Reliability and Potential of Inkjet-Printed Flexible Heaters with Adaptive Temperature Zones for High-Temperature and Long-Time Applications
Herein, the reliability and potential of inkjet-printed flexible heaters are demonstrated by a set of resilience tests, showing never before performed high-temperature application. The flexible heaters can operate up to 400 °C and failures are not caused by the printed conductor, but due to the temperature sensitivity of the base substrate only. Furthermore, these heaters show an accurate repetition rate with fluctuations <3% (2 °C within ten up–down cycles between 20 and 150 °C) and great long-lasting capability (200 h on-time operation at 150–180 °C). The bending strength has been proven by 10 000 cycles with resistance change less than 5%. Additionally, the adaptability and adjustability of defined temperature zones with the help of design rules for the layout definition are illustrated. These inkjet-printed heaters demonstrate a high potential of usage in high-temperature and flexible applications with the prerequisite of high stability and reliability
Intraoperative data analysis for the recognition of surgical phases and patient status
Die neusten Technologiefortschritte im Operationssaal (OP) haben erhebliche Verbesserungen in der interventionellen Medizin erzielt, jedoch traten durch die damit verbundene, gestiegene Komplexität massive Defizite auf. Daher werden situationsabhängige datengesteuerte, intelligente Systeme die Zukunft der Operationssäle gestalten. Diese intelligenten Systeme zielen darauf ab, Daten von medizinischen Geräten während der Operation zu analysieren und zu fusionieren, um den Ärzten und dem medizinischen Personal zusätzliche, tiefgründige Unterstützung zu bieten. Hierdurch wird das Situationsbewusstsein im OP gefördert, die medizinischen Teams werden entlastet, potenzielle Fehler und Komplikationen können reduziert oder vermieden und chirurgische Behandlungen optimiert werden. Die in dieser Arbeit vorgestellten Konzepte und Ansätze tragen zur Lösung von zwei grundlegenden Problemfeldern bei der Transformation aktueller Operationssäle in digitalisierte und intelligente Operationssäle bei. Ein Konzept betrifft die Analyse des chirurgischen Workflows, ein Weiteres die Erkennung des Patientenstatus durch die Analyse chirurgischer und physiologischer Daten.
Videobasierte Deep-Learning-Ansätze zur Erkennung chirurgischer Phasen wurden entwickelt, die speziell auf die Analyse laparoskopischer Videos zugeschnitten sind. Die Ziele dieser Ansätze ähneln den Zielen bereits bestehender Methoden, unterscheiden sich jedoch maßgebend in der Implementierung. Erste Ansätze umfassten die Erkennung von Phasen mit Hilfe einer hierarchischen dynamischen Neural-Network-Pipeline, eines Convolutional Neural Network (CNN) in Kombination mit einem nicht-linearen autoregressiven exogenen (NARX) neuronales Netzwerk oder Long Short-Term Memory (LSTM) Netzwerk. Bei weiterführenden Ansätzen wurde ein temporaler Attention- und Multi-Feature Fusions-Ansatz entwickelt, um die Erkennung chirurgischer Phasen, die Klassifizierung chirurgischer Instrumente und die schwach überwachte Lokalisierung von Werkzeugen durchzuführen. Des Weiteren wurde ein heterogener Datensatz erstellt, welcher Daten aus Chirurgie und Anästhesiologie während laparoskopischer Eingriffe enthält. Die potenziellen Vorteile dieses multidisziplinären Datensatzes wurden beispielhaft, anhand der klinisch relevanten Korrelation zwischen intraabdominalem Druck und der Lungenmechanik bei Patienten, dargestellt. Eine weiterführende Machbarkeitsstudie befasste sich mit einem computergestützten System, welches die automatische Generierung von Anästhesieberichten während laparoskopischer Operationen erlaubt.
