    Autumn leaves and chlorophyll degradation

    秋の樹木の紅葉はクロロフィルが分解されることによってみられる現象である.秋になると気温が下がり光合成ができなくなるため,落葉樹はクロロフィルを分解する.このように,クロロフィルの分解は植物の発達段階の最後に見られる現象である.そのため,クロロフィルの分解自体には生理学的役割はないとも考えられる.しかし,クロロフィルの分解を抑制したり促進したりすると,光化学系の量が変動する.また,クロロフィルの分解は老化の進行に影響を与える.このように,クロロフィルの分解自体も細胞の機能や個体の成長に影響を与える.本稿ではクロロフィルの分解機構を概説し,その生理学的役割について論じる.The autumn color of trees is caused by the degradation of chlorophyll. In autumn, deciduous trees degrade chlorophyll as temperatures drop and photosynthesis is no longer possible. Thus, chlorophyll degradation is a process seen at the end of the plant developmental stage. Therefore, chlorophyll degradation itself may not have a physiological role. However, when chlorophyll degradation is suppressed or accelerated, the amount of photosystems changes. Chlorophyll degradation also affects the progression of senescence. Thus, chlorophyll degradation itself also affects cell function and plant growth. Here we summarize the mechanisms of chlorophyll degradation and discuss their physiological roles

    光合成生産は,糖を合成する葉の活性(ソース活性)と,根や茎などの非光合成器官,貯蔵器官,展開中の若い葉などの糖消費活性(シンク活性)のどちらか低い方に律速される.シンク・ソース関係に注目して,個体や個葉の光合成系の構築,維持,老化などに関して著者自身,あるいは研究室の先輩や同僚が行ってきた40年にわたる生理生態学的な研究を,基本的な事項の解説を交えながら振り返った.Photosynthetic production is limited by the lower of the activity of the sugar-synthesising leaves (source activity) or the sugar-consuming activity of non-photosynthetic organs such as roots and stems, storage organs and unfolding young leaves (sink activity). Ecophysiological studies on the construction, maintenance, and senescence of the leaf and whole-plant photosynthetic systems conducted by the author, his superiors or colleagues for these 40 years are reviewed, with particular attention to sink-source relationships

    細胞内共生により生じたミトコンドリアとプラスチドは独自のDNA(オルガネラDNA)を保持している.これまでにモデル植物であるシロイヌナズナを用いた研究から,このオルガネラDNAはプラスチドとミトコンドリアに共局在するエキソヌクレアーゼDPD1により成熟花粉や老化葉といった組織特異的に分解されることが明らかとなっている.このオルガネラDNAの分解産物が回収され,リン酸など養分の利用効率向上に寄与する機構が提唱されている.加えて母性遺伝にも関与していることが明らかとなっている.DPD1ホモログは種子植物に広く保存されており,イネにおいてもシロイヌナズナと同様に花粉中のオルガネラDNA分解を担っていることが明らかとなっている.本稿ではオルガネラエキソヌクレアーゼDPD1の機能とその生理的意義について議論する.Mitochondria and plastids, originating from endosymbiotic events, retain their own DNA (organelle DNA).DPD1, Mg2+-dependent organelle exonuclease targeted to plastids and mitochondria, is involved in the tissuespecific organelle DNA (orgDNA) degradation and has an important role for the nutrient salvage mechanism in Arabidopsis. DPD1 homologues are well conserved in flowering plants. The function of DPD1 in rice is also investigated. OsDPD1 degrades orgDNA during pollen maturation. In addition, tobacco DPD1 degrades orgDNA during pollen maturation, which contributes to maternal inheritance. Furthermore, DPD1 homologues are fused to MutS, DNA repair enzyme, in moss plants. In this review, we will discuss the diversification of DPD1 exonuclease

    シトクロムb6 f複合体の構成サブユニットは核または葉緑体ゲノムにコードされ,複合体の実態が明らかになる前からPSIIとPSIとの電子伝達を行なう成分として変異株が単離され重要な知見を提供している.モデル生物が確立していく過程で,葉緑体遺伝子解析と葉緑体遺伝子改変の技術確立とともに変異株が作出された.同時にエレクトロクロミックシフトによる活性測定技術も確立しスクリーニングと変異株解析に大きく貢献している.近年になって確立した遺伝子編集技術が新しいモデル生物に適用され新たな変異株が単離されており今後の動向が注目される.Nuclear and chloroplast genes encode the cytochrome b6 f complex subunits. The b6 f mutants have been isolated as mutants, carrying essential components for the electron transfer between PSII and PSI that had been disrupted, even before isolating the b6 f complex. During the establishment of model organisms, the newly developed chloroplast gene manipulation and analysis techniques have been applied to generate b6 f mutants for their characterization. In recent years, gene editing technology has been used to isolate b6 f mutant strains in new model organisms

