Ruthenium(ii)-bisdemethoxycurcumin conjugate complexes as potent antitumour agents through simultaneous inhibition of 20S proteasome and HMG-CoA reductase

    No full text
The synthesis of new conjugated bisdemethoxycurcumin ligands, each incorporating a distinct acyl substituent-cyclic, aliphatic, heteroaromatic, or branched-in place of the native hydroxyl groups, has been reported. These ligands were employed to prepare the corresponding Ru(ii)-p-cymene complexes, which were comprehensively characterized by FT-IR, NMR spectroscopy, elemental analysis, and ESI-MS. Single-crystal X-ray diffraction was used to elucidate the solid-state structures of two ligands and two metal complexes. Density Functional Theory (DFT) calculations provided further insights into the structural and electronic features of both the free ligands and their complexes. The anticancer potential of the Ru(ii)-cymene compounds was assessed in vitro against a panel of human cancer cell lines (HepG2, Caco-2, and MCF-7), as well as non-tumorigenic controls. The complexes exhibited selective, cell-type-specific cytotoxicity, primarily mediated through proteotoxic stress-evidenced by proteasome inhibition and p62 accumulation-and HMG-CoA reductase-dependent downregulation of PCNA expression

Nutritional characterization of a fermented beverage from donkey milk

The present work is part of the Tool4MEDLife project (PRIMA program), which aims to in-crease awareness about the Mediterranean diet thanks to the production of innovative foods and educational toolkits. Donkey milk, due to its potential hypoallergenic properties, has been sug-gested as a good alternative for feeding young children affected by cow milk protein allergy (CMPA) when it is not possible to breastfeed. Furthermore, thanks to the presence of nutraceu-tical molecules with antioxidant, anti inflammatory, antimicrobial, antiviral, and antitumor activities, donkey milk is interesting also for elderly people. Considering the high quality char-acteristics associated with fermented milk, the aim of this work was the production of ferment-ed donkey milk by selected Lactic Acid Bacteria (LAB) able to synthesize folates (vitamin B9). The final product was also fortified with vitamin D3 and calcium. The levels of folates of natu-ral origin represent an optimal alternative to synthetic folate-fortified products, which have limited bioavailability, and possibly lead to the accumulation of unmetabolized folic acid. Five fresh pasteurized donkey milk samples, added with vitamin D3, calcium carbonate, Streptococ-cus thermophilus, Lactobacillus delbrueckii ssp. bulgaricus and Lactobacillus plantarum IMC 509 (Synbiotec), were fermented at a constant temperature of 38-40°C for 5 hours. Folate con-tent on the final product was determined by RP HPLC (Agilent 1260 Infinity LC system), using a column C18 HiQSil HS, 5 μm, 4.6 mm i.d., 250 mm (Kya Tech Corporation, Tokyo, Japan). The fermentation process led to the production of a fermented beverage with a final pH value of 4.4. The determination of folates by RP HPLC indicated mainly the presence of tetrahydro-folate (5.3 ± 0.081 M) which is a form of folate with high bioavailability. Other water soluble vitamins found in the final fermented donkey milk were vitamin C (110.16 ± 13.5 M), nicotin-ic acid, and nicotinamide (52.7 ± 0.05 M and 7.36 ± 0.15 M respectively), and also vitamin B12 (1.69 ± 0.022 M). Comparing the above results with the water soluble vitamin content of donkey milk present in the literature, it can be noticed that the vitamin content found in the fermented donkey milk beverage is considerably higher. These preliminary results indicated that the LABs used in these experiments are very promising to produce fermented donkey milk with enhanced nutraceutical properties. Inflammatory pain is a worldwide health problem, affecting primarily women. Nonetheless, most research conducted in this field has involved male subjects, and only 10% of the results have been obtained using females. Gold Lotion (GL) is a formulated product made from the peels of six citrus fruits that have been proven, via experimental data, to possess several biological properties, such as anti-inflammatory, anticancer, and antibacterial activities. This research aims to investigate the response to GL administration in two models of peripheral inflammation to assess potential sex differences. Carrageenan was used to induce mechanical allodynia and maximal edema within hours, serving as a model of acute inflammation, while Complete Freund’s Adjuvant (CFA) was used to obtain similar results within days, resulting in a model of chronic inflammation. Acute GL administration reduced mechanical allodynia in both models and both sexes, with no anti-inflammatory effects observed. In conclusion, this study demonstrates the potential of GL to alleviate inflammatory pain in both males and females, thereby broadening its therapeutic applications

