Comparison of Two Bacillus Strains Isolated from the Coastal Zone in Barley (Hordeum vulgare L.) Under Salt Stress

    No full text
Salt stress is one of the most important abiotic stress factors that negatively affects sustainable crop production, agricultural productivity, and microbial life. Increasing salt stress negatively affects the growth and development of barley, posing a threat to global food security. It is now known that inoculation of plant growth-promoting rhizobacteria (PGPR) has significant potential in increasing stress tolerance and yield in agricultural products. This study focused on the effects of Bacillus cereus CUN6 and Bacillus thuringiensis SIRB2, isolated from the coastal zone and tested for their PGPR capacities, on physiological (root length, shoot length, biomass, dry weight) and biochemical (total chlorophyll, total protein, hydrogen peroxide, lipid peroxidation, peroxidase activity (POX), catalase activity (CAT)) analyses in Hordeum vulgare L. seedlings under salt stress. The results showed that the two bacterial inoculations alleviated the negative effects of salt stress by increasing the root-shoot length, biomass, dry weight, chlorophyll content, and total protein content in barley plants. However, B. thuringiensis increased growth and development especially in root length, biomass, and dry weight compared to B. cereus. On the other hand, B. cereus significantly increased root length, biomass, and chlorophyll content under salt stress; these increases were 17%, 5%, and 7%, respectively. B. thuringiensis chlorophyll content increased by 4% in 300 mM NaCl compared to the control. When compared in terms of the antioxidant defense system, B. thuringiensis inoculation was more effective on CAT activity, while B. cereus inoculation was more effective on POX activity. Under salt stress, B. cereus and B. thuringiensis inoculation significantly decreased H2O2 content in barley; these decreases were 16% and 10%, respectively. Additionally, TBARs content was significantly decreased by B. cereus and B. thuringiensis inoculation under salt stress; these decreases were determined as 8% and 9%, respectively, compared to the control. These results indicated that both bacterial inoculations can alleviate the salt tolerance of barley seedlings by regulating antioxidant metabolism. This research focused on the potential of B. cereus and B. thuringiensis as biofertilizers against salt stress in barley based on physiological and biochemical analysis.Canakkale Onsekiz Mart University project [FDK-2021-3684]; Scientific and Technological Research Councilof Turkiye (TUEBITAK) [2211-A]We would like to thank the Canakkale Onsekiz Mart University project (FDK-2021-3684) for their support. This article is under the CC BY license (https://creativecommons.org/licenses/by/4.0/).This study investigates the ideological orientations of teachers and the potential transformation of these orientations over 14 weeks, focusing specifically on their conceptualizations of educational ideologies within conservative and liberal frameworks. The research employs an inventory-based assessment to capture participants’ ideological stances. Initial findings revealed participants’ tendency to associate ideological labels with political dualities, reflecting simplified views on educational ideologies. Throughout the study’s second phase, participants engaged in critical reading assignments and reflective discussions designed to expand their awareness and understanding of various educational ideological paradigms. Findings indicate a notable shift in ideological positioning, primarily driven by exposure to foundational texts and structured reflection sessions. In the final phase, participants re-took the initial inventory, allowing for a direct comparison of ideological stances. The comparative analysis demonstrated subtle yet significant shifts, particularly regarding participants’ views on the interrelations between educational ideology, social equity, and the intellectual demands of modern pedagogy. This study underscores the transformative potential of immersive engagement with diverse ideological perspectives, highlighting the need for professional development models that prioritize reflective dialogue and critical exploration of ideological content

A Dynamic Analysis of Cosmic Strings in the f(R,T) Gravity Model

    No full text
This study investigates the dynamics of cosmic strings within the framework of the f(R,T) gravity model, a modified theory of gravity proposed by Harko et al. in 2011. The f(R,T) theory extends Einstein’s general relativity by including functions of the Ricci scalar (R) and the trace of the energy-momentum tensor (T). Using the Friedman-Robertson-Walker (FRW) universe model, the study derives the field equations and explores the behavior of the key cosmological parameters, such as the scale factor, string energy density, cosmological term, deceleration parameter, and Hubble parameter. The analytical solutions show that the universe expands continuously with the increasing scale factor values. The study also demonstrates that energy density and the cosmological term decrease over time, while the Hubble parameter exhibits a similar declining trend. The results indicate that the model aligns well with the observed accelerated expansion of the current universe. These findings provide valuable insights into the role of cosmic strings and modified gravity theories in explaining large-scale cosmic evolution.Çanakkale Onsekiz Mart Universit

Wildlife considerations in local land use plans: The influence of different regional planning environments in New York and Vermont

