    Unveiling cellular changes in leukemia cell lines after cannabidiol treatment through lipidomics

    The present study was aimed at revealing the metabolic changes that occurred in the cellular lipid pattern of acute and chronic myeloid leukaemia cells following treatment with cannabidiol (CBD). CBD treatment reduced cell viability and initiated apoptotic and necrotic processes in both cancer cell lines in a time and dose-dependent manner, showing acute myeloid leukaemia (HL-60) cells greater sensitivity than chronic myeloid leukaemia ones (K-562), without differences in the activation of caspases 3/7. Then, control and treated cells of HL-60 and K-562 cell lines were studied through an untargeted lipidomic approach. The comprehensive characterization of cellular lipids unveiled several classes significantly affected by CBD treatment. Most of the differences correspond to phospholipids, including cardiolipins (CL), phosphatidylcholines (PC) and phosphosphingolipids (SM), and also triacylglycerols (TG), being many TG species increased after CBD treatment in the acute and chronic models, whereas phospholipids were found to be decreased. The results highlight some important lipid alterations related to CBD treatment, plausibly connected with different metabolic mechanisms involved in the process of cell death by apoptosis in cancer cell lines

    Two-point correlations conditioned on the turbulent/nonturbulent interface in a turbulent temporal jet

    This paper investigates one- and two-point correlations conditioned to the turbulent/ nonturbulent interface (TNTI) of a planar turbulent temporal jet. A classical average, which is taken with respect to a spatially averaged TNTI location, is compared to two types of conditional averages, which are conditioned with respect to the instantaneous TNTI. We show that the two conditional averages differ from the classical average, especially in the TNTI region and over a distance that is comparable to the standard deviation of the instantaneous TNTI location. All correlations scale in a self-similar manner with time when normalized with a characteristic jet width calculated from integral quantities of momentum and volume fluxes. This paper presents a two-dimensional framework of separation between the correlation points and physical position from the TNTI to show interesting behavior related to the location, size, and intensity of velocity fluctuations with respect to the TNTI. We observe that the conditional velocity correlation functions exhibit distinct peaks at well-defined separations, indicating the typical scales of turbulent motions influenced by the TNTI. The values of the relevant peaks clearly demonstrate that, even when studied in a conditional framework, the dynamics of the velocity fluctuations with respect to, for example, the enstrophy, shows the presence of length scales that are very large with respect to the width of the turbulent region. Interesting results emerge when the correlation where one point is always fixed on the TNTI and the other interrogates the turbulent zone is considered. This observable investigates the turbulent dynamics inside the turbulent zone with respect to the TNTI. We show that the peak correlations are in line with the standard deviation of the TNTI, suggesting a connection to the scale of the turbulent motions attached to the TNTI. These features are not captured in classical statistics or in prior TNTI studies relying solely on one-point quantities

    SUMO2/3 conjugation of TDP-43 protects against aggregation

    Cytosolic aggregation of the RNA binding protein TDP-43 (transactive response DNA-binding protein 43) is a hallmark of amyotrophic lateral sclerosis and frontotemporal dementia. Here, we report that during oxidative stress, TDP-43 becomes SUMO2/3-ylated by the SUMO E3 ligase protein PIAS4 (protein inhibitor of activated STAT 4) and enriches in cytoplasmic stress granules (SGs). Upon pharmacological inhibition of TDP-43 SUMO2/3-ylation or PIAS4 depletion, TDP-43 enrichment in SGs is accompanied by irreversible aggregation. In cells that are unable to assemble SGs, SUMO2/3-ylation of TDP-43 is strongly impaired, supporting the notion that SGs are compartments that promote TDP-43 SUMO2/3-ylation during oxidative stress. Binding of TDP-43 to UG-rich RNA antagonizes PIAS4-mediated SUMO2/3-ylation, while RNA dissociation promotes TDP-43 SUMO2/3-ylation. We conclude that SUMO2/3 protein conjugation is a cellular mechanism to stabilize cytosolic RNA-free TDP-43 against aggregation

    Integrated assortment, shelf space and inventory decisions in retail considering product deterioration with elasticity and positioning effects

    This paper investigates a problem faced by retailers when selling multiple products and specifically the decisions related to product assortment, shelf-space planning, inventory replenishment, and storage capacity allocation. Products deteriorate while in stock and are jointly replenished. Additionally, their demand is influenced by elasticity and positioning effects. The retailer’s objective is to maximise the profit by determining the optimal ratio between the surfaces assigned to the backroom facility and the display area, the optimal product assortment, the optimal space assignment and allocation, and the optimal coordinated inventory replenishment policy. A hybrid solution procedure, including a genetic algorithm, is proposed. Among the findings, it was observed that as the decay rate increases, it is preferable to reduce the surface dedicated to the backroom facility. In fact, to mitigate the negative impact of deterioration on profit, it appears desirable to reduce the order quantity and expand the product assortment

    Estradiol and Micronized Progesterone: A Narrative Review About Their Use as Hormone Replacement Therapy

    Hormone replacement therapy (HRT) currently represents the first-line treatment to manage and reduce menopausal symptoms. Standard regimens generally combine 17β-estradiol (E2) or conjugated equine estrogens (CEEs) with micronized progesterone (P4) or synthetic progestins. While synthetic progestins ensure endometrial protection against estrogen-induced stimulation of the endometrium, their impact on metabolic, cardiovascular, skeletal, and cognitive systems is heterogeneous and not always beneficial. In contrast, progesterone, as a micronized preparation (P4), allows for more physiological effects because it is chemically identical to endogenous progesterone. This narrative review provides an updated overview of the clinical benefits of HRT regimens based on E2/P4, with a focus on their impact on endometrial thickness, venous thromboembolism (VTE), cardiovascular diseases (CVDs), breast cancer risk, cognitive effects, bone protection, and quality of life (QoL)

    Muovendo dall'analisi della resa in italiano del titolo del dramma di Miller, il saggio riflette sul ruolo del critico della traduzione come osservatore di passaggi fra lingue e culture. Dopo aver ricostruito la genesi del dramma e la sua ambientazione nella comunità italoamericana di Red Hook, il saggio si sofferma sull'analisi della lingua dei personaggi passando poi in rassegna alcune traduzioni europee, spesso orientate a normalizzare l'oralità dell'originale. Ci si concentra poi sul caso italiano: dalla versione di Guerrieri per Visconti, alla recente versione di D'Amico e alla sua messa in scena da parte del regista attore Popolizio, riflettendo infine sulla traduzione come incontro incontro/scontro di poetiche