Die in dieser Arbeit implementierten Ansätze und Frameworks haben verschiedene potenzielle klinische Anwendungen, angefangen von der Optimierung des OP-Plans bis hin zum Situationsbewusstsein. Zum Beispiel könnten die entwickelten, video-basierten Ansätze implementiert werden, um Anästhesisten situationsabhängige Informationen zu visualisieren. Darüber hinaus schafft die statistische Korrelationsanalyse zwischen chirurgischen und anästhesiologischen Daten die wissenschaftliche Grundlage für viele Weiterentwicklungen des medizinischen Setups im OP. Individuelle Reaktion von Patienten / Patientinnen auf chirurgische Maßnahmen könnten in Echtzeit an Chirurgen und Anästhesisten übermittelt werden. In diesem Kontext könnte eine Vorhersage von Veränderungen der lungenmechanischen Eigenschaften der Patienten / Patientinnen ermöglicht werden, welche durch den, während des Eingriffs erhöhten, intra-abdominalen Druck induziert wird. Aus diesen Datenströmen könnten wiederum optimale Beatmungseinstellungen automatisch abgeleitet werden, um Anästhesisten zu unterstützen
Pharmacological Studies on TRPC6 Inhibitors
Based on the substantial challenges that precede the introduction of newly developed compounds for potential clinical application, comprehensive pharmacokinetic studies are crucial. These studies comprise the absorption, distribution, metabolism, and excretion (ADME) properties together with analysis of bioavailability and observations regarding their predicted effectiveness in preclinical models and possible toxicity are crucial to ensuring their safe and effective use in clinical practice. Recently, the diterpene (+)-larixol isolated from the conifer Larix decidua resin traditionally used for inhalation underwent a comprehensive chemical diversification and structure-related activity analysis. Its methylcarbamate congener, named SH045, was identified in functional assays as a highly potent, subtype-selective inhibitor of TRPC6. Therefore, the present work was dedicated to developing a method to analyze SH045 in biological samples, to use this method for the pharmacokinetic characterization of this compound, to look for signs of toxicity and finally to prove the inhibitory effectiveness of SH045 in an animal model of acute kidney injury (AKI). The results of these studies are published in three consecutive studies summarized below (Chai et al., 2021; Chai et al., 2022; Zheng et al., 2022a). As a requirement for future research and to pave the way for use of SH045 in animal disease models, this study aimed to develop a capable bioanalytical method and provide exploratory pharmacokinetic data for this promising derivative. Initially, a highly sensitive and specific LC-MS/MS method with enhanced precision was developed for quantification of SH045 in plasma samples. The Enhanced Product Ion (EPI) spectrum and optimized MS parameters were obtained to facilitate detection. Protein precipitation, a simple and cost-effective approach, was performed to extract SH045 from plasma. Notably, the method required only a small plasma volume (20 μl) for the LC-MS/MS analysis, enabling repeated blood sampling from a single mouse, thereby providing more robust and informative data while minimizing the number of animals required for the study. In accordance with the Bioanalytical Method Validation guidelines from the European Medicines Agency (EMA) and USA Food and Drug Administration (FDA), a comprehensive validation was conducted for the developed LC-MS/MS method of SH045 to ensure reliable routine results. The validation parameters evaluated included selectivity, carry-over, linearity, limit of detection (LOD), the lower limit of quantification (LLOQ), accuracy, precision, matrix effect, recovery, stability, and dilution integrity in the range of 2-1600 ng/ml. The LLOQ was determined to be 2 ng/ml (equivalent to 5.5 nM), which was slightly lower than the compound's IC50 for inhibiting TRPC6 current (5.8 nM), indicating the high sensitivity of the quantitative analysis. All validation results complied with the standards documented in the guidelines. The validated LC-MS/MS method was applied in an exploratory pharmacokinetic study in mice following a single intraperitoneal (i.p.) administration of SH045 at 20 mg/kg. As a result, SH045 displayed a relatively short primary half-life and rapid absorption after intraperitoneal administration. However, these may not limit the application of the analyte, as the plasma concentration after 5 hours was still higher than the IC50. Moreover, the high plasma protein binding may suggest a prolonged release of SH045 from plasma protein and tissue protein after distribution from the vascular system, which might affect the biological