Improvement of yield and its related traits for backbone hybrid rice parent Minghui 86 using advanced backcross breeding strategies
How to overcome yield stagnation is a big challenge to rice breeders. An effective method for quickly developing new cultivars is to further improve an outstanding cultivar. In this study, three advanced backcross populations under yield selection that consist of 123 BC2F2:4 introgression lines (ILs) were developed by crossing Minghui 86 (recurrent parent, RP) with three high-yielding varieties (donors), namely, ZDZ057, Fuhui 838, and Teqing, respectively. The progeny testing allowed the identification of 12 promising ILs that had significantly higher mean grain yields than Minghui 86 in two environments. A total of 55 QTLs that affect grain yield and its related traits were identified, which included 50 QTLs that were detected using the likelihood ratio test based on stepwise regression (RSTEP-LRT) method, and eight grain yield per plant (GY) QTLs were detected using chi-squared (c2) test. Among these QTLs, five QTLs were simultaneously detected in different populations and 22 QTLs were detected in both environments. The beneficial donor alleles for increased GY and its related traits were identified in 63.6% (35 out of 55) of the QTLs. These promising ILs and QTLs identified will provide the elite breeding materials and genetic information for further improvement of the grain yield for Minghui 86 through pyramiding breeding

Features of the expressed sequences revealed by a large-scale analysis of ESTs from a normalized cDNA library of the elite indica rice cultivar Minghui 63
The indica subspecies of cultivated rice occupies the largest area of rice production in the world. However, a systematic analysis of cDNA sequences from the indica subspecies has not been performed. The aim of the present study was to collect and analyze the expressed sequence tags (ESTs) of indica rice on a large scale. A total of 39 208 raw sequences were generated from a normalized cDNA library prepared by use of 15 different tissues of the indica cultivar Minghui 63. After trimming, processing and analysis, 17 835 unique sequences were obtained, each of which presumably represents a unique gene. Of these sequences, 2663 were novel, and at least 70 were indica specific. Comparison of the Minghui 63 sequences with the ESTs/full-length cDNAs in GenBank revealed a large number of deletion/insertion/substitution (DIS) at both the inter- and intra-subspecific levels. The overall number of polymorphisms in the expressed sequences was higher in the inter-subspecific comparisons than in the intra-subspecific comparisons. However, the extent of DIS-based polymorphism was highly variable among different rice varieties. In total, 15 726 unique sequences, including 697 novel sequences, were assigned to regions where large numbers of quantitative trait loci (QTLs) for agronomic traits had been detected previously. These results may be useful for developing new molecular markers for genetic mapping, detecting allelic polymorphisms associated with phenotypic variations between rice varieties, and facilitating QTL cloning by providing the starting points for candidate-gene identification
INTERVIEW WITH DR. REN MINGHUI, MINISTRY OF HEALTH, CHINA
Dr. REN is the Director-General of the Department of International Cooperation at the Ministry of Health (MOH) of the People’s Republic of China. Throughout his career, Dr. REN has been involved in numerous health policy research projects including the “Health care systems research in rural China” (MOH), “Financing and organization of health care services in poverty areas of China” (World Bank), “The pilot study of health insurance reform in urban China” (Chinese State Council) and the “Experiment in community health protection-policy exploration, training and demonstration program” (UNDP). In recent years, as a senior representative of the Chinese government, he served as a leading board member in a number of international organizations including WHO, UNAIDS, and the Global Fund to Fight AIDS, TB and Malaria. He is also the point of contact for many Chinese bilateral cooperation mechanisms in health. Dr. REN received his MD, MPH and PHD from Xi’an Medical University in 1987, Harvard School of Public Health in 1993 and Peking University in 2008, respectively. Dr. Lingling Zhang initiated this interview when she met with Dr. REN at the Harvard America-China Health Summit organized by the Harvard School of Public Health China Initiative. Dr. REN was one of the invited guests of honor at the Summit. Dr. REN responded to Dr. Zhang’s questions in writing after he returned to China
A targeted proteomic assay for the measurement of plasma proteoforms related to human aging phenotypes
Circulating polypeptides and proteins have been implicated in reversing or accelerating aging phenotypes, including growth/differentiation factor 8 (GDF8), GDF11, eotaxin, and oxytocin. These proteoforms, which are defined as the protein products arising from a single gene due to alternative splicing and PTMs, have been challenging to study. Both GDF8 and GDF11 have known antagonists such as follistatin (FST), and WAP, Kazal, immunoglobulin, Kunitz, and NTR domain-containing proteins 1 and 2 (WFIKKN1, WFIKKN2). We developed a novel multiplexed SRM assay using LC-MS/MS to measure five proteins related to GDF8 and GDF11 signaling, and in addition, eotaxin, and oxytocin. Eighteen peptides consisting of 54 transitions were monitored and validated in pooled human plasma. In 24 adults, the mean (SD) concentrations (ng/mL) were as follows: GDF8 propeptide, 11.0 (2.4); GDF8 mature protein, 25.7 (8.0); GDF11 propeptide, 21.3 (10.9); GDF11 mature protein, 16.5 (12.4); FST, 29.8 (7.1); FST cleavage form FST303, 96.4 (69.2); WFIKKN1, 38.3 (8.3); WFIKKN2, 32.2 (10.5); oxytocin, 1.9 (0.9); and eotaxin, 2.3 (0.5). This novel multiplexed SRM assay should facilitate the study of the relationships of these proteoforms with major aging phenotypes
Engineering study on lithium isotope separation by ion exchange chromatography
0000-0002-1437-2273We have been studying the lithium isotope separation by ion exchange chromatography for the engineering development. We performed the lithium isotope separation experiments by chromatography. From the results, we estimated the migration lengths required to obtain the 20 %, 50 %, and 90 % enriched lithium-6, the required lengths are 340, 1600, and 5800 m, respectively. We studied the detection methods of lithium adsorption band boundary in order to design the circular type chromatographic equipment, which type equipment is required for the industrial plant. Based on these investigations, we designed the automatic circular type chromatographic equipment with the boundary interface detector and manufactured the prototype equipment.journal articl
