to the search for therapeutic targets in atherosclerosis

ABSTRACT  The retinoic acid receptor-related orphan receptor α (RORα) is involved 
in the regulation of several physiological processes, including development, metabolism, 
and circadian rhythm. RORα-deficient mice display profound atherosclerosis, in which 
hypoalphalipoproteinemia is reportedly associated with decreased plasma levels of high-
density lipoprotein, increased levels of infl ammatory cytokines, and ischemia/reperfusion-
induced damage. The recent characterization of endogenous ligands (including cholesterol, 
oxysterols, provitamin D3, and their derivatives), mediators, and initiation complexes 
associated with the transcriptional regulation of these orphan nuclear receptors has facilitated 
the development of synthetic ligands. These findings have also highlighted the potential 
of application of RORα as a therapeutic target for several diseases, including diabetes, 
dyslipidemia, and atherosclerosis. In this review, the current literature related to the 
structure and function of RORα, its genetic inter-individual differences, and its potential as a 
therapeutic target in atherosclerosis is discussed. 
抄録　レチノイン酸受容体関連オーファン受容体 α (RORα)は、発生、代謝、概日リ
ズムなど、いくつかの生理的プロセスの制御に関与している。RORα欠損マウスは高
密度リポ蛋白の血漿レベルでの低下、炎症性サイトカインの増加、虚血 /再灌流によ
る障害と関連し、動脈硬化を呈することが報告されている。近年、これらの核内受
容体の転写制御に関わる内因性リガンド（コレステロール、オキシステロール、プ
ロビタミン D3、およびそれらの誘導体を含む）、メディエーター、開始複合体の特
徴が明らかにされ、合成リガンドの開発が進展している。これらの知見は、RORαが
糖尿病、脂質異常症、動脈硬化症などの疾患に対する治療標的としての可能性を示
している。本総説では、RORαの構造と機能、その遺伝的な個人差、および動脈硬化
の治療標的としての可能性に関する現在の文献を紹介する。

Drug-Metabolizing Enzymes-Mediated Inter-Individual Differences

ABSTRACT  Colossal inter-individual differences existing in biological responses to 
endogenous and exogenous substances are attributable to minute biological interplays, 
including nuclear receptors’ signaling and activities of drug-metabolizing enzymes. Nuclear 
receptors, such as estrogen receptor (ER), peroxisome proliferator-activated receptor β/δ 
(PPARβ/δ), and retinoic acid receptor-related orphan receptor α (RORα), are basically ligand-
inducible transcription factors. They are known to be robustly involved in the regulation 
of numerous physiological processes, such as metabolism, development, and reproduction 
via regulation of target genes, thus, rendering them as attractive drug targets. At the post-
transcriptional level, some microRNAs are suggested to be directly/indirectly involved in 
the regulation of CYP3A protein expression. Furthermore, at the post-translational levels, 
it has been evidently shown that there are functional protein–protein interactions between 
different kinds of drug-metabolizing enzymes i.e., P450 and uridine 5′-diphosphate (UDP)-
glucuronosyltransferase, UGT, which results in modulation of the enzyme(s) activities. In 
this Current Topics publication, 5 young promising scientists and their senior colleagues 
were invited to contribute papers for updating/improving our readers’ appreciation of the 
nuclear receptors- and drug-metabolizing enzymes-mediated inter-individual differences 
especially in the toxicological aspects. 1) Dr. Hirao-Suzuki highlights the significance of 
breast cancer malignancy stimulation by environmental chemicals, especially focusing on 
the interaction bisphenols and ERβ subtype in the enhanced malignancy. 2) Dr. Koga and 
Dr. Peters review the critical roles of PPARs including PPARβ/δ in the pathogenesis of 
alcoholic liver disease. 3) Dr. Matsuoka and Dr. Michihara summarize the structure/function 
of RORα and its potential utility as a therapeutic target for atherosclerosis. 4) Dr. Fujino, Dr. 
Sanoh, and Dr. Katsura evaluate the regulation of CYP3A expression in humans and rodents, 
coupled with individual differences in toxicological aspects, by focusing on the endo- and 
exogenous factors. 5) Dr. Miyauchi, Dr. Takechi, and Dr. Ishii recapitulate the functional 
protein–protein interactions between P450 and UGT, which imply a novel post-translational 
factor regulating interindividual differences in drug metabolism. I fi rmly believe that you, 
the reader, will essentially benefi t and develop a better understanding of the nuclear receptors 
and drug-metabolizing enzymes-driven inter-individual differences in toxicological aspects 
after reading this Current Topics.

