    Increasing the fraction of correct solutions in ensembles of protein-protein docking models by an iterative consensus algorithm

    : Protein-protein interactions play a pivotal role in numerous biological processes, making their structural prediction essential for molecular biology and medicine. Computational docking methods generate ensembles of models for protein complexes, but identifying correct solutions within these ensembles remains a challenge. This study introduces Iter-CONSRANK, an iterative consensus-based scoring algorithm designed to enrich the fraction of correct models in such ensembles. Building on the established CONSRANK algorithm, Iter-CONSRANK incorporates iterative filtering to progressively discard lower-ranked models based on inter-residue contact conservation, thus refining the ensemble. Performance evaluation using the 3K-BM5up dataset (~1.6 × 105 models), 30K-BM5 dataset (~6.4 × 106 models), and the CAPRI-derived Score_set (~2.0 × 104 models) demonstrated significant improvement in the proportion of correct solutions across model difficulty categories. Iter-CONSRANK effectively identified correct models within the top-ranking positions, outperforming over 150 other scoring functions. For moderately challenging targets, the algorithm enriched correct solutions by up to eightfold, making subsequent analyses more straightforward. The software is publicly available, enabling its application for pre-processing docking ensembles or independent scoring. Iter-CONSRANK is a promising tool for advancing the accuracy of protein-protein docking model evaluations

    Bi-Objective Portfolio Optimization Under ESG Volatility via a MOPSO-Deep Learning Algorithm

    In this paper, we tackle a bi-objective optimization problem in which we aim to maximize the portfolio diversification and, at the same time, minimize the portfolio volatility, where the ESG (Environmental, Social, and Governance) information is incorporated. More specifically, we extend the standard portfolio volatility framework based on the financial aspects to a new paradigm where the sustainable credits are taken into account. In the portfolio construction, we consider the classical constraints concerning budget and box requirements. To deal with these new asset allocation models, in this paper we develop an improved Multi-Objective Particle Swarm Optimizer (MOPSO) embedded with ad hoc repair and projection operators to satisfy the constraints. Moreover, we implement a deep learning architecture to improve the quality of estimating the portfolio diversification objective. Finally, we conduct empirical tests on datasets from three different countries’ markets to illustrate the effectiveness of the proposed strategies accounting for various levels of ESG volatility

    L’arduo compito del regolamento MiCAR (Markets in CryptoAssets Regulation) di introdurre una disciplina dettagliata e completa del mercato delle cripto-attività non ha sortito tutti gli effetti sperati. L’incertezza dell’àmbito applicativo, i problemi di coordinamento tra il MiCAR e le altre normative “correlate” nonché le criticità della “transizione” dei prestatori di servizi di cripto-attività, dal vecchio al nuovo regime normativo europeo, vengono analizzati alla luce dei più recenti interventi “regolamentari” e di soft law, che non sempre risultano risolutivi. I profili di “incompletezza” che ancora sussistono richiederanno, dunque, un nuovo intervento del legislatore europeo anche al fine di evitare disallineamenti e frammentazioni nelle legislazioni nazionali dei vari Stati membri.The MiCAR Regulation has raised interpretative issues regarding its scope of application and its coordination with related legislation from the outset. These interpretative problems are examined in light of recent regulatory and soft law developments. It is considered that the existing gaps will require intervention by the European legislator in order to avoid fragmentation among the Member States’ legal systems

    Experimental and Numerical Study on the Performance and Exhaust Emissions of a Single-Cylinder Natural Gas Large Bore Engine Equipped with Passive Pre-Chamber

    The maritime industry is among the most energy-intensive sectors, and achieving fleet decarbonization is crucial to significantly reduce greenhouse gas emissions. As a transitional fuel, natural gas (NG) presents a viable short-to-midterm solution. Compared to conventional marine fuels, NG has the potential to lower carbon dioxide emissions by approximately 20-30%. However, to fully leverage this potential on carbon footprint reduction, substantial advancements in combustion technologies are required. One promising approach to enhance the efficiency of SI NG engines is the implementation of Passive Pre-Chamber (PPC) technology. This strategy enables leaner combustion, improving thermal efficiency, mitigating the occurrence of knocking, and reducing NOx emissions. This study presents both experimental and numerical investigations to analyze the impact of charge dilution and ignition timing on the performance and emissions of a single-cylinder prototype NG PPC SI engine for marine application, retrofitted from baseline diesel architecture. Numerical simulations combining 1D and 3D CFD approaches were used to guide the combustion system and engine component design, optimizing valve timing, compression ratio, and fuel injection parameters to mitigate knock and improve thermal efficiency. Based on numerical simulations, excess air effects on thermodynamic efficiency and flame speed were evaluated. The experimental tests were conducted at 1500 rpm constant engine speed under different load conditions. Variation of the air-to-fuel ratio and spark advance were performed to characterize their effects on engine operation. The results were utilized to validate a 1D model, which demonstrated a high level of accuracy in reproducing the combustion evolution. PPC technology enabled high charge dilution (λ≈1.7). The coefficient of variation of IMEP remained below 1.5% throughout a wide range of λ values and combustion phasing conditions, indicating stable combustion. The engine exhibited indicated efficiencies of over 45%, marginally exceeding the predictions obtained from numerical simulations. This research underscores the potential of PPC technology in enhancing the efficiency and sustainability of NG-fueled marine engines, offering valuable insights to optimize combustion strategies for future low-emission propulsion systems
