Tremor, sensory-motor polyneuropathy and cerebral gliosis in klinefelter's syndrome
Klinefelter's syndrome (KS) represents one of the most frequent sexual chromosome abnormalities, with an extra-X chromosome added to the normal karyotype (XXY). Clinically, it is characterized by tall body habitus, infertility, gynecomastia, small penis and testes. Neurological involvement is usually present and tremor is frequently described as postural and kinetic tremor that resembles an essential tremor. Polyneuropathy is another neurological problem described in KS. It is an axonal neuropathy with the involvement of sensory-motor fibers. It may represent a complication or alternatively a coexisting morbidity. Cerebral gliosis in magnetic resonance images (MRI) was also described, with lesions in deep white matter (white matter lesions -WMLs). Sexual chromosome alterations with an increased number of chromosomes (XXXY, XXYY, etc.) more often present these MRI abnormalities. We describe three cases of KS patients carrying these alterations: tremor, sensory-motor polyneuropathy and leukoencephalopathy. These three abnormalities should be considered in all patients with KS
Characterizing the agroecological transition of Italian farming systems using FADN database
The Common Agricultural Policy 2023-2027 encourages the transition towards sustainable agriculture in line with the goals of the European Green Deal. Agroecology is gaining recognition as a valuable approach to enhance the sustainability of faming systems. However, there is limited understanding of the status quo of the farming systems and their readiness for transition, especially at large scales such as the national one. This paper investigates the current uptake of agroecology across the Italian farming systems, by adapting the Tool for Agroecology Performance Evaluation (TAPE) methodology to use secondary data from the Farm Accountancy Data Network (FADN). Besides, a panel between-variance regression was estimated to identify the key determinants of the transition degree. The results indicate that agroecology has not yet been widely adopted by the Italian farms. Additionally, factors such as the type of farming, farm management, farm economic and physical size, farm location, and farmer's socio-demographics play a significant role in explaining the variation in the transition intensity towards an agroecological production system. The findings provide a knowledgebase for developing targeted policies and monitoring progress in agroecological transition. The study confirms the viability of using the FADN to evaluate the degree of agroecological transition, while also revealing information gaps. Although essential environmental and social information will be included in the forthcoming Farm Sustainability Data Network, further integrative sources should be explored to capture multifaced aspects of agroecological transition within broader agri-food systems, while in-depth studies based on primary data collection could offer more detailed and nuanced insights

Uncertainties in anti-amyloid monoclonal antibody therapy for Alzheimer’s disease: the challenges ahead
Introduction: Alzheimer’s disease (AD), the leading cause of dementia, poses a significant burden on patients, caregivers, and healthcare systems worldwide. After two decades of extensive efforts, we are still without significantly effective disease-modifying drugs for AD. Although brain amyloid-β (Aβ) accumulation may predict cognitive decline, several drug candidates, including anti-Aβ monoclonal antibodies, have been developed and tested to reduce Aβ plaque burden effective, but without significant clinical success. Areas covered: The following review presents and discusses anti-Aβ monoclonal antibody therapeutics used to treat AD. The article considers both current approaches and alternatives. This article is multiple database searches (MEDLINE, EMBASE, Scopus, Ovid and Google Scholar) on all the available literature up to 1 February 2025. Expert opinion: Randomized clinical trials (RCTs) of anti-Aβ drugs in AD have not fully validated the Aβ cascade hypothesis. Nevertheless, eight anti-Aβ monoclonal antibodies have, thus far, made it to Phase III RCTs. Moving forward, the use of the Apolipoprotein E genotype and tau protein as alternative biomarkers can assist clinicians in providing patients with even more individualized and efficacious anti-Aβ monoclonal antibodies dosing regimens and reduce the risk of serious amyloid-related imaging abnormalities

High-throughput and sustainable B vitamins analysis in nutritional supplements, vegetables, and fruits via 2D carbon microfiber fractionation system coupled with mass spectrometry
B vitamins are essential for energy metabolism, nervous health, blood production, and the immune system. Their quantification in nutritional supplements and food is mandatory to manage a correct daily intake and dosage. In this study, a fast and sustainable method for the analysis of 8 B vitamins (VB1, VB2, VB3, nicotinamide, VB5, VB6, VB9, VB12) in real samples using a 2D-carbon microfiber fractionation system combined with a triple quadrupole mass spectrometer (2DμCFs-QqQ-MS/MS) is presented. The method shortens the analysis time to 5 min, showing high sensitivities (LODs: 1.7–13.7 ng mL−1, LOQs: 5.5–45.7 ng mL−1, obtained with standard solutions), good reproducibility (RSDs <15 %), and strong linear correlations (R2 > 0.997). When applied for B vitamin determination in nutritional supplements, results matched product label values at a 95 % confidence level, with batch reproducibility never exceeding 5 %. Furthermore, coupling a 2D-carbon microfiber fraction system with a quadrupole time-of-flight high-resolution mass spectrometry (2DμCFs-QTOF-HRMS) it is possible to carry out quick monitoring of B vitamin contents together with impurity evaluation in real samples
