Facile fabrication and characterization on alginate microfibres with grooved structure via microfluidic spinning
Alginate microfibres were fabricated by a simple microfluidic spinning device consisting of a coaxial flow. The inner profile and spinnability of polymer were analysed by rheology study, including the analysis of viscosity, storage modulus and loss modulus. The effect of spinning parameters on the morphological structure of fibres was studied by SEM, while the crystal structure and chemical group were characterized by FTIR and XRD, respectively. Furthermore, the width and depth of grooves on the fibres was investigated by AFM image analysis and the formation mechanism of grooves was finally analysed. It was illustrated that the fibre diameter increased with an increase in the core flow rate, whereas on the contrary of sheath flow rate. Fibre diameter exhibited an increasing tendency as the concentration of alginate solution increased, and the minimum spinning concentration of alginate solution was 1% with the finest diameter being around 25 mm. Importantly, the grooved structure was obtained by adjusting the concentration of solutions and flow rates, the depth of groove increased from 278.37 +/- 2.23 mu m to 727.52 +/- 3.52 mu m as the concentration varied from 1 to 2%. Alginate fibres, with topological structure, are candidates for wound dressing or the engineering tissue scaffolds.journal articl
Raman Laser Switching Induced by Cascaded Light Scattering
It is shown that, in multimode Raman lasers, cascaded light scattering (CLS) not only extends the optical frequency range, but can also modulate the laser dynamics. The origin of this phenomenon lies in the fact that many Raman lasing modes are directly correlated through CLS. The coupled‐mode equations only describe single‐mode cascaded Raman lasers and are insufficient for describing the multimode case. In this work, additional terms are introduced to account for intermodal interaction and, therefrom the physical mechanism behind the mode‐switching phenomenon is revealed. Additionally, mode‐switching controlled solely by a single‐mode pump in a whispering gallery mode (WGM) silica Raman laser is demonstrated. As the intracavity pump power is increased, laser switching happens between two adjacent WGMs in the same mode family.journal articl
Taxonomy of Arabian Temnothorax Mayr (Formicidae: Myrmicinae) with description of a new species enhanced by x-ray microtomography
Temnothorax elmenshawyi sp. n., a new ant species from the Asir Mountains of the southwestern region of the Kingdom of Saudi Arabia, is described based on the worker caste. The new species is a member of the T. exilis species group and is distinguished from the other species included in this group by the impressed metanotal groove, the short, acute and broadly-based propodeal spines, the finely punctate posterior half of cephalic surface, and absence of a median clypeal carina. Despite extensive collecting by the authors at the type locality, only two specimens are available for description, suggesting that this species may be rare and likely endemic to the Asir Mountains. The species description is complemented by still images of volume renderings of a 3D model and a 3D rotation video of the holotype based on x-ray microtomography (micro-CT), allowing remote in-depth examination of the specimen. Extended Temporal Association Memory by Modulations of Inhibitory Circuits
Hebbian learning of excitatory synapses plays a central role in storing activity patterns in associative memory models. Interstimulus Hebbian learning associates multiple items by converting temporal correlation to spatial correlation between attractors. Growing evidence suggests the importance of inhibitory plasticity in memory processing, but the consequence of such regulation in associative memory has not been understood. Noting that Hebbian learning of inhibitory synapses yields an anti-Hebbian effect, we show that the combination of Hebbian and anti-Hebbian learning can significantly increase the span of temporal association between correlated attractors as well as the sensitivity of these states to external input. Furthermore, these effects are regulated by changing the ratio of local and global recurrent inhibition after learning weights for excitation-inhibition balance. Our results suggest a nontrivial role of plasticity and modulation of inhibitory circuits in associative memory.journal articl
Nuclear AGO1 Regulates Gene Expression by Affecting Chromatin Architecture in Human Cells
The impact of mammalian RNA interference components, particularly, Argonaute proteins, on chromatin organization is unexplored. Recent reports indicate that AGO1 association with chromatin appears to influence gene expression. To uncover the role of AGO1 in the nucleus, we used a combination of genome-wide approaches in control and AGO1-depleted HepG2 cells. We found that AGO1 strongly associates with active enhancers and RNA being produced at those sites. Hi-C analysis revealed AGO1 enrichment at the boundaries of topologically associated domains (TADs). By Hi-C in AGO1 knockdown cells, we observed changes in chromatin organization, including TADs and A/B compartment mixing, specifically in AGO1-bound regions. Distinct groups of genes and especially eRNA transcripts located within differentially interacting loci showed altered expression upon AGO1 depletion. Moreover, AGO1 association with enhancers is dependent on eRNA transcription. Collectively, our data suggest that enhancer-associated AGO1 contributes to the fine-tuning of chromatin architecture and gene expression in human cells.journal articl
Dynamic stereoselective annulation via aldol-oxa-cyclization cascade reaction to afford spirooxindole pyran polycycles
Spiro polycyclic compounds bearing pyran ring systems are found in bioactive molecules, and we recently reported the construction of spirooxindole all-carbon polycycles. Here we show the development of catalytic stereoselective annulation reactions that afford spirooxindole pyran polycycles. Oxindole-derived spiro[4,5]decanes are reacted with arylglyoxal to construct a pyran ring via the formation of carbon-carbon and carbon-oxygen bonds through dynamic aldol-oxa-cyclization cascade reactions, leading to the formation of spirooxindole pyran polycycles bearing six stereogenic centers as single diastereomers. During the reaction, the starting material is isomerized to the diastereomer, and this is key to afford the product. Taking advantage of this isomerization, highly enantiomerically enriched single diastereomers of spirooxindole pyran polycycles are obtained. The reactions generating the spiro pyran polycycles show stereoselectivities distinct from those previously observed in the construction of all-carbon polycycles.journal articl
Multi-dimensional computational pipeline for large-scale deep screening of compound effect assessment: an in silico case study on ageing-related compounds
Designing alternative approaches to efficiently screen chemicals on the efficacy landscape is a challenging yet indispensable task in the current compound profiling methods. Particularly, increasing regulatory restrictions underscore the need to develop advanced computational pipelines for efficacy assessment of chemical compounds as alternative means to reduce and/or replace in vivo experiments. Here, we present an innovative computational pipeline for large-scale assessment of chemical compounds by analysing and clustering chemical compounds on the basis of multiple dimensions—structural similarity, binding profiles and their network effects across pathways and molecular interaction maps—to generate testable hypotheses on the pharmacological landscapes as well as identify potential mechanisms of efficacy on phenomenological processes. Further, we elucidate the application of the pipeline on a screen of anti-ageing-related compounds to cluster the candidates based on their structure, docking profile and network effects on fundamental metabolic/molecular pathways associated with the cell vitality, highlighting emergent insights on compounds activities based on the multi-dimensional deep screen pipeline.journal articl
