    In un contesto in cui in ambito sanitario, le competenze umane rischiano di essere superate dalla tecnologiae ridotte a meri strumenti funzionali alla produttività, questo studio sperimentale, radicato nella pedagogiadella formazione in area medica, indaga su un percorso educativo e pedagogico nelle Medical Humanities.L’analisi di tale Best Practice è finalizzata a valutare le competenze relazionali e comunicative del personalesanitario impegnato in un percorso di laurea magistrale in Scienze delle Professioni Sanitarie Tecnico-Dia-gnostiche presso l’Università di Foggia.     Advanced Imaging and Cytometric Techniques to Characterize Lipid Accumulation in Wolman Disease

    Wolman disease (WD) is a severe lysosomal storage disorder characterized by fatal lipid accumulation caused by the deficiency of a lipid metabolic enzyme, Lysosomal Acid Lipase (LAL), involved in the lysosomal hydrolysis of cholesterols and triglycerides. Due to the imbalance of lipid homeostasis, WD patients suffer from severe hepatosplenomegaly, hepatic failure, and adrenal calcification resulting in a premature infant death within the first year of age. In this work, we explored multiple imaging analyses to fully characterize the phenotype of LAL-deficient cells. In particular, we stained WD patients' fibroblasts for intracellular lipid droplets (LD) and lysosomes, and we analyzed staining intensity and granularity, as well as an increased number of LD and lysosomes using fluorescence wide-field microscopy, confocal microscopy, conventional, and image flow cytometry. Noteworthy, we showed that lipid homeostasis was restored upon delivery of a functional LAL transgene. Finally, since fibroblasts cannot be used as routine clinical tests as they are difficult to collect from WD patients, we confirmed our observations in LAL deficient human blood cell lines and in peripheral blood mononuclear cells (PBMC) from the LAL deficient (LAL-D) mouse model, as a proxy for easily accessible WD PBMC. Overall, we expect that this novel imaging analysis pipeline will help to diagnose WD, follow its progression, and evaluate the success of enzyme replacement therapy or gene correction strategies for WD as well as other lysosomal storage disorders

    Comparative Analysis of SiMoA and Ella Immunoassay Platforms for Measuring Serum Neurofilament Light Chain Levels in ATTRv With Polyneuropathy and Presymptomatic Carriers

    Background: Neurofilament light chains (NfL) represent reliable serum biomarkers of neuroaxonal injury. Due to their low serum levels, precise detection methods are critical. This study aimed to scrutinize the comparability of two techniques: Single Molecule Array (SiMoA) and Ella automated immunoassay, analyzing serum NfL levels in ATTRv presymptomatic subjects and polyneuropathy patients. Methods: Blood samples were processed and analyzed using commercial Ella and SiMoA kits. Statistical analysis included the Wilcoxon signed-rank test, Spearman correlation, Bland–Altman, and Passing-Bablok regression. ANCOVA models were used to compare NfL levels between cohorts. Results: The study measured serum NfL levels in 55 symptomatic and 55 presymptomatic ATTRv subjects. Median NfL concentrations were significantly higher with Ella (median 27.5 pg/mL) than SiMoA (median 15.9 pg/mL). Both methods showed a strong positive correlation (r = 0.8, p < 0.001), but Ella overestimated NfL by 42%. Bland–Altman analysis revealed a mean bias of 15.4 pg/mL, with limits of agreement between −41.1 and 72.0 pg/mL. The slope of the Passing–Bablok regression line was 0.58, and the intercept was 3.48 pg/mL, suggesting that the Ella platform tends to overestimate NfL concentrations compared to the SiMoA platform, especially at higher concentrations. Both methods effectively distinguished presymptomatic from symptomatic patients (p < 0.001 for both). Conclusions: Our findings underscore that both platforms are effective in measuring serum NfL, with Ella consistently overestimating, especially at higher concentrations. The difference between the two platforms must be taken into account when deeming the concentrations as pathological or normal, as well as when conducting longitudinal studies

    Role of blockchain in shaping trust and purchase intentions for beauty brands advertised online

    Corporate reputation is the public’s perception of a business and how people perceive an organization with their products, services, and behaviors. In the beauty industry, consumers are known to be more curious and conscious of the broader impact of their buying decisions. With significant purchasing power, consumers now prioritize safety and health considerations when it comes to beauty care. Our empirical study investigates the role of blockchain technology on consumers' perceptions of quality, consumers' trust, and their intention to buy beauty brands advertised on online beauty platforms, through an experiment. Drawing broadly on the theories of swift trust and trust-based marketing, the findings of the moderated-mediation analysis highlight that the application of blockchain technology on e-commerce platforms would lead to higher buying intentions for unpopular beauty brands than for popular beauty brands. The study is empirical evidence that highlights how blockchain's transparency and traceability characteristics could be important indicators of consumers' perceptions and trust in a beauty product brand which may lead to better brand image and corporate reputation

    Prevalence and Antimicrobial Resistance of Salmonella Strains Isolated from Chicken Samples in Southern Italy

    Salmonellosis is one of the most frequent foodborne zoonoses, and the consumption of contaminated poultry meat is considered the main source of Salmonella infections in humans. From 2021 to 2023, 384 chicken samples were collected from retailers in Apulia and Basilicata regions and analysed at the Istituto Zooprofilattico Sperimentale della Puglia e della Basilicata (IZSPB) laboratories. The Salmonella isolates were investigated to evaluate their phenotypic characteristics of antimicrobial resistance. A total of 125 (32.55%) samples tested positive for Salmonella spp. Three samples were simultaneously infected with strains of two different serotypes. Strains were classified into nine serotypes. Out of 128 strains, 112 (87.5%) isolates showed multidrug-resistant (MDR) profiles. Moreover, 16 isolates (12.5%) were ESBL producers with MDR profiles. Our data confirm that chicken products are a common source of Salmonella and highlight how S. Infantis was the most prevalent serotype (85.93%). Furthermore, Salmonella isolates showed antimicrobial resistance, which is very worrisome for animals and consumers. The high rate of antibiotic resistance along with the appearance of numerous MDR isolates is considered an actual concern and requires a robust surveillance network in a One Health concept
