Palytoxin-like compounds, including ovatoxins, are potent emerging toxins responsible for human respiratory poisonings following inhalation of contaminated marine aerosols. Periodic massive proliferations of the ovatoxin-producing organism (Ostreopsis cf. ovata) worldwide, particularly in the Mediterranean, have caused severe toxic outbreaks, drawing the attention of health authorities. At present, an efficient and sustainable sampling system for monitoring ovatoxins in seawater remains unavailable. Herein, different cyclodextrin (CD) polymers were investigated as a green and effective alternative to conventional and low-performing resins to detect ovatoxins in seawater. Spiking experiments using different concentrations of palytoxin or ovatoxins (namely 200 and 3.3 ng PLTX/mL or 200 ng OVTX-a/mL) were conducted and LC-HRMS was used to evaluate the suitability of CD polymers in capturing palytoxin-like compounds. Several conditions were tested for extracting polymer materials, including different extraction times (1.5 to 4 h), various solvent mixtures (acidic or alkaline), and organic modifiers (methanol or acetonitrile) at different ratios. Among the tested polymers, γ-CD-hexamethylene diisocyanate (HDI) resulted to be the most promising one, providing ovatoxin recoveries in the range 82-108% at a spiking level of 200 ng OVTX-a per mL. The best extracting condition was alkaline pH methanol:water 8:2 mixture, which showed the best palytoxin recovery in both high and low concentration spiking experiments. Finally, a time-dependent increase in the amount of ovatoxins captured by γ-CD-HDI disks deployed in O. cf. ovata cultures was observed. These findings provide valuable insights on the efficiency of passive sampling using CD polymers for capturing ovatoxins during O. cf. ovata bloom events

Background. Mud volcanoes are unique natural structures affecting the environment by continuous emissions of gas-liquid fluids, mud breccia and mudflows. Despite extensive research and a substantial dataset on mud volcanism, the understanding of the fluid dynamics and hydrogeological processes associated with these systems remains a subject of ongoing debate. Globally, there are more than 2,500 mud volcanoes distributed across 42 countries (Aliyev et al., 2015), highlighting their widespread occurrence. Azerbaijan represents a unique region for the study of contemporary mud volcanism, hosting more than 350 mud volcanoes within a relatively small territory. Among them, the Lokbatan mud volcano is the most active, with 28 recorded eruptions. In this context, a comprehensive approach to studying the activity of mud volcanoes, including satellite monitoring, is highly relevant. Methods. The study employed remote sensing techniques to analyze the hydro-fluid regime of the Lokbatan mud volcano. Specifically, satellite imagery was used to calculate a moisture index for the volcano field. For the first time, this index was remotely measured, enabling assessment of surface moisture dynamics as a proxy for fluid migration and subsurface activity. Results. The satellite-derived moisture index showed clear correlations with mud volcanic activity. Observations indicate that variations in the moisture index correspond to shifts in the fluid regime of the volcano, reflecting possible underground fluid migration or pressure changes prior to eruptions. Conclusions. The study demonstrates the potential of satellite-based monitoring in assessing the hydro-fluid regime of active mud volcanoes. The approach provides an effective, low-cost alternative to field-based observations and can support early warning systems for seismic or eruptive events. These findings contribute to the development of preventive strategies for managing geological hazards in seismically active regions

Starving the invader: disrupting iron uptake to defeat Staphylococcus aureus

    No full text
The rapid emergence of antibiotic-resistant Staphylococcus aureus strains underscores its role as a key contributor to the global antimicrobial resistance (AMR) crisis. A key factor in its virulence and persistence during infection is the ability to acquire iron, an essential nutrient restricted by the host’s defence mechanism known as nutritional immunity. To circumvent host-imposed iron limitation, S. aureus employs two sophisticated iron acquisition systems: the iron-regulated surface determinant (Isd) system, which scavenges heme-bound iron directly from host hemoglobin (Hb), and the siderophore-mediated system, which captures iron from host iron-binding proteins via small chelators, primarily staphyloferrin B (SB), only produced by the most invasive, coagulase-positive S. aureus strains. Here, we present two innovative strategies targeting S. aureus iron metabolism with potential therapeutic applications. The first involves a newly-identified compound (C35)1 that selectively binds Hb and inhibits its interaction with the IsdB receptor. Using both wild-type S. aureus and its isogenic in frame-deletion mutant (ΔisdB), C35 was shown to markedly inhibit S. aureus growth by blocking hemophore-mediated iron uptake, an essential pathway for survival during infection. The second approach targets SbnA, a key enzyme in SB biosynthesis. Searching for citrate (i.e., SbnA physiological inhibitor) analogues, the methyl esters of 2-phenylmaleic acid (2-PhMA) and of 2-phenylsuccinic acid (2-PhSA) were found to impair siderophore-dependent iron acquisition in S. aureus, and its subsequent growth under iron starvation2. Together, these dual approaches targeting both heme- and siderophore-mediated iron uptake pathways provide a robust foundation for the development of next-generation antimicrobial agents against S. aureus multidrug-resistant strains
