    “Her energy becomes our energy” Examining the self-efficacy beliefs of beginner learners of Japanese learning with native and non-native instructors

    Japanese as an additional language, while not the most common choice for language learners, enjoys significant popularity around the world thanks in part to media such as manga and anime becoming cultural mainstays even outside of their home country and Japan’s dominance as an economic heavyweight. However, the Japanese language is routinely touted as one of the hardest to learn due to its dissimilarity to popular Germanic and Romance languages such as English and French in areas such as its multiple independent writing systems, complex grammar and differing sentence structure (subject-object-verb). As a result, comprehending the elements that best cultivate learners’ self-efficacy during students’ initial months engaging in Japanese language study is crucial for those wishing to encourage student interest and continuation in the language. While one such element was hypothesized to be teacher cultural and linguistic identity, other equally relevant but unexpected factors came to light through the course of this research.In the current study, ten students from two different classes, five instructed by a native Japanese teacher and five instructed by a non-native Japanese teacher, were interviewed to capture their perceptions, preferences, expectations and reactions to the first four months of their Japanese learning experience. Their classes as a whole were also observed four times each to uncover further details regarding student interactions with each other and their instructors to ameliorate individual participant reports. Findings indicate that indeed instructor cultural and linguistic identity as either a native Japanese speaker or a French/English bilingual speaker significantly affected many students’ self-efficacy and drive to continue learning Japanese. Additionally, many instructor-centered pedagogical factors such as feedback, praise, and student engagement not only intersected with but went beyond teacher cultural and linguistic identity, as did course-centered factors such as lesson length, course content, curriculum design and testing, as well as student-centered factors such as time management challenges.Le japonais en tant que langue supplémentaire, bien que n'étant pas le choix le plus courant pour les étudiants de langues, jouit d’une popularité considérable dans le monde entier, en partie grâce aux médias tels que les mangas et les dessins animés qui deviennent des piliers culturels même en dehors de leur pays d’origine et à la domination du Japon en tant que puissance économique. Cependant, le japonais est couramment présenté comme l’une des langues les plus difficiles à apprendre en raison de ses différences face aux langues germaniques populaires telles que l’anglais et le français. Ses différences se retrouvent dans des domaines tels que ses multiples systèmes d’écriture indépendants, sa grammaire complexe et sa structure de phrase différente. En conséquence, il est essentiel de comprendre les éléments qui cultivent le mieux l’auto-efficacité et la motivation des étudiants au cours des premiers mois de leur séjour, afin de les encourager à poursuivre leur apprentissage de la langue. Alors que l’on supposait que l’un de ces éléments était l’identité culturelle et linguistique de l’enseignant, d’autres facteurs tout aussi pertinents mais inattendus ont été mis au jour tout au long de cette recherche.Dans la présente étude, dix étudiants de deux classes différentes, cinq dirigés par une enseignante japonais et cinq par une enseignante non-japonais, ont été interrogés pour saisir leurs perceptions, préférences, attentes et réactions au cours des quatre premiers mois de leur apprentissage du japonais. Leurs cours ont également été observés en intégralité quatre fois chacun pour révéler de plus amples détails concernant les interactions des élèves entre eux et avec leurs instructeurs. Ceci a été fait pour améliorer les rapports individuels des participants.Les résultats indiquent en effet que l'identité culturelle et linguistique de l'instructeur en tant que locuteur natif japonais ou bilingue anglais / français affectait de manière significative l'efficacité personnelle et l’enthousiasme pour continuer à apprendre le japonais de nombreux étudiants. En outre, de nombreux facteurs pédagogiques centrés sur l'instructeur, tels que les rétroactions, les éloges et l'engagement des étudiants, ont non seulement coïncidé avec l'identité culturelle et linguistique des enseignants, mais les ont dépassés, de même que des facteurs centrés sur le cours, tels que la durée des leçons, le contenu du cours, la conception du curriculum et les examens, ainsi que des facteurs centrés sur l'élève, tels que les défis liés à la gestion du temps

