Investigation of in-vitro antioxidant and cytotoxic potential of 1:1 mixture of grape seed and green tea ethanolic extract
This project report is submitted in partial fulfilment of the requirements for the degree of Bachelor of Pharmacy, 2018.Catalogued from PDF version of project report.Includes bibliographical references (page 65-71).Grape seed extract and Green tea (also known as Camellia sinensis) are two very common plants extracts that have been generally used in several food and beverage applications. Due to the health benefits associated with them, makes them a popular choice for our study. From various literature reviews, it has been revealed that these plants exhibit properties such as anti-oxidant, hypoglycemic, anti-microbial, anti-cancer, anti-smoking, have food safety applications and also provide synergistic effects when consumed with certain drugs such as 5-Fluorouracil. Since no in-vitro antioxidant and cytotoxic potential studies were carried out with the 1:1 mixture of grape seed and green tea extract in ethanolic solvent, this study was designed to observe whether their anti-oxidant potential and cytotoxic activity increases due to synergistic effects or not. Firstly, phytochemical screening revealed that the Grape seed extract (GSE) did not exhibit alkaloid and glycosides. Similarly, Green tea extract (GTE) showed the absence of alkaloid, saponin and glycosides. For better understanding of the antioxidant potential of 1:1 mixture of GSE and GTE, several in-vitro antioxidant tests were carried out. The DPPH free radical scavenging assay denoted that 96.42% of inhibition of free radicals was evident at 1200μg/mL of the extract. The IC50 was found to be 75.86μg/mL for the plant extract, whereas the IC50 was found to be 84.783μg/mL for the standard ascorbic acid which indicated that the extract exhibited more anti-oxidant activity than the standard. The Total Phenolic Content (TPC) was found to be 122.87 mg of gallic acid equivalent (GAE)/g at 1200μg/mL. The Total Flavonoid Content (TFC) and the Total Antioxidant Capacity (TAC) was found to be very low. Therefore, overall anti-oxidant potential was moderate. Cytotoxic activity was examined using an in-vitro cytotoxicity screening method known as MTT assay on cervical cancer cell line (HeLa). The results indicated that highest percentage of cell growth inhibition was seen for 2.5mg/mL concentration of sample in which about 37.97% of cell growth was inhibited giving IC50 value of 8.1 mg/mL. To conclude, it can be inferred that due to absence of synergistic effect, the ethanolic extract of 1:1 mixture of grape seed and green tea showed weak cytotoxic effect and moderate antioxidant potential.Raihanatul AnwarB. Local varieties of Allium Sativum has more effective antimicrobial property compared to imported variety
This thesis is submitted in partial fulfilment of the requirements for the degree of Bachelor of Science in Microbiology, 2018.Catalogued from PDF version of thesis report.Includes bibliographical references (page 46).Natural antibiotics or commonly known as herbal plants are used against various small diseases
throughout the history of mankind. Now we use synthetic antibiotics. However, these antibiotics are very
much ineffective against various microorganisms because of the antibiotic resistance. In current work we
used Bangladeshi and hybrid garlic against Vibrio cholerae, Staphylococcus aureus, Bacillus cereus and
Streptococcus pyogenes. The garlic samples were dried and ethanolic and methanolic extract were
prepared following a previously published protocol. The extracts were diluted by Dimethyl sulfoxide and
Distilled water at 75%, 50% and 25% for conducting agar diffusion tests. The prepared extracts were
tested on microorganisms by using minimal bactericidal concentration of garlic extract. Further, agar well
diffusion test were performed in order to determine the zone of inhibition. For determine the qualitative
analysis of the phytochemical properties of Bangladeshi and hybrid garlic, we did basic phytochemical
screening tests. Bangladeshi variety of garlic showed MBC of 62.5%, 67.5%, 52.5%, and 65% against
Vibrio cholerae, Staphylococcus aureus, Bacillus cereus and Streptococcus pyogenes respectively where
as MBC of 70%, 75%, 70%, and 70% was found for the hybrid variety against the same organisms. The
average diameters of inhibition-zone were 11mm, 12.5mm, 21.5mm and 18.65mm against Vibrio
cholerae, Staphylococcus aureus, Bacillus cereus and Streptococcus pyogenes respectively while the
same for hybrid variety were 8mm, 6mm, 20.67mm, and 7mm These results suggest that the local variety