抄録　内因性および外因性物質に対する生体応答には大きな個人差が存在し、これ
は核内転写因子を介したシグナルや薬物代謝酵素の活性を含めた緻密な相互作用
に起因する。エストロゲン受容体（ER）、ペルオキシソーム増殖剤応答性受容体 β/
δ(PPARβ/δ)、およびレチノイド関連オーファン受容体 α(RORα)は基本的にリガンド
誘導性の転写因子である。これらは受容体は標的遺伝子の調節を介して代謝、成長、
および再生などの多彩な生理的プロセスの制御に密接に関与している。したがって、
ここに挙げた受容体は創薬の標的として注目されている。転写後レベルでは、マイ
クロ RNAの中には、直接、あるいは間接的に CYP3Aタンパク質発現の調節に関与
しているものもある。さらに、翻訳後レベルでは、異なる薬物代謝酵素間（例えば、
P450とウリジン 5'-二リン酸（UDP）-グルクロノシルトランスフェラーゼ、UGT）
の機能的なタンパク質 -タンパク質相互作用の存在が示されている。このカレントト
ピックでは、分野に精通する 5人の気鋭の若手研究者とそれらの指導者に執筆を依
頼し、特に毒性学の点における核内受容体と薬物代謝酵素を介した個人差の理解に
関する最新の知見を紹介する。1） 平尾博士（広島国際大学）は、環境化学物質によ
る乳がん悪性化に注目し、特に特にビスフェノールと ERβサブタイプの相互作用に
着目したがん悪性化について解説する。2） 古賀博士（第一薬科大学）は、アルコー
ル性肝障害の発症機構における PPARβを含む PPARの重要な役割について概説する。
3） 松岡博士（福山大学）は、RORαの構造 /機能とアテローム性動脈硬化症の治療
標的としての RORαの可能性についてまとめた。4） 藤野博士（立命館大学）は、内
因性および外因性因子に着目し、人間とげっ歯類における CYP3A発現の調節を毒性
学の観点から評価した。5） 宮内博士（崇城大学）は、P450と UGT間の機能的タン
パク質 -タンパク質相互作用が薬物代謝における個人差を説明し得る新たな翻訳後調
節機構である可能性を示した。私は、みなさんがこのカレントトピックスを読み終
えた後、毒性学的観点における核内受容体と薬物代謝酵素による個人差の可能性に
ついて理解が深まることを期待する。

ABSTRACT  The present study focused on the development of an amphotericin B (AmB) 
nanoformulation for ophthalmic applications. Accordingly, AmB nanohydrogels (AHA/AmB) 
using alkyl glyceryl hyaluronic acid (Hyalorepair®, AHA), a hydrophobized hyaluronic 
acid, were prepared by employing the dialysis method, followed by assessments of physical 
properties, drug efficacy, and toxicity. In the AHA/AmB formulation, AmB existed in a 
selfaggregated and amorphous state in the hydrophobic environment of the AHA moiety. 
AHA/AmB was shown in vitro to interact with mucin, which is known to be expressed in 
the corneal epithelium and was expected to improve its corneal retention. Compared with 
the conventional AmB formulation, amphotericin B sodium deoxycholate, AHA/AmB had 
the same in vitro antifungal activity but signifi cantly lower in vitro toxicity. These fi ndings 
indicate that nanohydrogels prepared with AHA possess high fungal selectivity and serve as a 
promising system for ophthalmic AmB delivery.  
抄録　本研究では、眼科用途のためのアムホテリシン B（AmB）ナノ製剤の開発に
焦点を当てた。疎水化ヒアルロン酸であるアルキルグリセリルヒアルロン酸（ヒア
ロペア ®、AHA）を用いた AmBナノハイドロゲル（AHA/AmB）を透析法により
作製し、物性、薬効、毒性を評価した。AHA/AmB中において、AmBは AHAの疎
水性環境下で自己凝集した非晶質状態で存在した。AHA/AmBは角膜上皮に発現す
ることが知られているムチンと相互作用することが in vitroで示され、角膜の滞留
性を向上させることが期待された。従来の AmB製剤である amphotericin B sodium 
deoxycholateと比較して、AHA/AmBは in vitro抗真菌活性はそのままに、in vitro毒
性は有意に低いことが示された。これらの結果は、AHAで調製したナノハイドロゲ
ルが高い真菌選択性を有し、眼科用 AmB送達のための有望なシステムであることを
示している。