    On the prediction of mRNA subcellular localization with machine learning

    Cells are the basic units of life, and yet they are regulated by many delicate and to some extent, fragile, subcellular processes that are crucial to their survival. A simple genetic mutation could possibly clog up some important regulatory processes, or perturb the function of the product it encodes, which might ultimately bring the demise of the entire system. Therefore, it is important to gain more insights into the many control processes of cell and the regulatory factors associated with them, one prominent example of which would be the mechanism related to the RNA subcellular localization that we would focus on almost exclusively in this study from a computational perspective.RNA subcellular localization mechanism is one of the most important, yet under-appreciated, facets of the broader gene regulatory process, which helps with the cellular organization and regulation on gene expression, via transporting the RNA transcripts to their designated locations where their function, structure or translated proteins are needed. It is generally accepted as a fact that RNA trafficking mechanism is mediated between the trans-regulatory factors such as the RNA binding proteins, and the cis-acting elements — short snippets of the transcript that contain the RBP binding sites — which we call zipcode as they are considered to contain information on its address of delivery.The release of new RNA subcellular localization dataset has enabled us to build the first computational tool using state-of-the-art deep learning techniques, to predict the localization outcome for the protein-coding RNA from mere transcript sequence, and subsequently to identify the zipcode elements thereof. Our proposed method has achieved good accuracy compared to the baseline methods based on the k-mers features, despite the intrinsic difficulty that arise from the complex and stochastic interactions during trafficking events, as well as the limitations imposed by the available dataset.Les cellules sont les unités de base de la vie, et pourtant, elles sont régies par de nombreux processus subcellulaires délicats et, dans une certaine mesure, fragiles, qui sont essentiels à leur survie. Une simple mutation génétique pourrait éventuellement obstruer des processus de régulation importants ou perturber le fonctionnement du produit qu’elle encode, ce qui pourrait éventuellement entraîner la disparition de l’ensemble du système. Par conséquent, il est important de mieux comprendre les nombreux processus de contrôle de la cellule et les facteurs de régulation qui leur sont associés, un exemple frappant étant le mécanisme lié à la localisation sous-cellulaire de l'ARN sur lequel nous nous concentrerons presque exclusivement dans cette étude à partir de une perspective de calcul.Le mécanisme de localisation sous-cellulaire de l'ARN est l'une des facettes les plus importantes, mais sous-estimée, du processus plus général de régulation des gènes, qui facilite l'organisation cellulaire et la régulation de l'expression des gènes, via le transport des transcrits d'ARN vers leurs emplacements désignés, où leur fonction, leur structure ou des protéines traduites sont nécessaires. Il est généralement admis que le mécanisme du trafic d'ARN est régulé par des facteurs de régulation trans, tels que les protéines de liaison à l'ARN, et les éléments agissant en cis, des extraits courts du transcrit qui contiennent les sites de liaison à la RBP, qui nous appelons zipcode car ils sont censés contenir des informations sur son adresse de livraison.La publication d'un nouvel ensemble de données de localisation subcellulaire des ARN nous a permis de créer le premier outil de calcul utilisant des techniques d’apprentissage approfondi de pointe, afin de prédire le résultat de la localisation de l’ARN codant pour une protéine à partir d’une simple séquence de transcription, puis d’identifier le éléments de zipcode de ceux-ci. Notre méthode proposée a obtenu une bonne précision par rapport aux méthodes de base basées sur les caractéristiques de k-mers, malgré la difficulté intrinsèque résultant des interactions complexes et stochastiques lors d'événements de gestion de trafic, ainsi que des limitations imposées par l'ensemble de données disponibles

    Testing homogeneity in sparse Gaussian linear regression mixture models

    We present a method of determining homogeneity in Gaussian mixture models. A thorough review of the literature surrounding the establishment of the number of subpopulations in mixture models is presented. It is of interest to use a hypothesis testing procedure to arrive to a conclusion. We focus on the event that the response of a linear regression follows a Gaussian mixture of distributions with a large number of covariates. A modified EM algorithm estimates the unknown parameters while accounting for the difficult properties associated with Gaussian mixture models. Regularization methods, specifically the lasso and adaptive lasso, are introduced to simultaneously perform variable selection and determine the value of the statistic employed in the testing procedure. Cross-validation is employed over the entire modified EM algorithm in order to select the optimal value of the tuning parameter. Finally, the power and Type I error rates of the procedure are computed and discussed.Nous présentons une méthode qui sert à détecter l'homogénéité dans des modèles de mélanges de gaussiennes. Une révision complète de la litérature concernant l'établissement du nombres de sous-populations dans des modèles de mélanges est présentée. Nous sommes particulièrement intéressés à l'application de tests d'hypothèses afin d'atteindre une conclusion. L'accent est mis sur le cas de l'analyse de régression linéaire où la variable réponse suit un mélange de distributions gaussiennes avec plusieurs covariables. Les paramètres inconnus sont estimés à l'aide d'un algorithme EM modifié qui prend en compte les difficultés associées avec les modèles de mélanges gaussiennes. Des méthodes de régularisation, notamment le lasso et le lasso adaptif, sont introduits afin de choisir les variables et calculer la valeur de la statistique du test en même temps. La validation croisée de l'alogrithme permet de choisir le paramètre d'ajustement idéal. Finalement, l'efficacité statistique et l'erreur de type I de la démarche sont calculés et présentés