process of SH045. The large steady-state distribution volume of 25 l/kg is similar to the volume of distribution during the terminal phase of 25.5 l/kg, indicating that the compound strongly prefers the extravascular space. To obtain comprehensive pharmacokinetic data to support future studies, we thoroughly investigated the analyte's pharmacokinetic properties, including plasma kinetics, tissue distribution, elimination, and metabolism. Among the calculated plasma kinetics parameters, the extrapolated area under the plasma concentration-time curve from the last measurable time point to infinity was less than 6%, indicating that the measured time points sufficiently evaluated the pharmacokinetic parameters. Consistent with the exploratory pharmacokinetic study, SH045 exhibited a primary half life of 1.2-2.4 h in mice across different routes of administration and dosages. As expected by the high lipophilicity, SH045 showed extensive tissue distribution with preferential accumulation in the lungs compared to other tissues after intravenous administration, suggesting potential application for lung-targeted therapies in vivo. Five times repeated administrations, once daily via intravenous route, were expected to achieve steady-state levels to maintain therapeutic concentrations. No accumulation of SH045 was observed in the plasma or in tissues after repeated intravenous administration, indicating high elimination and metabolism, corroborated by the high concentration in urine samples. Despite the extensive tissue distribution, no acute tissue toxicity was observed in the liver, kidney and lung after single or repeated administration, as assessed by hematoxylin-eosin staining, suggesting the applicability of a higher dosage of SH045. The low bioavailability of the compound after oral administration may be due to first pass metabolism, prompting an investigation of the metabolic fate of SH045. When incubated with mouse liver microsomes (MLM) or human liver microsomes (HLM) in the presence of phase I and phase II cofactors in vitro, SH045 underwent rapid metabolism. The relatively high predicted hepatic clearance values, especially in mice, suggest that hepatic metabolism could be a major elimination pathway for SH045 in vivo. We then investigated several primary metabolic pathways of SH045 in HLM and MLM. Among the major cytochrome P450 enzymes, SH045 was primarily metabolized by CYP3A4 and CYP2A6, providing metabolites via oxidation reactions. With our experience in the quantification of SH045, we established an analysis method for all metabolites detected after incubation with liver microsomes using LC-MS/MS. Among the metabolic reactions, hydroxylation was the predominant pathway, along with reduction and phase II glucuronidation, indicating that the metabolism enhancing the drug's water solubility plays a significant role in the systemic clearance of SH045. Among these metabolites, a TRPC6 inhibitor with similar potency and prolonged half life might exist. To investigate this possibility, we performed a simple experiment with a FLIPR assay. However, no inhibition of Ca2+ influx in HEKhTRPC6-YFP cells by the metabolite solution after microsomal exposure of SH045 was found (data not shown). Having established the pharmacokinetic profile of SH045 and having observed that plasma and tissue concentrations of SH045 are still above the IC50 at 24 h after 20 mg/kg body weight intravenous administration, we consequently proved the effectiveness of SH045 in a preclinical model of AKI, a highly clinically relevant disorder involving TRPC6. These findings provide valuable insights into SH045 as a potential treatment option for symptoms found in the clinical setting and suggest further possibilities for SH045 or related compounds with improved pharmacokinetic features as a therapeutic option. Apart from these published data, further work was done on so-called photoswitchable inhibitors of TRPC6. Such compounds undergo reversible activity-related structural changes upon light exposure, enabling the modulation of target protein activity on demand (see Supplementary material). In summary, we developed a highly sensitive, selective and accurate LC-MS/MS method to quantify SH045, a second generation compound of TRPC6 blockers derived from a natural product, and its metabolites in biological samples. We have provided a comprehensive pharmacokinetic study of SH045. Despite the short primary half-life and low bioavailability after oral administration, the plasma concentration of SH045 (20 mg/kg) from the last collection was still higher than its IC50. Further, an extensive tissue distribution was observed. No acute tissue toxicity was found even after repeated administration, suggesting that a higher dosage of SH045 may be applicable. Together with a successful treatment of mice with tubulointerstitial fibrosis, SH045 may have a potential also to treat other TRPC6-related disorders. Dealing with the third generation of TRPC6 inhibitors it was shown that the photoswitchable compound azoLarixol acts as a highly potent and selective inhibitor that can modulate TRPC6 activity by light illumination. Moreover, it displayed a similar impact on native TRPC6 in rat pulmonary artery smooth muscle cells (rPASMCs), suggesting further suitable applications to investigate the mechanism or role of TRPC6 in physiological and pathological processes
