ABSTRACT　Lysosomes are organelles that play a crucial role in the degradation of 
endocytosed molecules, phagocytosed macromolecules and autophagic substrates. The 
membrane of lysosomes contains several highly glycosylated membrane proteins, and 
lysosome-associated membrane protein (LAMP)-1 and LAMP-2 account for a major portion 
of the lysosomal membrane glycoproteins. Although it is well known that LAMP-2 deficiency 
causes Danon disease, which is characterized by cardiomyopathy, myopathy and mental 
retardation, the roles of lysosomal membrane proteins including LAMP-1 and LAMP-2 in 
myogenesis are not fully understood. In this study, to understand the role of LAMP proteins in 
the course of differentiation of myoblasts into myotubes, we used C2C12 myoblasts and found 
that the protein and mRNA levels of LAMP-1 and LAMP-2 were increased in the course of 
differentiation of C2C12 myoblasts into myotubes. Then, we investigated the effects of LAMP-
1 or LAMP-2 knockdown on C2C12 myotube formation, and found that LAMP-1 or LAMP-2 
depletion impaired the differentiation of C2C12 myoblasts and reduced the diameter of C2C12 
myotubes. LAMP-2 knockdown more severely impaired C2C12 myotube formation compared 
with LAMP-1 knockdown, and knockdown of LAMP-1 did not exacerbate the suppressive 
effects of LAMP-2 knockdown on C2C12 myotube formation. In addition, knockdown of 
LAMP-1 or LAMP-2 decreased the expression levels of myogenic regulatory factors, MyoD 
and myogenin. These results demonstrate that both LAMP-1 and LAMP-2 are involved in 
C2C12 myotube formation and LAMP-2 may contribute dominantly to it.
抄録　リソソームは様々な生体分子の分解に関わる細胞小器官であり、リソソーム
膜には多数のリソソーム膜糖タンパク質が存在する。主要なリソソーム膜糖タンパ
ク質として LAMP-1 と LAMP-2 が知られており、LAMP-2 は心筋症、ミオパチーお
よび精神遅滞を特徴とするダノン病に関連することが良く知られているが、筋形成に
おける LAMP-1 および LAMP-2 の役割は十分に解析されていない。本研究において
C2C12 筋芽細胞の筋管細胞への分化の過程における LAMPs の役割の解明を試みたと
ころ、LAMPs のタンパク質量および mRNA 量が C2C12 の分化過程で増加すること
が明らかになった。また、LAMPs の発現抑制が C2C12 筋芽細胞の分化を抑制する
ことも明らかにした。さらに、LAMPs の発現抑制が筋特異的転写因子 MyoD および
myogenin の発現量を低下させることも見出した。これらの結果は、LAMP-1 および
LAMP-2 が C2C12 筋芽細胞の分化に関与することを示唆している。