might be more effective against the bacteria tested in this study. The phytochemical test result shows that
Bangladeshi Garlic contains protiens, tannins and phenols, alkaloids and tritenpenoids on the other hand
hybrid garlic has proteins and alkaloids but tritenpenoids was absent. Overall, the result shows that
Bangladeshi garlic might be more effective against microorganisms and it can act as a stepping stone to
use garlic for therapeutic purposes.Aakib PaevezB. Microbiolog
Cultural appropriateness of post Sidr housing reconstruction : a case of Sharankhola, Bagerhat
This dissertation is submitted in partial fulfillment of the requirements for the degree of Master in Disaster Management, 2018.Cataloged from PDF version of dissertation.Includes bibliographical references (page 86-89).Bangladesh is one of the most disaster prone countries in the world. Among the natural hazards cyclones and the associated storm surges are considered as the major threat to our 710km coastal belt with around 28 million people living within the risk zone. One of the major impacts of cyclone and storm surge is on human habitat severely disrupting the livelihood of the affected population. The usual response from the Government and different humanitarian organizations is providing emergency temporary shelter assistance and eventually a transitional or core shelter assistance following a ‘built back better’ approach focusing on resilience building and risk reduction. Cyclone Sidr, which hit our coast line in late 2007, caused massive destruction of settlements and was followed by an unprecedented reconstruction initiative by the government and NGOs changing the rural landscape permanently within a few years. Most of the agencies provided a cyclone resistant core shelter with potential for future addition and alteration by the owners. Such a multi-agency intervention, without proper consideration of rural culture of responsive built environment and indigenous materials and technologies may lead to a culturally inappropriate settlement degrading our rural landscape and heritage. This research aims to analyze cultural elements of our coastal settlement in general and evaluate the post Sidr assisted reconstruction works in the backdrop of cultural appropriateness. Sharankhola, one of the most intervened areas after Sidr, is selected as the study area of this research. The later alterations and modifications by the users will be studied as an indication of cultural inadequacies. Observation, focus group discussions and household survey in the study area will be conducted to ascertain user feedback and consciousness about culture and the outcome of the assisted shelters.Shams Mansoor GhaniM. Application of machine learning techniques on the context of livestock analysis
This thesis is submitted in partial fulfilment of the requirements for the degree of Bachelor of Science in Computer Science and Engineering, 2018.Cataloged from PDF version of thesis.Includes bibliographical references (pages 49-51).Livestock industry has been one of the fastest growing sectors of business and research in Bangladesh over the past decade. This rising, lucrative and profitable industry seems to be attracting a good number of enthusiastic investors to invest their capital, and make a contribution to the country’s overall GDP, and recover the deficit in meat production. However, due to lack of suitable resources, reliable data and information, proper knowledge and precise guidance, these investors are lagging behind in generating their expected outcomes. Moreover, these investors and farmers tend to make choices based on their experience only. To come to their assistance in making compatible decisions and provide with a profitable and efficient approach in the investor’s business expansion in Bangladesh, this research aims to establish an intelligent prediction methodology through regression analysis, by implementing data mining and supervised machine learning techniques. This research provides some cattle’s breed based analysis depending on different related factors, which includes age, current weight of cattle, the environment it is reared on, the diet plan and the geographical region it originated from. The models implemented in this research were, Linear Regression model, Ordinary Least Square Regression model, Polynomial Regression model and Decision Tree learning for attaining this prediction mechanism. For executing the analysis, an unanalyzed data set, having a period of 12 years, were collected from Bangladesh Livestock Research Institute, Savar Dairy Farm, and Meghdubi Agro farm.Md.
