Alternative Feeding Resources of Mediterranean Origin to Improve Lamb Meat and Pork Quality
The general objective of this doctoral research was to determine the effects of alternative feeding resources, which are of Mediterranean origin used in lamb and pig production, on meat quality. In the first and second experiments, the lambs were grouped into three to receive different dietary treatments: a barley-based concentrate diet, or two different levels of dried citrus pulp (24% or 35%) to replace barley in concentrate. The meat were stored aerobically for six days at refrigerated temperature, afterwards the colour stability and lipid oxidation (first experiment), and microbial spoilage (second experiment) were evaluated in the muscle longissimus dorsi. Dried citrus pulp strongly reduced meat lipid oxidation, while did not exert effects on colour stability and on microbial growth in meat over storage duration. In the third experiment, the pigs were grouped under dietary treatments concentrate-based finishing diet or two levels of carob pulp (8% and 15%) used to replace concentrate. The lipid oxidative stability and lipid composition were studied in the longissimus thoracis et lumborum. The use of carob pulp as concentrate replacer did not improve the lipid oxidative stability of pork but increased the levels of polyunsaturated fatty acids in meat as compared to concentrate fed pig meat. The findings demonstrate that the dietary citrus pulp or carob pulp could be potentially used as alternative feeds to obtain quality meat. Specifically, the presence of citrus pulp in lamb diets resulted in meat of higher resistance to lipid oxidation, while the inclusion of carob pulp improved the healthy fatty acid contents in pork that is increased in polyunsaturated fatty acids and decreased saturated fatty acids
Redox proteomics, thiol homeostasis and neurophysiologicla correlations in aging and neurodegeneration
The terms "aging" and "neurodegeneration" are often used in a broad and generalized manner. Actually, they are particularly complex and multifaceted processes, involving different biochemical systems.
Increasing evidence supports the notion that reduction of cellular expression and activity of antioxidant proteins and the resulting increase of oxidative stress are fundamental causes in the aging processes and neurodegenerative diseases. Within the frame of free radical hypothesis of aging, several lines of evidence suggest that accumulation of oxidative molecular damage is a causal factor in senescence. It is also increasingly evident that the mitochondrial genome may play a key role in aging and neurodegenerative diseases. Mitochondrial dysfunction is characteristic of several neurodegenerative disorders, and evidence for mitochondria being a site of damage in neurodegenerative disorders is partially based on decreases in respiratory chain complex activities in Parkinson s disease (PD), Alzheimer s disease (AD), and Huntington s disease (HD). Such defects in respiratory complex activities, possibly associated with oxidant/antioxidant balance perturbation, are thought to underlie defects in energy metabolism and induce cellular degeneration. Efficient functioning of mantainance and repair process seems to be crucial for both survival and physical quality of life. This is accomplished by a complex network of the so-called "longevity assurance processes", which are composed of several genes, termed vitagenes. Among these, heat shock proteins, also known as stress proteins and molecular chaperones, are highly conserved proteins for the preservation and repair of the correct conformation of cellular macromolecules, such as proteins, RNAs and DNA. Chaperone-buffered silent mutations may be activated during the aging process and lead to the phenotypic exposure of previously hidden features and contribute to the onset of multigenic diseases, such as age-related disorders, atherosclerosis and cancer. Recent studies have shown that the heat-shock response contributes to establishing a cytoprotective state in a wide variety of human diseases, including ischemia and reperfusion damage, inflammation, metabolic disorders, cancer, infection, trauma, and aging. The major neurodegenerative diseases, Alzheimer s disease (AD), Parkinson s disease (PD), amyotrophic lateral sclerosis (ALS), Huntington s disease (HD), and Friedreich s ataxia (FA), are all associated with the presence of abnormal. Given the broad cytoprotective properties of the heat-shock response, there is now strong interest in discovering and developing pharmacological agents capable of inducing the heat-shock response. These findings have opened up new perspectives in medicine and pharmacology, as molecules inducing this defense mechanism appear to be possible candidates for novel cytoprotective strategies. Particularly, modulation of endogenous cellular defense mechanisms such as the heat-shock response, and the proteasomal system, through nutritional antioxidants or pharmacological compounds may represent an innovative approach to therapeutic intervention in diseases causing tissue damage, such as neurodegeneration. Moreover, by maintaining or recovering the activity of vitagenes, it would be possible to delay the aging process and decrease the occurrence of age-related diseases with resulting prolongation of a healthy life span

With the beginning of the Information Age and the following spread of the information overload phenomenon, it has been mandatory to develop a means to simply explore, analyze and summarize large quantity of data. To achieve this purposes a data mining techniques and information visualization methods are used since decades.
