    The potential role of shearing load in the development of toe tip necrosis syndrome

    Toe tip necrosis syndrome (TTNS) is defined as a hind limb lameness disorder of feedlot cattle. TTNS is characterized by separation of the white line, necrosis of the third phalanx (P3) and abscess formation. The reason for white line separation with TTNS is unclear. One potential cause is that cattle, due to environmental conditions, damage the white line by scuffing their hooves on abrasive surfaces found in feedlots. This may result in small pathways for external biological materials and bacteria to enter the hoof, causing infection and initiation of TTNS. Also, an abraded worn claw may be more susceptible to local damage caused by small particles, further increasing potential for pathways to develop. The objectives of this study were: 1. Compare local stiffness along the white line of abraded claws and control claws. 2. Compare contrast-enhanced HR-pQCT image-based metrics of damage and pathway development of abraded claws, control claws and claws with TTNS. 3. Compare overall stiffness of abraded claws, control claws and claws with TTNS. Abrasion testing was performed on the lateral digit of healthy specimens. Local indentation stiffness testing was performed on these specimens as well as control specimens manually rasped to the same level of wear found with abrasion. Following that, the claws were submerged in a contrast agent and underwent suction. Specimens were then imaged using High-Resolution Peripheral Quantitative Computed Tomography (HR-pQCT). Images were segmented, with measures of mean intensity acquired. Estimates of overall stiffness were also acquired using load-displacement curves. State statistics were used to compare local stiffness, mean intensity and overall stiffness. Our results indicated a significant difference (p-value < 0.05) between local stiffness of abraded claws and control claws. Our findings also indicated differences in the mean intensity between these two groups. In addition, local damage was observed in HR-pQCT scans of abraded claws, whereas there was no evidence of local damage with control claws. Furthermore, we found that mean overall stiffness of abraded claws was similar to that of claws with TTNS and lower than control claws. These findings support the idea that abrasion is involved in TTNS pathogenesis

    Investigation of the Role of Amide I to Amide II Ratio and Alpha Helix to Beta Sheet Ratio of Faba Bean Seeds in the Determination of Microbial Protein Synthesis and Animal Performance and Metabolism in Ruminant Livestock Systems.

    The food and feed industries have benefited from the application of Infrared Spectroscopy as spectral features have been associated with nutrient quality and degradation characteristics. To our knowledge, the effects of steam pressure toasting (SPT) duration on spectral features of Faba bean seeds (FBS) have not been previously evaluated, and the influence of amide I to amide II ratio, and α-helix to β-sheet ratio (α_β) on the protein quality (solubility and digestibility), metabolism, and bioavailability in dairy cows has not been established. Thus, the objective of this research was to evaluate the role of the protein-related spectral ratios on the nutrient quality and protein metabolism of FBS in high lactating dairy cows through the application of laboratory and spectroscopic analysis, feed evaluation models (CNCPS, NRC2001, DVE), in situ nylon bag incubation, and in vitro batch culture techniques. Data were analyzed with Procedure MIXED of SAS 9.4 software using a RCBD model with the significance declared at P 0.05). The fat yield (kg/cow/day) tended to be lower with TMR_15 and TMR_30 compared to the other treatments (linear P = 0.05), milk urea nitrogen (MUN) was reduced from 12.2 to 11.1 mg/dl with TMR_0 to TMR_30, respectively (P < 0.01). The fat corrected milk (FCM3.5%) and energy corrected milk (ECM) tended to be slightly increased with TMR_7.5 than TMR_0 (P = 0.07, P = 0.08, respectively). Both parameters had a tendency to be reduced with TMR_15 and TMR_30 compared to the other treatments (P = 0.07, P = 0.08, ii respectively). Moreover, the actual milk yield, averaged pH, averaged NH3, and total VFA’s concentration were not significantly different among the dietary treatments (P > 0.05) with corresponding values reaching 39 kg/cow/day, 6.32, 102.7 mg/L, and 76.7 mmol/L, respectively. The feed efficiency (ECM/DMI) was reduced with TMR_15 and TMR_30 compared to TMR_0 (linear P = 0.02). In the rumen simulation technique (RUSITEC), CP disappearance decreased (linear P < 0.01) from TMR_0 to TMR_30, and the total microbial nitrogen (MN) increased (quadratic P = 0.03) from TMR_0 to TMR_15 (73.5 to 88.4 mg/day, respectively). In the present study, the application of mid-Infrared molecular analysis was helpful to determine changes in the protein molecular structures of FBS induced by SPT duration. Hence, the peak heights of α-helix, β-sheet, and the area ratio of amide I to amide II found in the present study were increased over SPT duration in the endosperm of FBS determined with Synchrotron radiation-based Infrared (IR) micro-spectroscopy (SR-IMS). Moreover, the areas of total amide, amide I, and amide II and the peak heights of α-helix and β-sheet were reduced over SPT duration when a mixture of the FBS’ pericarp, seed coat, aleurone cell, and endosperm was analyzed using Attenuated total reflectance- Fourier transformed IR spectroscopy (ATR-FTIR). A decreased ratio of α_β in FBS was found with increasing SPT duration with both SR-IMS and ATR-FTIR techniques. Additionally, SPT at 121 °C was effective to reduce protein and starch degradability in the rumen with different responses observed with different SPT duration in FBS among the treatments. The truly digested protein in the small intestine (DVE-MP) increased with SPT. Cows fed TMR’s with 10% inclusion of raw FBS yielded 39.1 and 43.9 kg/cow/day of milk and FCM, respectively, with 3.90% fat, 2.98% protein, and 4.55% lactose contents in the milk. Cows fed TMR’s with 10% inclusion of steam-pressure toasted FBS for 7.5 min yielded 39.9 and 44.4 kg/cow/day of milk and FCM, respectively, with 3.95% fat, 2.97% protein, and 4.53% lactose contents in the milk. Finally, cows fed TMR’s with 10% inclusion of steam-pressure toasted FBS for 15 and 30 min yielded on average 39.4 and 41.2 kg/cow/day of milk and FCM, respectively, with 3.79% fat, 2.98% protein, and 4.56% lactose contents in the milk. The optimal processing, time, and level of inclusion should be considered when FBS are used in combination with more traditional sources of protein and starch to improve the production performance of high lactating dairy cows

