    Pembentukan Kokristal Asam Usnat – N-Methyl-DGlucamine dengan Metode Penguapan Pelarut dan Pengaruhnya terhadap Penurunan Interleukin-8 pada Tikus Inflamasi

    The objective of the research was to prepare the cocrystal of usnic acid -N-methyl-D-glucamine in order to increase solubility, dissolution rate, and decrease IL-8 levels in rats with inflammation. Cocrystal was prepared with a ratio of 1:1 equimolar by a solvent evaporation method using etanol. Characterizations were conducted using Powder X-ray diffraction (PXRD), Differential Scanning Calorimetry (DSC), infrared spectroscopy (FT-IR), and Scanning Electron Microscopy (SEM). The solubility test, dissolution rate test, and determination of interleukin-8 (IL-8) levels in rats were also performed. CO2 free distilled water was used as a media in the solubility test. Meanwhile, CO2 free distilled water with 0.25 % w/v Sodium Lauryl Sulphate (SLS) were used as the media for dissolution rate studies. Both tests were conducted using a UV-Vis spectrophotometer. The determination of IL-8 levels in rat was performed by using an ELISA method. Powder X-ray Diffraction (PXRD) diffractogram showed a new peak compared to diffraction peaks of each component and physical mixture that confirmed the formation of cocrystal, and peak intensity decreased that shown formation of eutectic mixture. The appearance of the DSC thermogram from cocrystal showed different endothermic peaks for both constituent components. The FT-IR spectrum of cocrystal showed a shift of the wave number spectrum compared to pure usnic acid and N-methyl-D-glucamine. Microscopic analysis by SEM showed a change of crystal morphology. The solubility and dissolution rate of cocrystal increased significantly (p<0.05) by 24 and 23 times compared to the pure usnic acid. Cocrystal could reduce IL-8 levels in rats with inflammation equivalent to acetosal (P <0.05). This study concludes, that cocrystal of usnic acid–Nmethyl-D-glucamine can enhance solubility and dissolution rate, and decrease IL-8 levels in rats with inflammatio

    Fabrikasi dan Studi Stabilitas Self-Nano Emulsifying Propolis menggunakan Minyak Kesturi sebagai Pembawa

    Propolis is a poorly-water soluble resin produced by bees with antibacterial, antioxidant and anti-inflammatory activities. Self-nano emulsifying (SNE) is one of the drug delivery methods to improve drug solubility of poorly water-soluble. This study aimed to prepare a formulation of transparent and stable of propolis loading SNE with castor oil, cremophor RH40, and PEG 400 as the carriers. The preparation of SNE was conducted by mixing propolis with the vehicle. The spontaneous nanoemulsion area was constructed by the ternary phase diagram. The characterization includes particle size, zeta potential, transmittance, thermodynamic stability, robustness to dilution, and accelerated stability test. The nanoemulsion area was found in Formulae (F) 1 to F9 with a range composition of castor oil (10-30%), cremophor RH 40 (40-80%), and PEG 400 (10-40%). The nine formulas showed transmittance values at 96-99%, particle size 10-40 nm zeta potential less than -40 mV and stable during thermodynamic stability test. The accelerated stability test showed that F5 (20: 70: 10) produces the optimal SNE formula with no phase separation and precipitation.     Diversifikasi komoditas pangan lokal serta ekstentifikasi dengan pemanfaatan lahan sub optimal dapat dijadikan sebagai solusi menjaga ketersediaan dan keamanan pangan di tengah pandemi COVID-19. Hanjeli merupakan komoditas pangan lokal yang berpotensi untuk dikembangkan pada lahan sub optimal, hal ini dikarenakan hanjeli mudah dibudidayakan dan memiliki kemampuan adaptasi yang tinggi diberbagai macam kondisi lingkungan, serta mengandung nutrisi yang cukup tinggi. Hanjeli cocok untuk dibudidayakan di Sumatera Barat karena kondisi geografis dan iklim yang sesuai dengan syarat tumbuh bagi hanjeli, namun pemanfaatan komoditi ini masih sangat minim di Indonesia, karena keterbatasan pengetahuan masyarakat yang menganggap bahwa hanjeli ialah hanya tanaman liar ataupun gulma. Untuk itu pengembangan hanjeli dengan teknologi yang tepat diharapkan mampu mendukung program pemerintah dalam menjaga stabilitas ketahanan pangan nasional di tengah pandemi COVID-19. Penelitian ini bertujuan untuk: (1) mengetahui pengaruh berbagai tingkat kepadatan populasi terhadap pertumbuhan dan hasil hanjeli pada lahan sub optimal; (2) mengetahui populasi optimum untuk meningkatkan pertumbuhan dan hasil budidaya hanjeli pada lahan sub optimal. Penelitian ini akan dilaksanakan pada bulan Juli hingga November 2020 di Kota Padang, Provinsi Sumatera Barat. Rancangan percobaan yang digunakan pada penelitian ini adalah Rancangan Acak Kelompok (RAK) dengan 3 kelompok ulangan serta terdiri dari 8 perlakuan tingkat kepadatan populasi. Kepadatan populasi yang digunakan dalam penelitian ini ialah A1 = Populasi 62.500 tanaman/ha (jarak tanam 40 x 40 cm) , A2 = Populasi 50.000 tanaman/ha ( jarak tanam 50 x 40 cm ), A3 = Populasi 40.000 tanaman/ha ( jarak tanam 50 x 50 cm ), A4 = Populasi 33.333 tanaman/ha ( jarak tanam 50 x 60 cm ), A5 = Populasi 28.571 tanaman/ha ( jarak tanam 50 x 70 cm ), A6 = Populasi 25.000 tanaman/ha ( jarak tanam 50 x 80 cm ), A7 = Populasi 22.222 tanaman/ha ( jarak tanam 50 x 90 cm ), dan A8 = Populasi 20.000 tanaman/ha ( jarak tanam 50 x 100 cm ). Data pengamatan dianalisis secara statistik untuk mengetahui perlakuan yang berpengaruh dengan menggunakan uji F pada taraf 5% dan data yang berbeda nyata akan dilakukan uji lanjut menggunakan uji DNMRT pada taraf 5%. Rencana luaran penelitian di tahun ke-1 ini adalah jurnal nasional terakreditasi dan draft buku. Hasil penelitian menujukkan bahwa jarak tanam 50 x 70 cm, 50 x 80 cm, 50 x 90 cm dan 50 x 100 cm memberikan pengaruh terbaik pada variabel tinggi tanaman, diameter batang, jumlah anakan serta jumlah daun.