ABSTRACT  Increased permeability of endothelial cells lining the blood vessels in the 
brain leads to vascular oedema and, potentially, to stroke. The tight junctions (TJs), primarily 
responsible for the regulation of vascular permeability, are multi-protein complexes 
comprising the claudin family of proteins and occludin. Several studies have reported that 
downregulation of the claudin domain containing 1 (CLDND1) gene enhances vascular 
permeability, which consequently increases the risk of stroke. However, the transcriptional 
regulation of CLDND1 has not been studied extensively. Therefore, this study aimed to 
identify the transcription factors (TFs) regulating CLDND1 expression. A luciferase reporter 
assay identified a silencer within the first intron of CLDND1, which was identified as a 
potential binding site of the myeloid zinc fi nger 1 (MZF1) through in silico and TFBIND 
software analyses, and confi rmed through a reporter assay using the MZF1 expression vector 
and chromatin immunoprecipitation (ChIP) assays. Moreover, the transient overexpression 
of MZF1 significantly increased the mRNA and protein expression levels of CLDND1, 
conversely, which were suppressed through the siRNA-mediated MZF1 knockdown. 
Furthermore, the permeability of FITC-dextran was observed to be increased on MZF1 
knockdown as compared to that of the siGFP control. Our data revealed the underlying 
mechanism of the transcriptional regulation of CLDND1 by the MZF1. The fi ndings suggest 
a potential role of MZF1 in TJ formation, which could be studied further and applied to 
prevent cerebral haemorrhage. 
抄録　脳血管内皮細胞における物質透過性の亢進は、血管浮腫を引き起こし、脳卒
中を惹起する。血管における物質透過性は、クローディンやオクルディンにより形
成される密着結合により調節される。これまで我々は、脳出血誘発マウスにおいて
クローディンファミリーに属する CLDND1の発現低下により物質透過性が亢進する
ことを報告している。これらのことから、脳卒中発症の原因である物質透過性の亢
進は、CLDND1の発現低下によっても誘発されると考えられる。しかし、CLDND1
の転写調節はあまり研究されていない。そこで、本研究では、CLDND1発現に影響
を及ぼす転写因子の同定を試みた。レポーター解析の結果、CLDND1の第１イント
ロン内にサイレンサーが示された。サイレンサーに結合する転写因子を TFBIND解

析で検索したところ、Myeloid Zinc Finger 1（MZF1）が見出された。MZF1発現ベクター
を用いたレポーター解析およびクロマチン免疫沈降法によりMZF1がサイレンサー
に結合し、アクチベーターとして作用することが示唆された。また、MZF1一過性発
現系においてMZF1発現増加に伴う CLDND1の有意な発現増加が示された。siRNA
を用いたMZF1ノックダウンにおいて CLDND1の有意な発現低下が示された。さら
に、MZF1ノックダウン細胞における FITC-デキストランの透過性は、コントロール
群と比較して有意な増加が示された。これらの結果は、タイトジャンクション形成
におけるMZF1の潜在的な役割を示唆し、脳出血の予防に応用できる可能性がある。
