Effect of human survivin-2B-specific cytotoxic CD8+ T lymphocytes on CD44+/- HSC-2 and HSC-3 oral cancer cells
Despite advancements in the treatment of oral cancer, cancer survival rates remain low, highlighting the need for new therapeutic strategies targeting cancer stem-like cells. Cancer stem-like cells are a small population of cancer cells within tumors that drive recurrence and metastasis. They are often resistant to conventional treatments. Immunotherapy has shown promise against cancer stem-like cells, particularly with the use of cytotoxic T lymphocytes targeting specific markers. Survivin, an apoptosis protein inhibitor, is overexpressed in several malignancies, including oral cancer, and is associated with tumor recurrence and reduced survival. Survivin-2B-specific cytotoxic T lymphocytes were produced and evaluated for their ability to target CD44+ (cancer stem-like cells) and CD44- cells (non-cancer stem-like cells), respectively, from oral cancer cell lines (HSC-2 and HSC-3, respectively). Quantitative polymerase chain reaction (qPCR) analysis confirmed similar survivin-2B expression in both cell types. Cytotoxic T lymphocyte assays revealed the effective lysis of both cancer stem-like cells and CD44- cell populations, supporting the potential of survivin-2B-specific cytotoxic T lymphocytes to overcome cancer stem-like cell-associated resistance. These findings suggest that survivin-2B peptide vaccines are effective in preventing cancer relapse by targeting cancer stem-like cells, with future directions aimed at developing multipeptide "cocktail" vaccines to reduce the risk of immune evasion.journal articl
高校訪問データベースを通してみえた入試広報の課題─札幌医科大学医学部を事例に─
本研究は、札幌医科大学医学部の高校訪問活動においてデータベースを構築・運用することで明らかとなった入試広報の課題と改善策を示すものである。FileMaker プラットフォームを用いて過去10 年分の訪問記録や受験・合否状況を一元管理し、検索機能を強化することで情報の活用効率を向上させた。統合されたデータの分析により、合格者がいない出願校への対応や道外高校への広報戦略の必要性、訪問記録の比較による改善点が明らかとなった。また、報告書の標準化やフィードバック活用、戦略的な訪問計画を実施することで、訪問活動の質向上と志願者確保の強化を目指したい。本研究では、データベース活用の有効性を示し、今後の広報活動の発展に寄与することを目指した。This study aims to identify the challenges and improvements in admission public relations revealed through the construction and operation of a database for high school visit activities at the Sapporo Medical University Faculty of Medicine. Using the FileMaker platform, visit records and examination results from the past ten years were centrally managed, and search functionality was enhanced to improve the efficiency of information utilization.
Analysis of the integrated data clarified issues such as addressing applicant schools with no successful candidates,the necessity of public relations strategies for high schools outside Hokkaido, and improvements identified through comparisons of visit records. Additionally, by standardizing reports, utilizing feedback, and implementing strategic visit planning, we aim to enhance the quality of visit activities and strengthen applicant recruitment.
This study demonstrates the effectiveness of database utilization and seeks to contribute to the further development of admission public relations activities in the future.departmental bulletin pape
行動嗜癖の現状と課題
行動嗜癖(behavioral addiction)とは,アルコールや麻薬などの精神作用物質ではなく,ある特定の行動や一連の行動プロセスを依存対象とする「依存症」の一種である.2013年に公表された精神障害の診断および統計マニュアル第5版(DSM-5)で,ギャンブル障害は物質依存症と同じカテゴリーに分類された.さらに,2018年に公表された国際疾病分類第11回改訂版(ICD-11)では,ゲーム障害が初めて疾病として採択された.しかし,行動嗜癖と物質依存を同じカテゴリーとすることには,専門家の中でも議論が分かれている.行動嗜癖の概念が広がる中,その実態や生物学的病態の解明が今後の課題である.いずれにせよ,早期介入,心理療法,リハビリテーション(作業療法)は,行動嗜癖や物質依存症治療の中心である.本稿では,最初に行動嗜癖の概念について述べ,ギャンブル障害とゲーム障害について概説する.departmental bulletin pape
A Novel Tight Junction-Nuclear Receptor Signaling Pathway Regulating Cancer Progression
Nuclear receptors (NRs) are lipid ligand-binding transcription factors, with 48 members having been identified in humans to date. They are involved in diverse physiological processes, including development and homeostasis, and are also implicated in the pathogenesis of various diseases, most notably cancer. While NR activity is primarily regulated by specific ligand binding, posttranslational modifications, particularly phosphorylation, also play a critical role in modulating their function. Recently, we identified a novel signaling pathway linking claudins (CLDNs), cell-cell adhesion proteins, to NRs. CLDN-mediated cell-cell adhesion activates Src family kinases (SFKs), leading to serine phosphorylation of several NRs. This newly-discovered CLDN-NR pathway contributes to epithelial differentiation in stem cells and promotes cancer progression.
