    Yuanji Zhang, known as Xiaozhai (courtesy name) and Jusheng (pseudonym), was born in Zhenjiang, China. Zhang was a Hanlin Academy bachelor and a central figure of Hundred Days’ Reform (a national political movement in 1898). After the reform fails, Zhang dedicated himself to publication business and served as a director and president of Commercial Press. He believed that education and culture can help revitalize the country. During his tenure of the president in Commercial Press, he organized classic documents and edited text books and made Commercial Press became the most influential educational institution in China.張元濟,字筱齋,號菊生,浙江人。曾為翰林院庶吉士,積極投入維新運動。政變失敗後,張元濟投入出版事業,曾任商務印書館所長及董事長等職務。任職其間整理古籍、參與教科書編定並推廣教育,期望利用教育及文化以振興國業。How can one allow himself or herself not to study every day?     Development of Mn(III) Porphyrins as Gd-free Contrast Agents for Targeted MR Imaging

    Personalized medicine requires precision diagnosis, and a targeted medical imaging method addressing this requirement is highly anticipated. Targeted magnetic resonance imaging (MRI) requires a contrast agent (CA) capable of detecting specific cellular or molecular biomarkers in a sensitive and selective manner. Currently, mainstream MRI CAs are Gd(III)-based, which is limited by low contrast efficiency (r1) and toxicity of free Gd(III) upon metal dissociation. To address these limitations, we are developing a new class of Gd(III)-free CAs based on the biocompatible Mn(III) porphyrin (MnP), which is desirable for their high r1 at high fields, stability against metal dissociation, and versatility for structural modification to achieve functional diversity. In this thesis, the development of MnP-based CAs for targeted MRI is explored. In chapter 2, we studied MnP2, the first MnP-based blood pool agent (BPA) on its binding behavior with its target, human serum albumin (HSA). We unexpectedly discovered a novel sequential binding model, in which MnP2 binds first to a kinetically favorable site, followed by migration to a thermodynamically favorable hydrophobic site (subdomain IB), causing r1 decrease in MnP2, possibly due to limited water accessibility. Based on the crucial findings, in chapter 2, a series of MnPs designed to overcome the water accessibility issue, either by twisting MnP2’s geometry (chapter 3) or incorporating smaller, more polar functional groups (chapter 4) were developed as next-generation BPAs. Amongst them, MnTriCPP especially showed significant r1 increase at high fields upon HSA binding (chapter 4). In chapter 5, MnTriCP-B(OH)2 was developed as a saccharide-targeted CA based on boronic acid-diol interactions, and the binding was found to be pH dependent. In chapter 6, MnTriCP-NTA was designed to target His-tag, a post-translational modification widely utilized in protein purification. Finally, in chapter 7, MnMOB and MnEOB were developed as MRI probes for organic anion transport proteins (OATPs). In vitro and in vivo evaluations show that MnEOB is highly specific and sensitive towards OATP uptake, demonstrating its potential for targeted liver imaging as well as gene reporting. The potential for improved engineering project management and the promotion of sustainable development are both enhanced by Building Information Modeling (BIM), a sophisticated technology that is generally acknowledged. This study delves into the use of building information modeling (BIM) in project management, focusing on how it contributes to the attainment of sustainable development objectives. This study provides an overview of building information modeling (BIM), its history and evolution, and its complex link with sustainable project management via a thorough literature analysis. Particularly noteworthy is a case study that shows how BIM helps with quality assurance, scheduling, and cost control. As shown in the case study, the results demonstrate the critical role of BIM in enhancing management efficiency and conserving resources. The study also highlights potential avenues for future research and provides actionable recommendations for improving present procedures

    Translational control of autophagy rejuvenates immune responses

    As our body's guardian, the immune system maintains systemic health through removal of pathogens, damage and cancer. Ageing of the immune system is associated with compromised immune responses as well as decreased tumour surveillance and is therefore a key risk factor for major diseases in the elderly. Adaptive immune responses are mediated by T and B lymphocytes, and failure in adaptive immunity is a particular hallmark of the ageing organism. Here we show that autophagy is impaired in aged murine B lymphocytes, and loss of autophagy causes severely reduced B cell responses. Our data demonstrate that B cell senescence can be reversed in an autophagy-dependent manner by spermidine, a naturally occurring polyamine metabolite. Mechanistically, our study reveals that the translation factor eIF5A, that requires spermidine for its activation, regulates the expression of the master autophagy/lysosomal transcription factor TFEB. Importantly, we show in humans that spermidine, eIF5A and TFEB levels decrease with age and may serve as ageing biomarkers. Taken together our results indicate that the translational control of autophagy by eIF5A is dysregulated with ageing, and identify a novel pathway with therapeutic implications

