TW-01, a piperazinedione-derived compound, inhibits Ras-mediated cell proliferation and angioplasty-induced vascular restenosis
Purpose: Vascular smooth muscle cell (VSMC) proliferation plays a critical role in the pathogenesis of atherosclerosis and restenosis. This study investigated piperazinedione derived compound TW-01-mediated inhibitory effects on VSMC proliferation and intimal hyperplasia.
Methods: Cell proliferation was determined using [H-3]-thymidine incorporation and MTT assay; cell cycle distribution was measured using flow cytometry; proteins and mRNA expression were determined using western blotting and RT-PCR analyses; DNA binding activity of nuclear factor-kappa B (NF-kappa B), as measured using enzyme linked immunosorbent assays (ELISA); in vivo effects of TW-01 were determined using balloon angioplasty in the rat.
Results: TW-01 significantly inhibited cell proliferation. At the concentrations used, no cytotoxic effects were observed. Three predominant signaling pathways were inhibited by TW-01: (a) extracellular signal-regulated kinase (ERK)1/2 mitogen-activated protein kinase (MAPK) activation and its downstream effectors of c-fos, c-jun, and c-myc; (b) DNA binding activity of nuclear factor-kappa B (NF-kappa B); and, (c) Akt/protein kinase B (PKB) and cell cycle progression. Furthermore, TW-01 also inhibited Ras activation, a shared upstream event of each of these signaling cascades. In vascular injury studies, oral administration of TW-01 significantly suppressed intimal hyperplasia induced by balloon angioplasty.
Conclusion: The present study suggests that TW-01 might be a potential candidate for atherosclerosis treatment. (C) 2016 Elsevier Inc. All rights reserved

Pristidia ramosa can be easily distinguished from P. cervicornuta sp. nov. by the embolus distinctly longer, the presence of basal teeth on the RTA, RTA expanded, with a blunt tip, and by the absence of TA (Figs 13, 14). Yu, Sun & Zhang (2012: 45) highlighted the differences of this species from P. prima. Description. See Yu, Sun & Zhang (2012). Intraspecific variation. There is almost no difference between the male from Taiwan (HUBU-TW- 130177, Figs 10, 12–14) and the holotype from Jiangxi (Yu, Sun & Zhang 2012: figs 1–7, 12–14). However, some intraspecific variation is exhibited by females from different localities, mostly related to different degrees of sclerotization. The female from Taiwan (HUBU-TW- 130178, Fig. 11) is distinctly larger in size and darker in color than the paratype from Jiangxi (Yu, Sun & Zhang, 2012: fig. 8). Furthermore, in HUBU-TW- 130178, the epigynal ventral plate is more sclerotized (Fig. 15) than in the paratype (Yu, Sun & Zhang 2012: fig. 9); both spermathecae and bursae are totally transparent in HUBU-TW- 130178 (Fig. 16), while more sclerotized in the paratype (Yu, Sun & Zhang 2012: figs 10, 16). Natural history. The spiders inhabit forests in low mountain areas (210–800 m). This is in accordance with data of the type locality provided by Yu, Sun & Zhang (2012) and Huang & Chen (2012). Distribution. Mt. Jinggang in Jiangxi, Mt. Fanjing in Guizhou and New Taipei City in Taiwan, China. The present data showed the extension of the known range of this species of about 560 km to the northwest (Mt. Fanjing) and of 750 km to the southeast (Taiwan) from the type locality (Fig. 17). Remark. Although we have not examined the type specimens of C. expansa, the long filiform embolus, the expanded RTV with a subapical flange, the bell-shaped atrium, and the course of vulvar duct system shown in the original illustrations (Huang & Chen 2012, fig. 15A–G), leave no doubts on this synonymy. Huang & Chen (2012) was published in May, 2012 (day of publication not specified), while Yu, Sun & Zhang (2012) was published in May, 3, 2012. According to the article 21.3 of the International Code of Zoological Nomenclature, the publication date of Huang & Chen (2012) must be specified as May, 31, 2012. Sticholotis petila Yu & Pang (Figures 41–43, 169–173, 297) Sticholotis petila Yu & Pang in Yu et al., 1993: 467. Diagnosis. This species is very similar to S. ruficeps in dorsal color pattern, but can be distinguished by its elytra with arcuate row of large punctures at mid length near suture (Fig. 41) and distinctly different shape of the penis capsule (Fig. 170). Description. TL: 2.32–2.44 mm, TW: 1.98–2.11 mm, TH: 1.05–1.19 mm, TL/TW: 1.16–1.18; PL/PW: 0.43– 0.44; EL/EW: 0.94–0.97; HW/TW: 0.38; PW/TW: 0.63. Head reddish brown, mouthparts yellow. Pronotum reddish brown and scutellum black. Elytra dark brown, with narrow yellowish brown band along lateral margins, each elytron with two large yellowish spots, anterior very large, irregular, at middle of basal margin (touching margin), posterior small, 1/2 size of anterior spot, at apical third (Figs 41–43). Underside reddish brown, middle of meso- and metaventrite black. Legs reddish brown. Body almost circular in outline, hemispherical, shiny and glabrous. Head frontal punctures medium and densely distributed, 0.5–1.5 diameters apart, with scattered short setae, interocular distance of eyes 0.58x head width (Fig. 43). Pronotal punctures fine and conspicuous, 1.0–2.0 diameters apart, slightly smaller than those on head. Elytral punctures medium, similar to those on pronotum, 1.0–1.5 diameters apart; each elytron with arcuate row of large punctures near suture at mid length. Elytral margins wide, visible from above. Prosternal punctures inconspicuous, with long sparse setae. Mesoventral surface shagreened, punctures coarse, with scattered short setae. Metaventrite broad and slightly shagreened, flat medially, covered by coarse and dense punctures distributed at center. Elytral epipleuron broad, complete to apex, feebly foveate to accommodate femoral tips of hind legs. Wings well-developed. Male genitalia: Penis short and slender, strongly curved at base, capsule broadening basally and with a small inner process (Figs 170–171); tegmen with penis guide slightly shorter than paramere, in lateral view thin and straight; parameres very thin (Fig. 172). Female genitalia: Ovipositor elongate and narrowly triangular, styli inconspicuous (Fig. 173). Specimens examined.