    No full text
Connecting large-scale conservation efforts to local land use planning processes is an important challenge. How conservation themes get incorporated into local land use planning depend not only on higher-level policies but also the local community characteristics. This research seeks to understand this process by looking at two landscape linkages, one located in New York and the other across the state border of New York and Vermont. This exploratory study aims to investigate the extent of the wildlife conservation language in the existing land use plans of towns located within the linkage sites using plan content analysis. The differences in content analysis results across three regional environments are further investigated to understand the influence of local variables through statistical analysis, and of regional variables through a more close-up evaluation of the plans. Results indicate that the wildlife language in the land use plans is influenced by the regulatory environment, information sharing by the regional planning agencies, development pressure and some contextual variables such as population density and education. Large-scale conservation efforts could focus on these differences among multiple scales and collaborate with regional planning agencies in order to increase success with implementation.Northeastern States Research CooperativeWe would like to thank the two anonymous reviewers for their comments and suggestions. Thanks to the payment methods used in international trade, buyers and sellers in different countries can conduct business while speaking the same financial language and being aware of their risks and responsibilities. For traders in different countries who wish to engage in trade but lack mutual trust, the letter of credit (LC) payment method secures the buyer's goods while also ensuring the seller's payment. Due to this feature, the letter of credit is considered a safe haven for both buyers and sellers. However, despite its advantages, the LC payment method also carries risks for both importers and exporters. This study aims to contribute to the literature by first evaluating the payment methods used in international trade to provide a clearer understanding of the advantages of LC compared to other payment methods. Then, the LC payment method is examined in detail, and the risks associated with it for both importers and exporters are identified. To achieve this objective, a comprehensive literature review was conducted to examine previous studies and explain the conceptual framework of the topic. Subsequently, a field study was carried out to collect data on the risks of LC payment from the perspectives of importers and exporters. In this context, qualitative data collection techniques were employed, and interviews were conducted with representatives of importer and exporter firms, as well as bank officials. The findings from these interviews indicate that both importer and exporter firm representatives consider the LC payment method to be the most secure payment method for their transactions. However, importer representatives highlighted product and shipment risks as their primary concerns, whereas exporter representatives emphasized payment and buyer risks as their most significant challenges

Identification and inhibition of PIN1-NRF2 protein-protein interactions through computational and biophysical approaches

    No full text
NRF2 is a transcription factor responsible for coordinating the expression of over a thousand cytoprotective genes. Although NRF2 is constitutively expressed, its stability is modulated by the redox-sensitive protein KEAP1 and other conditional binding partner regulators. The new era of NRF2 research has highlighted the cooperation between NRF2 and PIN1 in modifying its cytoprotective effect. Despite numerous studies, the understanding of the PIN1-NRF2 interaction remains limited. Herein, we described the binding interaction of PIN1 and three different 14-mer long phospho-peptides mimicking NRF2 protein using computer-based, biophysical, and biochemical approaches. According to our computational analyses, the residues positioned in the WW domain of PIN1 (Ser16, Arg17, Ser18, Tyr23, Ser32, Gln33, and Trp34) were found to be crucial for PIN1-NRF2 interactions. Biophysical FP assays were used to verify the computational prediction. The data demonstrated that Pintide, a peptide predominantly interacting with the PIN1 WW-domain, led to a significant reduction in the binding affinity of the NRF2 mimicking peptides. Moreover, we evaluated the impact of known PIN1 inhibitors (juglone, KPT-6566, and EGCG) on the PIN1-NRF2 interaction. Among the inhibitors, KPT-6566 showed the most potent inhibitory effect on PIN1-NRF2 interaction within an IC50 range of 0.3-1.4 mu M. Furthermore, our mass spectrometry analyses showed that KPT-6566 appeared to covalently modify PIN1 via conjugate addition, rather than disulfide exchange of the sulfonyl-acetate moiety. Altogether, such inhibitors would also be highly valuable molecular probes for further investigation of PIN1 regulation of NRF2 in the cellular context and potentially pave the way for drug molecules that specifically inhibit the cytoprotective effects of NRF2 in cancer.Ministry of National Education of Republic of Turkiye within the scope of the YLSY scholarship program; COST Action [CA20121]; COST (European Cooperation in Science and Technology)The authors would like to thank the Ministry of National Education of Republic of Turkiye within the scope of the YLSY scholarship program for funding (AO). The Potential of the Inclusion of Prosopis farcta Extract in the Diet on the Growth Performance, Immunity, Digestive Enzyme Activity, and Oxidative Status of the Common Carp, Cyprinus carpio, in Response to Ammonia Stress

    No full text
Herbal feed additives have benefits in aquaculture, as they can improve growth performance, welfare, and stress resistance. Hence, the effects of dietary Prosopis farcta extract (PFE) on the growth parameters, digestive enzyme activity, antioxidant defense, innate immunity responses, and resistance to ammonia stress in the common carp, Cyprinus carpio, were studied. Fish (15.14 +/- 0.72 g) were fed on diets without the PFE (PFE0) or those fortified with 0.5% (PFE0.5), 1% (PFE1), or 2% (PFE2) PFE for 60 days and then subjected to ammonia stress for 24 h. The growth rate, feed efficiency, and amylase-, lipase-, and protease-specific activities in the PFE1 and PFE2 treatments showed significant elevations compared to these values in PFE0. The intestinal protease-specific activity significantly increased in all of the PFE treatments compared to that in the PFE0 treatment. Serum total protein and immunoglobulin significantly increased in the PFE1 treatment, whereas serum albumin and alternative complement activity significantly increased in the PFE2 treatment compared to these values in the PFE0 treatment. The PFE2 treatment significantly mitigated post-stress elevations in serum cortisol, glucose, malondialdehyde levels, alanine aminotransferase, and aspartate aminotransferase activities. The PFE1 treatment significantly mitigated post-stress elevations in CAT activity and decreases in SOD and lysozyme activity. In conclusion, 1-2% dietary PFE supplementation can improve the growth performance, health, and resilience to environmental stressors of the common carp.RUDN University [numero; 040728-2-000]; RUDN University Scientific Projects Grant SystemThis publication was supported by the RUDN University Scientific Projects Grant System, project & numero; 040728-2-000