    Coming to life: the illustrated novel and Theodor von Holst's creation of the 1831 "Frankenstein" frontispiece

    Henry Colburn and Richard Bentley published Mary Wollstonecraft Shelley’s Frankenstein in 1831 with the additions of an authorial introduction, revisions, and Theodor von Holst’s frontispiece image. Holst’s illustration was the first-ever published image of Frankenstein and his Creature and engages with a number of diverse elements from the novel’s publishing, public, and scientific histories. This thesis acts to place Holst’s image back into a very specific time through the examination of Frankenstein’s publications, author, audiences, theatrical precedents, and notions of Romantic melancholy and Gothic wonder. Additionally, I examine how practices in academic art, traditions of anatomy, medical portraiture, and an alchemical revival reflect the novel’s past and respond to its present 1831 socio-political conditions. By utilizing the medium of the book itself as a handled, illustrated novel, Frankenstein’s 1831 edition comes to life as a cultural “Frankenstein’s monster.”En 1831, Henry Colburn et Richard Bentley ont publié une nouvelle édition révisée de Frankenstein de Mary Wollstonecraft Shelley comprenant une nouvelle introduction de l’auteure et l’image frontispice de Theodor von Holst. L’illustration de Holst constitue la première représentation visuelle publiée de Frankenstein et de sa Créature. Elle évoque de nombreux éléments découlant de l’histoire du roman quant à sa publication, au public et à la science. Cette thèse détermine comment l’illustration de Holst représente une époque particulière par l’examen des publications de Frankenstein, de son auteure, de son public, de ses précédents théâtraux et des notions de mélancolie romantique et d’émerveillement gothique. Elle explore la relation entre les pratiques dans l’art académique, les traditions en anatomie, en portrait médical et le renouveau de l’alchimie qui puisent dans le passé de ce roman et qui reflètent les conditions socio-politiques contemporaines de 1831. En utilisant le livre, objet qui peut être manipulé comme média pour ce roman illustré, l’édition de 1831 de Frankenstein donne vie au « Monstre de Frankenstein » et à sa culture

    Computer-assisted proofs for a nonlinear Laplace-Beltrami equation on the sphere

    In this study, we prove the existence and local uniqueness of radially symmetric solutions of nonlinear partial differential equations via a rigorous numerical method. We introduce the concepts of Banach spaces of geometrically decaying sequences and explore the domains of convergence of the Taylor and Chebyshev expansions. These notions constitute the basis for the Radii Polynomial Theorem on Banach spaces of infinite sequences, which is the main tool employed to obtain the proofs by using a combination of Taylor and Chebyshev coefficients of the solutions. We also introduce briefly the notions of interval analysis to justify the rigor of our computer-assisted results.Dans cette étude, nous prouvons l'existence et l'unicité locale de solutions d'ÉDPs non linéaires et symétriques via une méthode numérique. Nous introduisons des concepts sur les espaces de Banach de suites à décroissance géométrique et explorons les domaines de convergence des séries de Taylor et de Chebyshev. Ces notions constituent les bases du Théorème du Polynôme de Rayons sur les espaces de Banach de suites infinies, qui sera l'outil principal pour obtenir les preuves en utilisant une combinaison de coefficients de Taylor et de Chebyshev. Nous introduisons également des notions de base sur l'analyse d'intervalle pour justifier la rigueur des résultats obtenus numériquement

    FOXA1: Its role and its interaction with prolactin hormone signaling in triple negative breast cancer tumorigenesis