In the last years a new research field is gaining importance: Visual Analytics, an outgrowth of the fields of scientific and information visualization but includes technologies from many other fields, including knowledge management, statistical analysis, cognitive science and decision science. In this dissertation the combined effort of the mentioned research fields will be analyzed, pointing out different way to combine them following the best practice according to several application cases
Acting as therapeutic support in Parkinson's disease. A Field experiment
In the past, the clinical approach to Parkinson s disease focussed primarily on motor symptoms, whereas the current trend emphasises the importance of non-motor symptoms, thus moving towards a more complex type of assistance aimed at enhancing each patient s residual ability and, as far as possible, at rehabilitating their impaired daily activities. So, there is a growing amount of artistic activities that appear to positively influence the patients emotional and relational sphere as well as their and their family s quality of life (QoL).
Over a period of 8 months, a group of patients with a moderate form of idiopathic Parkinson s disease, on stable L-dopa and L-dopa antagonists, together with a few family members, took part in a weekly theatre workshop culminating in a show given to the public. The patients were evaluated both before and at the end of the eight-month workshop using eight Assessment Scales indicating the state of progression of the disease (H&Y, UPDRS, PDQ8, PDSS, NMSS, FES, GFQ and MMSE). The results of these assessments were then compared to verify if the theatre workshop had had any influence and if so to what extent. The result analysis showed that modest improvements were associated with movement parameters, while parameters regarding memory, attention and concentration, quality of sleep and especially relational aspects were affected in a statistically significant manner, allowing us to say that the patients quality of life (QoL) clearly improved thanks to acting
Linking Exploration and Exploitation to the Venture Capital Industry. An investigation on the rationales adopted, decision-making, and performance
The aim of this dissertation research is to link exploration and exploitation rationale to the Venture Capital industry, understanding the measurement dimensions and the effects on both decision-making and performance of VCs.
Three studies constitute the body of the empirical research in this dissertation. Each study is focused on specific structure of relationships from the research framework with a twofold aim. First, frame exploration and exploitation orientation in a novel context of investigation - the VC industry - and find evidence for the impact of a more explorative orientation (vs a more exploitative orientation) on decision-making and performance in VC companies. Second, make specific contributions to streams of strategic management and entrepreneurship literature, on the one side, and practice, on the other side.
In the first study we conceptualize exploration and exploitation orientation in the VC context, framing exploration and exploitation in terms of greater versus narrower search for knowledge resources in non-local domains.
The second study applies the measurement instrument proposed in the first study to test to what extent exploration and exploitation orientation impact on decision-making performance.
The third study explores the impact of exploration orientation on VC performance under high levels of uncertainty.
A quantitative approach is adopted to conduct the empirical investigation. To empirically test the hypotheses of the first two studies we managed a survey sent to 90 Canadian venture capitalists. Data collected have been analyzed through a multivariate methodology. Furthermore, to empirically test for hypotheses developed in the third study we collected data from the Thomson One Banker Database and ran the analysis on a dataset consisting of over 1.400 VC investments. Results have been presented and discussed.