    The relationship between the authority of conscience and the authority of the state is a perennial issue in both political and ethical philosophy. The extreme emphasis on the authority of the state or conscience leads to the undermining of a free society. This thesis will explore the issues at play in this interaction from the perspective of natural law by exploring the proper domain and orientation of each sphere of authority. It will then demonstrate how the application of natural law principles offers a practical solution to the fraught issue of conscientious objection in a medical context. The solution that the application of natural law offers to this social issue properly respects the freedom of conscience necessary for a free society while allowing for the reasonable limits that governments need to maintain and enact the legal order necessary for a society

    Impact of Radiative Exposures on the Mechanical Properties of Fire-Resistant Fabrics

    Firefighters’ protective garments are designed to protect them from elevated temperature and heat flux environments. Typically, these garments are made of three layers: an outer shell, a moisture barrier, and a thermal liner. At present, new firefighters’ garments must meet performance criteria specified in test standards, but these do not apply to in-use garments. As the performance of this clothing may degrade with use, quantitative methods for determining the useful life of these garments without destroying them is a need for the fire service. One area of such research has been the development of correlations between near infrared (NIR) spectral results and changes in fabric properties. Research at the University of Saskatchewan (U of S) has shown that NIR measurements can be correlated to deterioration in mechanical strength after thermal ageing. This study examined the performance of two Kevlar®/PBI fabrics after exposures to heat fluxes ranging from 10 to 70 kW/m2 for durations ranging from 15 to 1200 s. These exposures were conducted using a cone calorimeter and have been selected as representative of the wide range of conditions expected on the fireground. After examination using an NIR spectrometer, tensile testing was conducted on the specimens. The tensile strength values for thermally aged fabrics were then compared against criteria in standards for new clothing. Three correlations between exposure and duration were developed based on multi-variable linear regressions, multi-variable nonlinear regressions, and single-variable nonlinear regressions. These correlations were constructed from this research and past research datasets. These correlations aim to predict the degradation these types of fabrics experience, and could be used in future degradation. It was found that the multi-variable nonlinear regression correlation was the most successful across different exposures, while the single-variable nonlinear regression was able to predict degradation more accurately to an average standard error of 4%

    "Avengers Assemble!": The Implications of Assembling an Edition of the Marvel Cinematic Universe on Textual Scholarship

    In the last couple of decades, the word “social” has become increasingly ubiquitous not just in everyday life with the advent of social media sites such as Facebook, Instagram and Twitter but also in the field of textual scholarship with the increased attention paid to the construction of “social editions.” Concomitantly, in the film world, the format of the cinematic universe is becoming one of the main modes of storytelling with unprecedented success of the Marvel Cinematic Universe (MCU). As film critics like Graeme McMillan claim that “whatever lightning Marvel captured in its onscreen bottle is not a trick that’s easily replicable,” tackling the cinematic universe might seem to be a daunting task for screenwriters and filmmakers. For textual scholars who have been struggling to create a truly “social” edition, however, the MCU’s success only presents an opportunity to achieve their goal and exponentially expand the field. Taking the MCU as a case study, this paper explores the implications of approaching a cinematic universe as a subject for textual editing and argues that because of its massively collaborative, transmedial, and inherently commercial nature as a work, the MCU invites reconsiderations of current practices in textual scholarship. In conversation with other attempts at social editing, this paper demonstrates that the collaboration and transmediality behind the MCU not only allows it to fit but also encourages the expansion of current definitions of the text. As well, it argues that attempting to edit the MCU keeps the creation of a truly “social” edition in the realm of possibility exactly because of its commerciality. Ultimately, in theorizing what an edition of the MCU would look like, this paper asserts that editing the MCU would also require a reconsideration of what “scholarly editing” means, both in terms of what constitutes “scholarly” work and what the duties of editors and editions are, and where and when they should look for meaning