Toward ‘Cultures of Engagement’? An exploratory comparison of engagement patterns on Facebook news posts
Information production, dissemination, and consumption are contingent upon cultural and financial dimensions. This study attempts to find cultures of engagement that reflect how audiences engage with news posts made by either commercial or stateowned news outlets on Facebook. To do so, we collected over a million news posts (n = 1,173,159) produced by 482 news outlets in three Scandinavian countries (Denmark, Norway, and Sweden) and analyzed over 69 million interactions across three metrics of engagement (i.e. comments, likes, and shares). More concretely, we investigate whether the patterns of engagement follow distinct patterns across national boundaries and type of outlet ownership. While we are skeptical of metrics of engagement as markers of specific cultures of engagement, our results show that there are clear differences in how readers engage with news posts depending on the country of origin and whether they are fully state-owned or private-owned outlets
A Mechanochemically Active Metal-Organic Framework (MOF) Based on Cu-Bis-NHC-Linkers: Synthesis and Mechano-Catalytic Activation
Porous coordination polymers, more commonly known as metal-organic frameworks (MOFs), are constructed from metal ions and organic linkers which form a robust network-like structure similar to very dense polymeric networks. In this work the mechanochemical activation of a MOF is reported, based on the previous results on the mechanochemical activation of latent Cu(I)-based N-heterocyclic carbenes (NHCs) embedded covalently in various polymers. A mechanochemically triggered copper-catalyzed azide-alkyne cycloaddition (CuAAC) is achieved using an NHC-MOF, constructed from 1,3-bis(4-carboxyphenyl)imidazolium chloride as organic linker. A Cu(I) bis(NHC) is embedded into the MOF analogous to ([Zn4O{Cu(L)2}2]) using the organic bis-NHC ligand, in turn generating a largely amorphous MOF. On activation by ultrasound, the MOF containing 4.66% of Cu shows mechanochemical activation to obtain Cu(I), subsequently triggering a CuAAC with conversions up to 26.5% as monitored by the reaction of benzylazide and phenylacetylene to yield 1-benzyl-4-phenyl-1H-1,2,3-triazole over a period of 20 sonication cycles. With this knowledge in solution, there are prospects to transfer this mechanochemical activity into polymer networks with embedded MOFs to detect stress in hybrid materials

Eine de novo Variante im Gen CNOT9 war besonders überzeugend und somit wurde ein neues Krankheitsbild beschrieben, welches zu Entwicklungsverzögerung und Epilepsie führt
Parasoziale Interaktionen und kollektive Kreativität: Eine Fallstudie der Taylor Swift Fan Community: Eine quantitative Untersuchung zum Einfluss digitaler Fan- Interaktionen und gemeinschaftlicher Kreativität auf das Gemeinschaftsgefühl bei Live- Konzerten
Digitale Medien haben die Beziehung zwischen Künstler:innen und ihren Fans grundlegend verändert. Besonders in Online-Fangemeinschaften sind dynamische Interaktionsformen entstanden, die über bloßen Konsum hinausgehen und eine aktive Teilhabe an kreativen und gemeinschaftsstiftenden Prozessen ermöglichen. Diese Bachelorarbeit untersucht, wie parasoziale Interaktionen, kollektive Kreativität und transmediales Storytelling innerhalb der Taylor-Swift-Fangemeinschaft zusammenwirken und welchen Einfluss sie auf das Zugehörigkeitsempfinden der Fans haben.
Der theoretische Rahmen stützt sich auf Konzepte der parasozialen Interaktion, kollektiven Kreativität und des transmedialen Storytellings. Er zeigt auf, wie Fans digitale Plattformen nicht nur zur Entwicklung emotionaler Bindungen an Künstler:innen nutzen, sondern auch aktiv zur Fankultur beitragen – etwa durch kreative Aktivitäten und geteilte narrative Praktiken. Im Fall von Taylor Swift zeigt sich dieses Zusammenspiel beispielsweise in Fanchants, selbstgebastelten Freundschaftsarmbändern oder der gemeinschaftlichen Entschlüsselung sogenannter „Easter Eggs“.
