No full textThe 5'-terminal cloverleaf (CL)-like RNA structures are essential for the initiation of positive- and negative-strand RNA synthesis of entero- and rhinoviruses. SLD is the cognate RNA ligand of the viral proteinase 3C (3Cpro), which is an indispensable component of the viral replication initiation complex. The structure of an 18mer RNA representing the apical stem and the cGUUAg D-loop of SLD from the first 5'-CL of BEV1 was determined in solution to a root-mean-square deviation (r.m.s.d.) (all heavy atoms) of 0.59 A (PDB 1Z30). The first (antiG) and last (synA) nucleotide of the D-loop forms a novel ‘pseudo base pair’ without direct hydrogen bonds. The backbone conformation and the base-stacking pattern of the cGUUAg-loop, however, are highly similar to that of the coxsackieviral uCACGg D-loop (PDB 1RFR) and of the stable cUUCGg tetraloop (PDB 1F7Y) but surprisingly dissimilar to the structure of a cGUAAg stable tetraloop (PDB 1MSY), even though the cGUUAg BEV D-loop and the cGUAAg tetraloop differ by 1 nt only. No full textWerner A. Untersuchungen zur funktionellen Bedeutung von Gangliosiden bei der E-Selektin-vermittelten Zell-Zell-Adhäsion. Bielefeld (Germany): Bielefeld University; 2001.Die Infektionsabwehr in vivo beruht neben der humoralen auch auf der zellulären Immunantwort. Dabei wird vom Organismus u.a. die Strategie der Phagocytose zur Beseitigung von Erregern angewendet. Die dafür verantwortlichen Zellen sind u.a. Granulocyten, die aus dem Blutstrom zum Ort der Entzündung in das Gewebe geleitet werden müssen. Der Gesamtprozess der Adhäsionskaskade besteht aus vier Schritten und beginnt mit dem Verlangsamen der Granulocytengeschwindigkeit durch "Anbinden" und "Rollen" (engl.: "tethering" und "rolling"). Mit dem zweiten Schritt werden die Granulocyten aktiviert und adhärieren schließlich im dritten Schritt fest an die Endothelzellen. Der vierte und letzte Schritt beinhaltet die Durchdringung der Endothelzellbarriere (Diapedese) und das anschließende Einwandern in das darunterliegende Gewebe. Cytokin-induzierte Endothelzellen exprimieren transient E-Selektin, das membranständig auf der Plasmamembranoberfläche präsentiert wird und die Granulocyten über Protein-Kohlenhydrat-Wechselwirkungen in situ "einfängt". Diese Interaktion stellt den ersten Schritt der Kaskade dar. Als Ligand des E-Selektins wurde das sialyl-Lewis x-Epitop (sLe x; Neu5Ac[alpha]2-3Gal[beta]1-4(Fuc[alpha]1-3)GlcNAc-R) entdeckt. Dieses Epitop wurde sowohl auf Glykoproteinen als auch auf granulocytären Gangliosiden detektiert.
Das Arbeitsziel der vorliegenden Arbeit war die Untersuchung der funktionellen Bedeutung dieser Ganglioside für die E-Selektin-vermittelte Zell-Zell-Wechselwirkung. Dazu wurden ein statisches und ein dynamisches Zell-Zell-Adhäsionssystem entwickelt. Als Modellzelllinien wurden HL-60 Zellen statt primärer Granulocyten und E-Selektin-transfizierte CHO-Zellen (CHO-E) bzw. primäre humane Nabelschnurendothelzellen (HUVEC) statt primärer mikrovasculärer Endothelzellen eingesetzt.
Massenspektrometrische Untersuchungen belegten die grundsätzliche strukturelle Übereinstimmung der detektierten Ganglioside von humanen Granulocyten und HL-60 Zellen. Es wurden fünf verschiedene humane Granulocytengangliosidfraktionen, zwei HL-60 Gangliosidfraktionen und eine humane Hirngangliosidfraktion mittels HPTLC getrennt und im Overlay-Assay auf Anwesenheit von CSLEX- und E-Selektin-Epitopen getestet. Anhand von statischen und dynamischen Zell-Zell-Adhäsionstests wurde die funktionelle Bedeutung von Gangliosiden bei der E-Selektin-vermittelten Zell-Zell-Adhäsion nachgewiesen
Appropriate Similarity Measures for Author Cocitation Analysis
No full textWe provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
