Total RNA was isolated from bMSCs (4 × 104 cells/well) using the PureLink RNA Mini Kit (12183018A, Invitrogen, Thermo Fisher Scientific) following the manufacturer’s instructions. cDNA was synthesized in a final reaction volume of 20 µL using the High-Capacity cDNA Reverse Transcription Kit with RNase Inhibitor (4368814, Applied Biosystems, Thermo Fisher Scientific). Quantitative RT-PCR was performed on the CFX96 Real-Time PCR Detection System (Bio-Rad) using TaqMan Gene Expression Assays (Life Technologies, Monza, Italy) for target genes: Cyclin Dependent Kinase Inhibitor 1A (CDKN1A) (Hs003555782_m1) which encodes p21 and mitochondrially encoded NADH dehydrogenase-1 (MT-ND1) (Hs02596873_s1), with GAPDH (Hs99999905_m1) serving as the internal reference gene. Gene expression levels were determined using the 2−ΔΔCt method

RESEARCH OBJECTIVES:
1- precise characterization of sleep endophenotype and genomic profile in a group of patients with Schizophrenia, recorded during the first episode of psychosis (FEP) and in matched control samples.
Research steps:
1.1 - Detailed characterization of sleep endophenotype and genomic profile in a group of patients with Schizophrenia, recorded during the first episode of psychosis (First Episode Psychosis, FEP) and in matched control samples.
1.2 - Follow-up psychodiagnostic assessments to obtain more information
on the correlation between thalamo-cortical dysfunction (altered spindle activity) and
longitudinal clinical phenotype in all FEP subjects with a confirmed diagnosis of Schizophrenia (SCZ).
1.3 - Blood sampling of subjects with a confirmed diagnosis of SCZ for a genetic
characterization aimed at estimating the load of common variants at individual level, stratifying them according to risk. Selection on the basis of particular allelic variants and/or significant impairment of the Thalamic Reticular Nucleus (TRN), suggested by higher sleep spindle deficits, of patients with SCZ for cellular reprogramming.
2 - in follow-up sample, identification of neural correlates, focusing on structural and functional brain connectivity related to gamma-band oscillations, using functional magnetic resonance imaging (fMRI), structural magnetic resonance imaging (sMRI), and electrophysiology (in particular simultaneous electroencephalography with fMRI (EEG - fMRI), at rest and during a task).
3 - generation of GABAergic interneurons from patient-specific induced pluripotent stem cells (human iPSCs, hiPSCs) to identify specific molecular and functional alterations that reflect GABAergic dysfunctions at the cellular level