ABSTRACT　 Lysosome-associated membrane protein-1 (LAMP-1) is a type I membrane 
glycoprotein with a COOH-terminal lysosome-targeting signal peptide of G1YQRTI5-COOH. 
This sequence is categorized as a tyrosine-based motif of GYXXΦ where Φ is a bulky 
hydrophobic amino acid residue. Lysosomal localization of LAMP-1 varies by changing 
the Φ amino acid residues. In this study, we conducted computer-based molecular modeling 
for structures of the COOH-terminal wild-type and mutant peptides (GYQTI, GYQTL, 
GYQTF, GYQTM, and GYQTV) complexed with a medium subunit of adaptor protein 
complex-2 (μ2 of AP-2), a key molecule in vesicular transport to endosomes and lysosomes. 
Tyr2 of all the peptides is critical for their rigid binding to μ2 by well fitted hydrogen 
bonds and hydrophobic interactions between Tyr2 and the surrounding amino acids of μ2. 
The Φ hydrophobic side chains of LAMP-1-derived COOH-terminal peptides are located 
exclusively within a hydrophobic pocket formed by five hydrophobic amino acid moieties on 
β-sheets of μ2, whereas slight but significant differences were observed in the spatial position 
of the side chain of a Φ residue in the hydrophobic pocket, causing their differential affinities 
with μ2. These results could explain, at least in part, why the COOH-terminal variants of 
LAMP-1 show different lysosomal localization in late endosomes and lysosomes.  
抄録　リソソーム膜タンパク質 -1（LAMP-1）は、C- 末端に G1YXX Φ5 モチーフ ( Φ：
かさ高い疎水性アミノ酸残基 ) を持つ。本研究では、5 種類の GYXX Φ（Φ＝ I,L,F,M,V）
ペプチドとアダプタータンパク質（AP-2）のμ 2 サブユニットとの複合体を分子モデリ
ングで検討した。その結果、すべてのペプチドにおいて Tyr2 は、μ 2 のアミノ酸残基
と水素結合および疎水性相互作用によって強固な結合を形成するのに重要であった。ま
た、Φ残基の側鎖は、μ 2 のβシート上の 5 つの疎水性アミノ酸部分によって形成され
る疎水性ポケット内に位置するが、その空間位置のわずかではあるが有意な違いがμ 2
との親和性の差異を引き起こしていた。これらの結果は、LAMP-1 の C 末端変異体によっ
て後期エンドソームとリソソームでリソソームの局在化が異なる理由を説明できる。

ABSTRACT　 A mixture of pharmaceuticals having a xanthine skeleton, theophylline, 
proxyphylline, diprophylline and (-)-epigallocatechin-3-O-gallate (EGCg) in 
water created a sticky precipitates, which were thought to be 2:2 complexes of the 
pharmaceuticals and EGCg.  The molecular capture ability of the pharmaceuticals having 
a xanthine skeleton by EGCg was estimated by the amount of the pharmaceuticals 
included in the precipitates of the complexes, and measured by the integrated value of 
proton signals in the quantitative 1H NMR spectra.
 Based on changes in chemical shifts of proton signals of the pharmaceuticals with a 
xanthine skeleton in 1H NMR spectra by adding standard amounts of EGCg, the xanthine 
skeleton of the pharmaceuticals was considered to exist in the hydrophobic space formed 
by the three aromatic A, B, B’ rings of EGCg, and a part of the proxyphylline and 
diprophylline side chains existed out of the hydrophobic space. 
In the 1H NMR spectra of the mixture of (R)- and (S)-proxyphylline, (R)- and (S)-
diprophylline and an equimolecular amount of EGCg, the N3-CH3 signal of (R)- and 
(S)-proxyphylline, and (R)- and (S)-diprophylline was clearly observed as two singlets. 
This suggested that EGCg recognized the chirality of proxyphylline and diprophylline in 
water.  
抄録　キサンチン骨格をもつ医薬品 テオフィリン、プロキシフィリン、ジプロフィ
リンと (-)- エピガロカテキン -3-O- ガレート（EGCg）の混合物の水溶液からは、そ
れらの医薬品と EGCg との 2:2 錯体であると考えられる粘着状物質を与えた。キサ
ンチン骨格をもつ医薬品の EGCg による分子補足能は、錯体である沈殿に含まれる
それらの医薬品の量で評価した。その際、定量 1H NMR スペクトルにおける積算
値を用いて測定した。EGCg を一定量ずつ加えていくことにより 1H NMR スペク
トルにおけるそれら医薬品の化学シフト値の変化をもとにして、キサンチン骨格が
EGCg の A 環、B 環、B’ 環の３つの芳香環によって形成される疎水性空間の中に存
在していると考えられる。また、プロキシフィリンとジプロフィリンの側鎖の一部
もその疎水性空間の外に存在していると考えらる。
　(R) 体 と (S) 体の混合物のプロキシフィリンとジプロフィリンと、等モル量の

EGCg の 1H NMR スペクトルにおけて、いずれも N3-CH3 のシグナルが２つのシン
グレットとして明確に分離した。　これは，EGCg によりプロキシフィリンとジプ
ロフィリンの不斉を水中で認識したことを示唆している。