    中国では二千年にもわたり皇帝を頂点とする政治体制が存続していた。なかでも中国明代の政治は強大な皇帝権力が特徴として挙げられる。なぜ皇帝政治が存続したのかを明らかにするためには、強大な皇帝権力を肯定的にとらえていた人物の思考を理解することが必要である。そこで本稿では明代後期の思想家管志道の思考と当時の政治状況がどのように関連しているのかを明らかにするため、管志道の奏議『奏疏稿』収録の各序文を用いて万暦五年に発生した内閣首輔張居正の奪情問題における管志道の立場とその後の管志道の自己イメージ形成について検討した。その結果、管志道は張居正奪情問題においては保身を優先し、この政治的体験によって政治主体としての士大夫の責任から逃避しようという思想にたどりついたことを明らかにした。In order to understand the factors that sustained the monarchy in Premodern China, this essay focuses on Guan Zhidao 管志道 (1536-1608), a scholar bureaucrat in the late Ming. He argued that all people should obey laws issued by the Son of Heaven and scholar bureaucrats should not play an active role in the political sphere. Chiefly using the prefaces in a collection of Guan's memorials known as the Zouyigao 奏議稿, this paper explains the relation between the formation of Guan's thought and his political stance regarding the issue of filial mourning by Zhang Juzheng 張居正 (1525-1582) in 1577 and thereafter. In 1577, Zhang Juzheng, a Chief Grand Secretary who was politically powerful, lost his father. According to Ming law, Zhang was required to leave the court and return to his hometown in order to practice filial mourning for three years, but Zhang ventured not to do so. He had the emperor order him not to practice filial mourning. Five officials who belonged to Hanlin Academy and Ministry of Justice then opposed this move, but they were punished at court with a caning of from sixty to eighty strokes and seriously injured. After Zhang's death, the five officials who had been beaten were reinstated and promoted. According to the previous studies, Guan also lost his official post over this matter and thereafter he devoted himself to a scholarly life unconnected to the political sphere. The prefaces of Zouyigao show Guan's stance on the issue of filial mourning. One preface titled Wanyanshucaozhuiyan 万言書草綴言, whose author was Zhang Hao 張浩, a disciple of Guan, shows that Guan was a consultant and a supporter to the anti-Zhang officials. But he did not offer direct opposition to Zhang and insisted that he had no official post and no responsibility to speak out. At that time, Guan was waiting for a vacant post in Beijing, so he did not wish to openly oppose Zhang, but on the other hand, some of the anti-Zhang officials were closely related to him. For example, Zhao Yongxian 趙用賢 (1535-1596), a Hanlin Academy official who was caned at court, employed Guan as a tutor for his family. It must be admitted that Guan had tried to protect himself during the strife over the issue of Zhang's filial mourning and this experience caused him to lose confidence in his position that scholar bureaucrats had responsibility for political matters. Thus, in order to escape responsibility as a scholar bureaucrat, he ultimately came to advocate that scholar bureaucrats should act as subordinates to the Son of Heaven and should not play an active role in politics. After Zhang's death in 1582, Guan started to call for his own reinstatement. He had his acquaintances write prefaces to his memorials in order to embellish his image in the dispute over filial mourning. At that time, many scholar bureaucrats honored not only the five officials who had been beaten but also other officials who took a critical stance toward Zhang over the filial mourning issue, even if they had not been punished. This situation was brought about by the reinstatement of Wang Xijue 王錫爵 (1534-1614) in 1584. Wang had criticized Zhang over the issue of filial mourning and resigned afterward. Wang's reinstatement with full honors spurred Guan's call for reinstatement. This was due to the fact that Guan and Wang were from the same hometown of Taicang 太倉, and they had a close relation through their religious faith in Wang's daughter Wang Tanyang 王曇陽. After her death in 1580, Guan and Wang still kept in close communication. Wang later became a Grand Secretary, but he gradually lost the favorable reputation he had garnered as an opponent of Zhang due to disputes over the selection of a successor to the emperor. This outcome influenced Guan's political circumstances. Guan finally left the political sphere because of their close political stance. Guan's political failures created a gap that separated his political thought from that of other famous scholar bureaucrats of the Donglin party 東林党 who believed that they should play an active role in the political matters

    Reimagining permanence in architecture: How adaptable and circular design principles can enhance the value and durability of buildings

    The misguided idea of permanence in architecture has proven to be an illusion (Brand, 1994; Ford, 1997). Present-day solutions such as the adaptive and circular design of buildings strive to enable frequent changes in buildings to satisfy the needs of users in our transient society. However, this challenges the idea of permanence in architecture, a concept that people profoundly believe in (Ford, 1997).By outlining the new notion of permanence as identified by Katrina Touw (2006) and the principles of adaptable and circular design as identified in recent literature studies (Asker et al., 2021; Hamida et al., 2022). Then establishing a framework which shows the relation between the two topics used to evaluate five case studies. This thesis seeks to identify if the new approach to building design strives to achieve permanence and in what way.We can conclude that by thinking about the inherent value of the building at its end-of-life whether it be to maintain, reuse, recycle, or repurpose, the stakeholders involved in the design process actively engage in achieving permanence.AR2A011Architectural History ThesisArchitecture, Urbanism and Building Science