Die Studie basiert auf einer quantitativen Online-Umfrage (N = 306), die untersucht, wie parasoziale Bindungen mit kreativer Teilhabe und dem Gemeinschaftserleben bei Live-Konzerten zusammenhängen. Die Ergebnisse zeigen, dass parasoziale Interaktionen das Zugehörigkeitsempfinden deutlich stärken. Kreatives Engagement wirkt hingegen selektiver und entfaltet besonders dann Wirkung, wenn es ritualisiert auftritt. Transmediales Storytelling fungiert als strukturierender Rahmen, in dem emotionale Nähe und kollektive Ausdrucksformen zusammenlaufen. Die Arbeit leistet einen empirisch fundierten Beitrag zur Fankulturforschung, indem sie das Zusammenspiel von emotionaler Bindung, kreativer Beteiligung und medialer Gemeinschaftsbildung beleuchtet.
Abstract
Digital media have fundamentally transformed the relationship between artists and their fans. Especially within online fan communities, dynamic forms of interaction have emerged that go beyond consumption and enable active participation in creative and community-building processes. This thesis investigates how parasocial interactions, collective creativity, and transmedia storytelling intersect within the Taylor Swift fan community and how they influence fans’ sense of belonging.
The theoretical framework draws on the concepts of parasocial interaction, collective creativity, and transmedia storytelling. It highlights how fans use digital platforms not only to develop emotional bonds with artists, but also to contribute to fan culture through creative activities and shared narratives. In the case of Taylor Swift, this interplay becomes visible in practices such as fanchants, friendship bracelets, and the collaborative decoding of so-called 'Easter Eggs.'
The study is based on a quantitative online survey (N = 306) examining how parasocial bonds relate to creative participation and community experience at live concerts. The findings show that parasocial interactions significantly enhance the feeling of belonging. Creative engagement, by contrast, appears to have more selective effects and is particularly impactful when ritualized. Transmedia storytelling serves as a structuring framework in which emotional closeness and collective modes of expression converge. This thesis contributes to fan culture research by empirically demonstrating how emotional attachment, creative participation, and mediated community are interconnected
Intentions in Prosody: Investigations on Expression, Comprehension and Neural Bases of Social Intentions in Speech
Für unsere Kommunikation ist es zentral, die Absichten unseres Gegenübers zu verstehen. Obwohl die Intention, warum etwas gesagt wird, der Kern einer Unterhaltung ist, drücken wir sie oft nur indirekt und nicht in den gesagten Worten aus. Nichtsdestotrotz ist das gegenseitige Verständnis meist reibungslos, was darauf schließen lässt, dass weitere Mechanismen an unserer Wahrnehmung von Intentionen im Sprachsignal beteiligt sind. Ziel der vorliegenden Arbeit war es, herauszufinden, wie Prosodie zur Übertragung der Intention einer Sprecherin oder eines Sprechers beiträgt und welche neuronalen Korrelate am Intentionsverständnis durch Prosodie beteiligt sind. Dazu wurde das prosodische Signal verschiedener Intentionsausdrücke in einer Reihe von akustischen und perzeptuellen Messungen analysiert (Experimente 1 bis 3). Darüber hinaus wurde untersucht, welche Hirnregionen und kognitiven Mechanismen aktiviert werden, wenn Hörer:innen die Absicht der Sprecher:innen durch das prosodische Signal wahrnehmen (Experiment 4). Die Ergebnisse der Experimente zeigten, dass es charakterische akustische Konfigurationen für den Ausdruck von kommunikativen Absichten des Gesprächspartners gibt. Die intentionalen Prosodien wurden von Hörer:innen zuverlässig erkannt. Außerdem beruht die Wahrnehmung von Intentionen in Prosodien auf verteilten neuronalen Netzwerken, die funktionell miteinander verbunden sind. Neben Arealen der auditorischen und konzeptuellen Abstraktion des eingehenden Signals entlang des ventralen auditorischen Pfades waren auch Theory-of-Mind-Areale an dem Verstehen von intentionalen Prosodien beteiligt. Dies deutet auf ein Modell hin, in dem auditorisch-prosodische Kategorisierung und soziale Inferenzprozesse zusammenarbeiten, um die wahrgenommene Prosodie in eine kohärente Repräsentation der Sprechabsicht zu übersetzen. Die Erkenntnisse dieser Studien zeigen auf, dass Prosodie ein wirkungsvoller Kanal für erfolgreiche Kommunikation ist