    Triple Negative Breast Cancer (TNBC) is the most resistant subtype of breast cancer. The lack of expression of estrogen, progesterone, and human epidermal growth factor 2 receptors in TNBC leads to the limited use of targeted therapy. Therefore, treatment with chemotherapy remains the main approach for treating TNBC with low therapeutic benefits and increased risk of resistance. Pro-differentiation based therapies by reversing breast cancer cells into an epithelial phenotype have recently been studied as a promising therapeutic approach for TNBC. Indeed, the laboratory of Dr. Ali showed that restoring prolactin pro-differentiation pathway reversed the mesenchymal aggressive phenotype of TNBC into the epithelial form with less aggressive behavior, less invasiveness, and lower metastasis. In addition, they identified a TNBC subgroup of patients that preserved prolactin receptor (PRLR) expression to show better overall survival and better prognosis. By metagenes cluster analysis, the TNBC-PRLR subtype is characterized by the up regulation of luminal-like differentiation genes which are Forkhead Box A1 (FOXA1) and androgen receptor (AR). Our aim is to determine the effect of FOXA1 on the epithelial mesenchymal transition (EMT) of breast cancer cells and how it may modulate the antitumorigenic role of prolactin and its signaling pathway in breast cancer. To achieve our objectives, we measured the expression of FOXA1 at the mRNA and protein levels after treating TNBC-PRLR cell lines with prolactin. We also produced CRISPR knock out of FOXA1 in these cell lines and measured the protein level of both E-cadherin, an epithelial marker and vimentin as a mesenchymal marker. As expected, the knock-out cells showed a decrease in the level of E-cadherin and an increase in vimentin compared to the parental cells. Finally, we conclude that FOXA1 can be a potential pro-differentiation marker in breast cancer cells. Further studies are needed to investigate this role of FOXA1 and its use as a target for personalized medicine in triple negative breast cancer.Le cancer du sein triple négatif (TNBC) est le sous-type de cancer du sein le plus résistant. Le manque d'expression des récepteurs des œstrogènes, de la progestérone et du facteur 2 de croissance épidermique humain dans le TNBC conduit à une utilisation limitée du traitement ciblé. Par conséquent, le traitement par chimiothérapie demeure la principale approche pour le traitement de la TNBC avec de faibles avantages thérapeutiques et un risque accru de résistance. Les thérapies fondées sur la différenciation par inversion des cellules cancéreuses du sein en un phénotype épithélial ont récemment été étudiées comme une approche thérapeutique prometteuse pour le TNBC. En effet, le laboratoire du Dr Ali a montré que le rétablissement de la voie de différenciation de la prolactine a inversé le phénotype mésenchymateux agressif de la TNBC dans la forme épithéliale avec un comportement moins agressif, moins invasif et moins de métastase. De plus, ils ont identifié un sous-groupe de patients TNBC dont l'expression du récepteur de la prolactine (PRLR) est élevée, la survie globale meilleure et le pronostic meilleur. Par l'analyse des groupes de métagènes, le sous-type TNBC-PRLR est caractérisé par la régulation ascendante des gènes de différenciation de type luminal qui sont FOXA1 et AR. Notre objectif est de déterminer l'effet de FOXA1 sur la transition mésenchymateuse épithéliale (EMT) des cellules cancéreuses du sein et comment il peut moduler le rôle antitumorigène de la prolactine et sa voie de signalisation dans le cancer du sein. Pour atteindre nos objectifs, nous avons mesuré l'expression de FOXA1 au niveau de l'ARNm et des protéines après avoir traité les lignées cellulaires TNBC-PRLR avec la prolactine. Nous avons également produit CRISPR knock out de FOXA1 dans ces lignées cellulaires et mesuré le taux de protéines de la E-cadhérine, un marqueur épithélial et de la vimentine comme marqueur mésenchymateux. Comme on s'y attendait, les cellules éliminées ont montré une diminution du taux de E-cadhérine et une augmentation de la vimentine par rapport aux cellules parentales. Enfin, nous concluons que FOXA1 peut être un marqueur pro-différenciation potentiel dans les cellules du cancer du sein. D'autres études sont nécessaires pour étudier ce rôle de FOXA1 et son utilisation comme cible de la médecine personnalisée dans le cancer du sein triple négatif

    Removing user fees for facility-based delivery services: a difference-in-differences evaluation from ten sub-Saharan African countries.

    BACKGROUND: Several countries in sub-Saharan Africa have recently adopted policies that remove user fees for facility-based delivery services. There is little rigorous evidence of the impact of these policies on utilization of delivery services and no evaluations have examined effects on neonatal mortality rates (NMR). In this article, we estimate the causal effect of removing user fees on the proportion of births delivered in facilities, the proportion of births delivered by Caesarean section, and NMR. METHODS:We used data from Demographic and Health Surveys conducted in 10 African countries between 1997 and 2012. Kenya, Ghana and Senegal adopted policies removing user fees for facility-based deliveries between 2003 and 2007, while seven other countries not changing user fee policies were used as controls. [...] CONCLUSIONS:Our findings suggest removing user fees increased facility-based deliveries and possibly contributed to a reduction in NMR. Evidence from this evaluation may be useful to governments weighing the potential benefits of removing user fees

