Impact of Waste-HydroChar on the Rheological Behavior, Physical Properties, and Aging Resistance of Bitumen
In line with circular principles and the reuse of waste products, this study investigates the use of a waste-derived additive sourced from civil waste to modify the rheological and physical properties, as well as the aging resistance, of bitumen. Different dosages of waste-hydrochar (HC), produced via hydrothermal carbonization of digested sewage sludge, specifically 2%, 4%, and 10% by weight, were introduced to the bitumen, and the materials were characterized in terms of their rheological, physical, and aging behavior. Two aging protocols, e.g., short-term thermal aging and UV irradiation aging, were followed to evaluate the aging resistance of the bitumen with and without waste-hydrochar. The results obtained suggest that bitumen containing waste-hydrochar exhibits similar rheological and physical properties to bitumen without an additive, indicating the potential for using this waste material as a suitable bitumen additive. Furthermore, the presence of waste-hydrochar does not reduce the short-term thermal or UV irradiation resistance of bitumen, again suggesting the potential for using this waste material as a suitable bitumen additive. Finally, the results obtained have been compared with those of bitumen containing high-cost biochar, highlighting the potential to replace high-cost biochar with low-cost, waste-hydrochar

Beyond the stop: Oxadiazole TRIDs restore LRBA protein expression in nonsense-driven primary immunodeficiency
Nonsense mutations are among the genetic causes of LRBA (lipopolysaccharide-responsive beige-like anchor) deficiency, a rare autosomal-recessive immunodeficiency disorder. These mutations introduce premature stop codons, leading to the loss of LRBA protein expression. Following the recent market withdrawal of ataluren, the only approved translational readthrough-inducing drug (TRID), there is an urgent need for alternative therapeutic options. In this study, we investigated the efficacy of three 1,2,4-oxadiazole-based TRIDs—NV848, NV914, and NV930—using primary fibroblasts from a patient homozygous for the R1683X nonsense mutation. All compounds restored full-length LRBA protein with correct cytoplasmic localization, as confirmed by western blot and immunofluorescence, outperforming ataluren in readthrough efficiency. NV848 exhibited the strongest activity and uniquely increased LRBA mRNA levels, suggesting transcript stabilization. In contrast, NV930 and NV914 induced readthrough without stabilizing mRNA. Global proteomic profiling revealed minimal off-target effects for NV848, limited protein modulation by NV914, and widespread variations of 828 proteins by NV930, affecting pathways related to vesicular transport and mRNA splicing. However, network analysis revealed poor connectivity among differentially expressed proteins, with LRBA unrelated to any regulated cluster. These findings highlight the reported molecules as promising candidates for precision therapy in LRBA deficiency and shed light on the broader cellular impact of TRIDs
Defining the role and competencies of the medical oncologist in diagnostic and therapeutic care pathways: Consensus recommendations from the Italian association of medical oncology (AIOM)
Background: Diagnostic and Therapeutic Care Pathways (DTCPs) are clinical governance tools aimed at managing the care of specific patient populations through the coordinated application of standardized, evidence-based interventions by multidisciplinary teams. Their primary goal is to ensure equitable, timely, and cost-effective access to high-quality care. Materials and method: A panel of recognized opinion leaders, endorsed by the Italian Association of Medical Oncology (AIOM), was convened to develop a consensus document defining the role of the medical oncologist within Multidisciplinary Oncology Groups (MOGs) and DTCPs. Employing the RAND/UCLA Appropriateness Method in its "consensus conference" format, the panel evaluated a series of statements derived from a review of the scientific literature and expert-generated Good Practice Points (GPPs). These statements addressed five key areas. To further support each topic, illustrative case studies were presented. Results: A total of 21 articles met the inclusion criteria, yielding 88 evidence-based recommendations. Additionally, panel members contributed 9 further GPPs based on clinical expertise. Of the 97 total recommendations, 95 received a relevance score above 7, while 2 scored between 4 and 6.9; none scored below 4. Conclusions: This consensus effort and the resulting document represent a comprehensive evaluation of the available evidence regarding the role of medical oncologists within MOGs and DTCPs. The objective is to propose standardized criteria for the optimal management of cancer patients (pts) across all phases of care from initial diagnosis and staging to treatment, follow-up, and end-of-life support

Le tecnologie immersive di realtà virtuale (VR), realtà aumentata (AR) e metaverso rappresentano innovazioni che stanno ridefinendo le modalità di interazione con i contenuti digitali, delineando nuove frontiere per la creazione di valore aziendale e ponendo questioni rilevanti ai tradizionali modelli di valutazione economica. Il capitolo affronta la problematica della valutazione di tali sistemi, sia come asset tecnologici autonomi che come componenti del valore aziendale complessivo. La valutazione di queste tecnologie richiede una base informativa multidimensionale che integra dati tecnico-economici, informazioni di mercato competitivo, metriche di engagement utente e fattori normativi socio-culturali. Le metodologie applicabili spaziano dagli approcci tradizionali (flussi di cassa scontati, multipli di mercato, asset-based) a tecniche più innovative come l’analisi delle opzioni reali e l’approccio venture capital.

An important case study of industrial archaeology in the Vesuvian area is the connection between the "classical" site of Oplontis and the "industrial" complex of the Royal Arms Factory of Torre Annunziata, the Vanvitellian plant, cutting-edge for its time, which employed over 3,000 workers specialized in mechanics and precision carpentry for the production of bladed weapons and firearms. The proto-industrial facility, dating back to the second half of the 18th century, was powered by the hydraulic energy of the Sarno Canal, an engineering feat constructed by architect Domenico Fontana way back in 1599, which served to power the local mills. Today, this site of significant cultural interest, recognized as a UNESCO World Heritage Site, is the focus of an ambitious restoration and enhancement plan with a dual objective: to restore the historic and monumental portion and to repurpose the subsequent 19th-century additions. The goal is to preserve the site's industrial heritage while simultaneously recreating vital spaces for cultural, educational, and research laboratories, thus projecting the industrial past into the community's future
