsibilities. Through a phenomenological lens, the study uncovered skills utilized by four experienced independent school heads. The study used full-range leadership theory to further interpret the lived experiences of the school heads. The purpose of the inquiry is two-fold. First, according to the review of literature, most independent school leaders felt unprepared to lead when they accepted their first leadership position. Graduate level leadership programs tend to prepare leaders for the majority of positions in public schools. The preparation of independent school leaders could impact the sustainability of these schools. The second reason for the inquiry is the lack of research that has taken place in independent school leadership. This study uses qualitative methods of interviews to gain a rich description through a phenomenological lens of the journey of an independent school leader as well as insight into the skills needed to lead an independent school. Emerging themes such as constancy of purpose, relational approach, personality, and decision making that drove the ability to perform many of the transformational leadership skills. How Much Do Faculty Think Students Should Pay for Course Materials? A Survey of Instructors' Use of Current Course Materials and OER Use
No full textIntroduction: Campus affordability initiatives promote the adoption of free, low-cost, and open educational materials. Coordinators first need to understand faculty usage of instructional course materials, textbook selection authority, and their price tolerance for, knowledge of, and use of open educational resources (OER). Methods: Faculty at a medium-sized research university in the Midwest were invited to participate in an online survey about their current use of instructional course materials, their knowledge of material cost, and student purchasing habits. The survey had a completion rate of 128 responses, representing all teaching ranks on campus. Results: Instructors assign a wide range of materials and rarely hear from students about their inability to purchase textbooks. The majority find US$100 or more an acceptable price and believe that more than 70% of students purchase all required items. Over half of respondents make the final decision about their materials, and factually accurate content is the top priority when making selections. Eighty percent have some awareness of OER, but OER use is low. Discussion: Response rate was too low to make general assumptions but suggests areas for further exploration and provides base data when working with departments. A faculty member’s selection authority could be limiting OER use and impacting students. Discussions need to take place about an acceptable price for department courses and how faculty decide to require materials. Conclusion: Outreach to campus partners should promote high-quality materials as instructors consider this the most when selecting resources. Further examination of how selection authority impacts OER use and student success need to occur
