    Mechanism of smoothened localization to the primary cilium in hedgehog signal transduction

    honors thesisCollege of Social & Behavioral ScienceBiologyBenjamin MyersThe Hedgehog (Hh) pathway is a vital cellular signaling pathway involved in embryonic development and general tissue homeostasis in vertebrates. Aberrant activation of hedgehog signaling has implications in developmental disorders and multiple forms of cancer [1,28,41]. The pathway is activated when Hh proteins bind to their receptor Patched (PTCH1) which relieves inhibition on G-protein coupled receptor (GPCR) like protein, Smoothened (SMO). SMO can then bind cholesterol, activating the protein and triggering GRK mediated phosphorylation and PKA binding, preventing phosphorylation of gliomaassociated (GLI) transcription factors and allowing for Hh target genes to be transcribed [3,13,15,35]. Hedgehog singling takes place in the primary cilium, an antennalike, membrane bound signaling hub found along the periphery of most cells in vertebrates. [6,11,30,34]. Importantly, protein synthesis does not occur in the primary cilium. Therefore, ciliary signaling function requires proteins to be targeted to the ciliary membrane [30]. Targeting and entry of signaling proteins to the primary cilium is a tightly regulated process, mediated by a variety of mechanisms, most of which are not conserved across different ciliary signaling pathways. Even though SMO is known to signal via the primary cilium, the mechanism by which it localizes to the organelle is not entirely clear. To address this question, we developed cell lines containing specific SMO C-tail truncations and completed a variety of drug-based assays followed by antibody staining and confocal microscopy to visualize SMO localization in vitro. While the current understanding of SMO ciliary localization is based on ubiquitination of the third intracellular loop (ICL3) of inactive SMO and shuttling from the cilium, previous work indicates that the C-tail of SMO is necessary for ciliary localization and contains two, partially redundant, ciliary localization sequences (CLS1 and CLS2) [7,19,22,25]. Our preliminary work illustrates a preliminary mechanism by which localization via the membrane-proximal CLS1 is independently mediated through activity-dependent PKA/GRK2/3 interactions with SMO, while the more membrane-distal CLS2 is mediated by ubiquitination of inactive SMO. Understanding the intricate mechanism by which SMO localizes to the primary cilia is crucial in developing treatments to Hedgehog-related pathologies

    Role of the oncogene E26 tranformation-specific sequence-i in arteriovenous fistula maturation failure

    honors thesisCollege of EngineeringBiomedical EngineeringYang-Ting ShiuAbout 15% of the adult United States (U.S.) population is affected by chronic kidney disease (CKD) [1]. Almost 70% of end-stage kidney disease patients in the U.S. need hemodialysis to survive [2]. A vascular access point must be created for hemodialysis to occur. The current gold standard for vascular access points is the arteriovenous fistula (AVF) as it lasts the longest [3]. After its creation, an AVF must successfully mature to be useful. AVF maturation failure often occurs due to excess cell proliferation into the AVF\u27s lumen, called neointimal hyperplasia. When AVF maturation failure occurs, hemodialysis can no longer occur because blood cannot flow through the vascular access point at the needed rate. The aim of this project is to investigate whether E26 transformation-specific sequence-1 (ETS-1) causes neointima hyperplasia after arteriovenous fistula creation surgery in a rat model with chronic kidney disease. The leading hypothesis is that ETS-1 inhibition decreases neointima hyperplasia in the AVF following vascular injury. Histological analysis of ETS-1 gene knockout (KO) and wild-type (WT) rats with AVFs in healthy and induced CKD models was performed, and morphometric analysis was used to quantify the open lumen percentage of AVFs in all four groups. No statistically significant difference in open lumen percentage was found at both Week 1 (on average, WT with CKD: 86.3% at n=4; KO with CKD: 78.9% at n=2) and Week 4 (on average, WT with CKD: 31.9% at n=4; KO with CKD: 63.6% at n=2) between the four experimental groups. Further analysis of rats at both Week 1 and Week 4 is needed to draw conclusions regarding the relationship between ETS-1 inhibition and neointimal hyperplasia formation. Further translational research into inhibition of the ETS-1 pathway may be worthwhile and lead to the development of therapies for prevention of neointimal hyperplasia, improved AVF maturation rates, and reduced need for revisit surgeries, enhancing the quality of life for patients with CKD

    The interview with Dr. Glen Fifield, a 95-year-old retired music educator, details his musical career and his immense contribution to the music program in Carbon County, Utah. Starting with his father\u27s cornet, his passion led to serious music study and a teaching career that saw him and woodwind specialist Derral Siggard build a celebrated band program in Price, which was invited to march in the Rose Parade. They developed the Master Musician program to motivate students to achieve individual excellence. During his time in Price, Fifield also played trumpet in the legendary Jimmy Dart Orchestra, a big band dance group that performed across the state. He later taught at the College of Eastern Utah and finished his career at Utah State University. Dr. Fifield praised the vital contributions of Janis Siggard, who served as a tremendous musician and accompanist for hundreds of student solos. He and Derral Siggard consciously worked to uphold the musical tradition established by earlier educator E.M. Williams, proudly referring to their group as A Band With a Heritage

    This patient demonstrates 4 common types of intrusions/nystagmus that may be seen in cerebellar ataxia: 1) square wave jerks (SWJ, a saccadic intrusion), 2) downbeat nystagmus (seen minimally in primary gaze, mainly in lateral gaze), 3) gaze-evoked nystagmus, 4) rebound nystagmus. The presence of SWJ can make it more challenging to diagnose nystagmus, but the presence of downbeat, gaze-evoked and rebound nystagmus is highly suggestive of cerebellar flocculus/paraflocculus dysfunction