Implications are derived for both theory and practice. Limitations of the study and future research are outlined
Effetto degli inibitori delle tirosino chinasi nelle cellule di carcinoma poco differenziato della tiroide
Premessa. I carcinomi poco differenziati della tiroide sono refrattari ai trattamenti convenzionali per cui negli ultimi anni si sono condotti diversi studi finalizzati ad identificare le alterazioni molecolari di più frequente riscontro e che potrebbero essere responsabili della scarsa efficacia delle terapie classiche. Bersagli molecolari interessanti sono: il recettore per l EGF e quello per l IGF-I poiché entrambi iperespressi nel carcinoma tiroideo. Di recente, le case farmaceutiche hanno sintetizzato vari inibitori molecolari rivolti verso entrambi i recettori, per esempio il Gefitinib inibitore di EGFR e l NVP in grado di bloccare IGF-IR. Tuttavia, gli studi preclinici condotti finora hanno dato risultati poco soddisfacenti, in termini di efficacia antitumorale, per la possibile insorgenza di meccanismi di resistenza.
Oggetto dello studio. In questo studio, su una serie di linee cellulari di carcinoma tiroideo, abbiamo valutato i meccanismi molecolari di resistenza al blocco farmacologico di EGFR ed IGF-IR, usando come inibitori il Gefitinib e l NVP.
Risultati 1 (Inibizione di EGFR): Valutando l espressione di EGFR, mediante western blot, è stato visto che le linee di carcinoma tiroideo analizzate iper-esprimevano tale recettore e che il Gefitinib era in grado di inibire la sua fosforilazione. Contrariamente, questo farmaco si è mostrato scarsamente efficace nella riduzione della proliferazione delle cellule neoplastiche e della fosforilazione di ERK, mediatore a valle di EGFR. Questo dato è stato ottenuto nella maggior parte delle linee cellulari testate, ad eccezione delle linee WRO e Hth-74 che sono risultate sensibili al farmaco. Il Gefitinib non è risultato efficace in tutte quelle linee cellulari recanti mutazioni dei geni che portano all attivazione costitutiva della via di ERK, tra cui BRAF (V600E), HRAS (G12A/Q61R) o riarrangiamento RET/PTC1. Alla luce di ciò, in questo studio abbiamo valutato gli effetti dell inibizione dell oncogene BRAF(V600E) e di RET sulla sensibilità delle cellule di carcinoma tiroideo al Gefitinib. Abbiamo trovato che l inibizione di BRAF, mediante l inibitore selettivo PLX4230, ripristina gli effetti del Gefitinib in tutte le cellule in possesso della mutazione del BRAF. Risultati simili sono stati ottenuti con inibitori di RET.
Risultati 2 (Inibizione di IGF-IR): Dopo aver valutato gli effetti antiproliferativi dell NVP-AEW541 in un gruppo di linee cellulari tumorali tiroidee abbiamo selezionato linee sensibili e resistenti. In tutte le linee, sia sensibili che resistenti, l NVP ha inibito la fosforilazione di IGF-IR. Nelle cellule sensibili, l NVP ha esercitato effetti antiproliferativi ed è stato in grado di inibire la via di segnale dipendente da AKT (pAKT) in maniera più efficace rispetto alla via di ERK1/2. Viceversa, nelle linee resistenti si è osservato un aumento di attivazione di ERK1/2 dopo NVP e nessuna inibizione di pAKT. Nelle FF1 aventi un alto rapporto IR:IGF-IR, abbiamo trovato che EGFR viene attivato in seguito al trattamento con NVP. Questo dato non è stato riscontrato nelle C643, le quali esprimono alti livelli di IGF-IR e bassi livelli di IR. Pertanto, abbiamo valutato l efficacia di un trattamento combinato volto ad inibire sia IGF-IR che EGFR o le vie a valle di esso quali MAPK. Abbiamo trovato che l inibizione di IGFIR associata a quella di EGFR o di ERK, è in grado di superare i meccanismi di resistenza all inibizione singola di IGF-IR.
Conclusioni. Questi risultati indicano che in alcune linee di carcinoma tiroideo la resistenza al Gefitinib è dovuta principalmente alla mutazione del BRAF(V600E) e che tale resistenza può essere superata utilizzando il Gefitinib in combinazione con inibitori del BRAF.