LEGGERE L’ARCHITETTURA, SCRIVERE LA STORIA. Intervista a GIORGIO CIUCCI

Commento all'art.

Retrospective analysis of compliance with Reg. EC 1/2005: examination of Trade Control and Expert System for the transport of horses to and from Southern Italy

    No full text
This study retrospectively analysed horse movements recorded in the European Trade Control and Expert System (TRACES) arriving in, departing from and crossing Apulia, during 2022–2023. Data from 290 TRACES were analysed to identify the most common inaccuracies and regulatory non-compliances, as well as the difference in journey conditions between horses that did and did not arrive at a slaughterhouse. The dataset included 3254 horses recorded in the 290 TRACES, of which 2946 were for slaughter. Descriptive statistics, chi-square, Kruskal-Wallis, or Mann-Whitney tests were performed. The most common routes for slaughter were from Poland and France to Apulia, while for other destinations were from Italy to Germany. Horses transported for slaughter were moved in significantly larger groups (mean 22 vs. 2 horses) and given less space per animal (1.57 vs. 3.25 m2) compared to other destinations (p < 0.001). Regardless of destinations and type of horses, namely registered and non-registered, the analysis revealed that journey durations, conditions, inaccuracies, and regulatory non-compliances were similar. Furthermore, 75 TRACES incorrectly classified horses as Registered when they were shipped for slaughter, which impedes verification of welfare standards. Many of the inaccuracies and non-compliances documented were associated with a specific transporter (p < 0.001). Our findings emphasise that TRACES needs improvements: many data fields were incorrectly completed, likely due to its complexity and vague policies. Shorter journeys and more space should be enforced to protect horse welfare during transport. Additionally, the TRACES’ portal for horses should ensure equal welfare standards for all transported horses, regardless of registration or destination

    No full text
AIM: To define the role, competencies and clinical focus of specialised breast cancer nurses and to explore post-basic academic training programmes available in Western countries. DESIGN: A scoping review was reported following the PRISMA-ScR framework and aligned with JBI methodology. METHODS: The research question was formulated using the Population, Concept and Context model. Searches were performed across four databases (Medline, Embase, Cochrane Library, CINAHL and Gray Literature) between July and September 2024. Article screening and data extraction were independently performed by two authors. Descriptive analysis was employed to interpret the data. RESULTS: From an initial 1499 records, 143 studies were selected, with 16 included in the final analysis. The studies, published between 2001 and 2024, evaluated the role of breast cancer nurses in oncology, focusing on their impact on patient care, health outcomes, quality of life, psychological support and breast cancer nurses-led interventions. Two studies assessed educational programmes, identifying a diverse range of postgraduate degrees and specialised training options tailored to meet the specific needs of this essential nursing role. CONCLUSIONS: Breast cancer nurses are crucial in delivering comprehensive care and improving QoL for breast cancer patients. They contribute to enhancing clinical outcomes through patient education and support. Ongoing professional development and expanded education are essential to meet evolving patient needs. IMPLICATIONS FOR THE PROFESSION: Breast cancer nurses play a pivotal role in improving patient outcomes by delivering education, psychological support and holistic care. To keep pace with the increasing complexity of breast cancer management, it is essential to prioritise expanded academic training and continuous professional development. These efforts will ensure BCNs are well prepared to address evolving patient needs and provide high-quality, comprehensive care. PATIENT OR PUBLIC CONTRIBUTION: No patient or public contribution

Giancarlo De Carlo. Il Campus universitario di Urbino

    No full text