    酸化・還元を基盤とした翻訳後修飾であるレドックス制御は,光合成システムの制御に重要な役割を果たしている.この制御系は従来,鍵因子であるチオレドキシンが電子伝達系によって生じる還元力を特定の標的酵素に伝達し,その酵素を活性化するというシンプルなものとして考えられていた.ところが近年,植物葉緑体のレドックス制御系は,数多くの制御因子および標的酵素が関わる複雑な還元力伝達ネットワークによって構成されていることが明らかになってきた.本稿では,最近の研究成果から見えてきたレドックス制御系の実体について,今後の研究展望を交えながら概説する.The redox regulation system is one of the post-translational mechanisms, which is important for photosynthesis.In recent years, many regulatory factors and target enzymes have been identified in plant chloroplasts, raising a possibility that the redox regulation system is composed of highly divergent redox pathways. We discuss a current understanding of the molecular basis and physiological importance of this regulatory network, with future perspectives of this research field

    Sustained energy dissipation in overwintering evergreens

    寒冷圏の常緑植物は,冬期は,光合成電子伝達を抑制する必要がある.光化学系IIを夏季の半分程度までに減らすことで電子伝達を抑制している植物も存在するが,もっとも重要なのは光化学系IIにおいて励起エネルギーを熱として放散し,電子伝達を抑制することである.冬季の常緑植物において持続的に活性化している熱放散(持続的熱放散)の分子メカニズムについて,本稿では,提唱されている主要な6つの仮説,すなわち,光阻害,スピルオーバー,ゼアキサンチンによる熱放散,Early lightinduced proteinによる熱放散,電荷再結合について概説する.Evergreen plants in cold regions need to suppress photosynthetic electron transport during winter. Some plants achieve this by reducing the level of photosystem II (PSII) to about half of its summer level, but the most important mechanism in general is increasing the dissipation of excitation energy as heat in PSII, thereby suppressing electron transport. This article summarizes recent progress toward understanding the molecular mechanisms of thermal dissipation (sustained heat dissipation) that are continuously active in evergreen plants during winter, focusing on six main hypotheses: photoinhibition, spillover, zeaxanthin-dependent thermal dissipation, thermal dissipation by Early light-induced protein, and charge recombination

    Structure and function of the LH1–RC complexes from purple phototrophic bacteria living in extreme environments

    紅色光合成細菌は酸素発生を伴わない原始の光合成生物であり,酸素発生型光合成生物が利用しない低エネルギーの近赤外光を吸収して,光―物質変換を営んでいる.その中心的な役割を担うのが光捕集1反応中心(LH1–RC)複合体である.構造および機能的に比較的シンプルであり,さまざまな極限環境に生息する種が存在するため,光合成タンパク質の構造機能相関を調べる上で非常に魅力的なツールである.本稿では,極限環境で生息する紅色光合成細菌の中から好熱性紅色細菌,好塩性紅色細菌,低エネルギーを利用する紅色細菌に焦点を絞り,ここ10年ほどで明らかになってきたLH1-RCの構造と機能について紹介する.Purple photosynthetic bacteria are ancient phototrophs that do not evolve oxygen. These organisms absorb lowenergy near-infrared (NIR) light—unused by oxygen-evolving phototrophs—and convert the NIR light energy into chemical energy, which is necessary for carbon assimilation. The key protein of purple bacterial photosynthesis is a light-harvesting 1–reaction center (LH1–RC) complex. Purple bacteria are relatively simple in their structures and functions and can survive under various extreme environments. Therefore, they serve as an excellent model for studying the structure-function relationships of photosynthetic proteins. Here, we focus on thermophilic, halophilic, and low-energy-utilizing species among purple photosynthetic bacteria living in extreme environments, and introduce the structure–function relationships of the LH1-RC complexes, which have been revealed in the last decade