Inoltre, l iperespressione di IR e di EGFR possono entrambi contribuire alla resistenza dell'inibizione di IGF-IR. Una terapia combinata con altri farmaci molecolari mirati, come inibitori di EGFR o MAPK potrebbero superare questi meccanismi di resistenza

Con il presente lavoro abbiamo cercato di ricostruire e contestualizzare, attraverso anche il reperimento di materiale archivistico e bibliografico inedito, la figura dell intellettuale siciliano non vedente Vincenzo Tedeschi Paternò Castello (1786-1858).
La tesi è articolata in tre capitoli. Il primo è dedicato a una presentazione generale della vita e delle opere di Vincenzo Tedeschi Paternò Castello, come pure del contesto storico italiano ed europeo nel quale il nostro visse ed operò. Il secondo capitolo ripercorre le tappe della travagliata carriera universitaria e il percorso scientifico di uno studioso aperto a quasi tutte le branche del sapere (dal diritto naturale alla filosofia, dall anatomia alla fisica e alla matematica). Nel terzo capitolo, infine, si ricostruisce il coinvolgimento di Tedeschi nell attività politica e amministrativa catanese.

Chronobiological plasticity in Honeybee and its association with differences in the organization of the molecular pathway between central and peripheral clocks: the brain and flight muscle tissues.
Circadian rhythms govern the behavior, physiology, and metabolism of living organisms, enabling them to anticipate changes of environmental conditions. Within Insects, in honey bee s colonies an ontogeny in circadian rhythms of behavior was suggested to be related to an age- based division of labor, to best serve their social organization: young adult tend to perform tasks in the nest, nursing brood around the clock, with no circadian rhythms for the 2 3 first weeks, and then shift to foraging outside for the remainder period of life, enabling visiting flowers at time of maximal nectar and pollen availability. In addition to light, social cues is one of the primary zeitgeber that entrain the locomotor activity rhythm in honeybees, as notably showed by the robustness of circadian activity of young nurses soon after their release far from social interactions with the conspecifics. Such context dependent plasticity in circadian behavior was often associated with a variation in the main core clock genes mRNA expression profile in the brain, but it has not been yet examined whether such variations extend to other body tissues of the honeybee. In this thesis, I identified for the first time oscillating transcription of core clock genes in the flight muscle of workers, which temporal cycling pattern differed significantly in LD and under DD, in a task rather than age dependent manner. Although the autonomy of such oscillator from central orchestration has yet to be determined, the consistent temporal cycling pattern of mRNA level enriched for genes highly involved in muscle metabolism, suggest exciting future opportunities to discover additional clock controlled genes in the honeybee. The ensemble of progresses, might give answer to the kind of relationships between different clocks, involving eventually total and partial autonomy, especially in such an excellent model in chronobiology: the Honeybee
Archeologia del territorio tra Siracusa e Catania in età romana e medievale
Le ricerche condotte nel corso dell ultimo decennio nell area megarese (Augusta, Melilli e Priolo Gargallo, SR) hanno permesso di delineare chiaramente lo sviluppo e le trasformazioni del paesaggio rurale tra l età romana e il Medioevo. I risultati più interessanti sono stati evidenziati per il Tardoantico e l Altomedioevo grazie alle recenti acquisizioni e seriazioni delle ceramiche. Il quadro che è stato delineato ha permesso di evidenziare una generale continuità di vita tra l età repubblicana e il IV secolo e tra il VII e il IX secolo, con limitati fenomeni di abbandono, mentre è chiara una cesura avvenuta nel V secolo e, successivamente tra il IX e il X secolo, quest ultimo certamente a causa dello scontro bellico tra Bizantini e Arabi, con l abbandono di gran parte degli insediamenti del territorio. Parallelamente, lo sviluppo degli insediamenti rupestri, con la funzione di siti-rifugio, conferma da un lato l incertezza di questa fase, ma parallelamente gli insediamenti costieri indicano una continuità di vita che si protrae senza cesure fino al IX secolo. Proprio su questo punto sono state concentrate le ricerche che hanno sottolineato, come in altri contesti del Mediterraneo, che tra VIII e IX secolo gli scambi sulla lunga distanza proseguirono, certo su scala minore, e non furono interrotti dalle vicende belliche