    Antimicrobial hierarchically porous graphene oxide sponges for water treatment

    Water treatment technologies based on graphene oxide (GO) sponges show promise due to their high surface area and versatile chemistry, yielding an excellent adsorption affinity for different contaminants. However, the bacteria removal capacity and the intrinsic antimicrobial properties of GO sponges are not well understood. While GO has been successfully functionalized with antibiotics or metal biocides, these antimicrobials present cytotoxicity concerns. Natural antimicrobial agents such as antimicrobial enzymes, peptides, and polymers hold promise in this respect due to their relatively low cost, biocompatibility, and ability to readily functionalize GO by covalent bond formation with oxygen-containing functional groups. In this work, the antimicrobial enzyme lysozyme, antimicrobial peptide nisin, and antimicrobial polyamide ε-poly-l-lysine were used to covalently functionalize the surface of a hierarchically porous GO sponge. The antimicrobial activity of the functionalized material was demonstrated against two model organisms: the Gram-positive B. subtilis and Gram-negative E. coli. The performance of the porous material in a simulated filtration context was evaluated using packed column experiments, and an improved bacterial retention of both strains by the functionalized sponge was demonstrated. Furthermore, samples of spent sponge after filtration were evaluated with a membrane integrity assay demonstrating antimicrobial activity in a continuous flow mode

    QMC-D and Nano Tweezer Measurements to Characterize the Effect of soil cellualse on the depositon of PEG-coated TiO2 Nanoparticles in Model Subsurface Environments

    Upon release to the environment, engineered nanoparticles (ENPs) undergo several chemical, physical and biological transformations that may affect their fate, transport, and bioavailability. The impact of ENP transformations (e.g., coating with natural organic matter or heteroaggregation with natural colloids) on ENP fate and transport has been systematically examined; however, the influence of soil enzymes that are ubiquitous in soils has not been considered. In this study, we examined the effect of a model extracellular soil enzyme (cellulase) – either free in an artificial porewater or adsorbed on a model aquifer grain (silica) surface – on the deposition kinetics of polyethylene glycol-coated titanium dioxide nanoparticles (PEG-nTiO2). A quartz crystal microbalance with dissipation monitoring (QCM-D) was used to study the interaction of PEG-nTiO2 with bare and cellulase-coated silica surfaces as well as in the presence of free cellulase over a range of sodium chloride concentrations. Significant reduction in PEG-nTiO2 deposition rates was observed in the presence of cellulase indicative of strong repulsive interactions between the nanoparticles and the layer of cellulase adsorbed on the silica surface. QCM-D observations were supported by measurements of the PEG-nTiO2–surface interaction energies using an optical NanoTweezer apparatus revealing more repulsive particle–surface interaction energies for the cellulase-coated silica. QCM-D measurements also indicated formation of more viscoelastic films in the presence of cellulase compared to bare silica except at the lowest ionic strength (IS) studied (10 mM NaCl). Overall, this work shows the potential for increased mobility of ENPs in subsurface environments in the presence of extracellular soil enzymes, motivating the need for further studies on the fate and behaviour of ENPs in the presence of these ubiquitous biomolecules

    Experimental assessment of the sensitivity of an estuarine phytoplankton fall bloom to acidification and warming

    We investigated the combined effect of ocean acidification and warming on the dynamics of the phytoplankton fall bloom in the Lower St. Lawrence Estuary (LSLE), Canada. Twelve 2600 L mesocosms were set to initially cover a wide range of pHT (pH on the total proton scale) from 8.0 to 7.2 corresponding to a range of pCO2 from 440 to 2900 µatm, and two temperatures (in situ and +5 ∘C). The 13-day experiment captured the development and decline of a nanophytoplankton bloom dominated by the chain-forming diatom Skeletonema costatum. During the development phase of the bloom, increasing pCO2 influenced neither the magnitude nor the net growth rate of the nanophytoplankton bloom, whereas increasing the temperature by 5 ∘C stimulated the chlorophyll a (Chl a) growth rate and maximal particulate primary production (PP) by 76 % and 63 %, respectively. During the declining phase of the bloom, warming accelerated the loss of diatom cells, paralleled by a gradual decrease in the abundance of photosynthetic picoeukaryotes and a bloom of picocyanobacteria. Increasing pCO2 and warming did not influence the abundance of picoeukaryotes, while picocyanobacteria abundance was reduced by the increase in pCO2 when combined with warming in the latter phase of the experiment. Over the full duration of the experiment, the time-integrated net primary production was not significantly affected by the pCO2 treatments or warming. Overall, our results suggest that warming, rather than acidification, is more likely to alter phytoplankton autumnal bloom development in the LSLE in the decades to come. Future studies examining a broader gradient of temperatures should be conducted over a larger seasonal window in order to better constrain the potential effect of warming on the development of blooms in the LSLE and its impact on the fate of primary production
