    Blocking of orexin receptors in the paraventricular nucleus of the thalamus has no effect on conditioned fear

    The paraventricular nucleus of the thalamus (PVT) projects to the central nucleus of the amygdala and recent experimental evidence indicates a role for the PVT in conditioned fear. Furthermore, the PVT contains a high density of orexin receptors and fibers and acute injections of orexin antagonist into the PVT produce anxiolytic effects. The present study was done to determine if administration of a dual orexin receptor antagonist (DORA) in the region of the PVT interfered with the expression of conditioned fear in rats exposed to cued and contextual conditioning paradigms. Infusion of 0.5 µl of the DORA N-biphenyl-2-yl-1-{[(1-methyl-1H-benzimidazol-2yl) sulfanyl] acetyl}-L-prolinamide at a concentration of 0.1, 1.0, and 10 nmol had no effect on the freezing produced by exposing rats to an auditory cue or the context associated with foot shock. In contrast, the 1.0 and 10 nmol doses were anxiolytic in the social interaction test. The results of the present study do not support a role for orexin receptors in the PVT in the expression of learned fear. The finding that the 1.0 and 10 nmol doses of DORA in the PVT region were anxiolytic in the social interaction test is consistent with other studies indicating a role for orexins in the PVT in anxiety-like behaviors

    Peritraumatic startle response predicts the vulnerability to develop PTSD-like behaviors in rats: a model for peritraumatic dissociation

    Peritraumatic dissociation, a state characterized by alteration in perception and reduced awareness of surroundings, is considered to be a risk factor for the development of post-traumatic stress disorder (PTSD). However, the predictive ability of peritraumatic dissociation is questioned for the inconsistent results in different time points of assessment. The startle reflex is an objective behavioral measurement of defensive response to abrupt and intense sensory stimulus of surroundings, with potential to be used as an assessment on the dissociative status in both humans and rodents. The present study examined the predictive effect of acoustic startle response (ASR) in different time points around the traumatic event in an animal model of PTSD. The PTSD-like symptoms, including hyperarousal, avoidance, and contextual fear, were assessed 2-3 weeks post-trauma. The results showed that 1) the startle amplitude attenuated immediate after intense footshock in almost half of the stress animals, 2) the attenuated startle responses at 1 hour but not 24 hours after stress predicted the development of severe PTSD-like symptoms. These data indicate that the startle alteration at the immediate period after trauma, including 1 hour, is more important in PTSD prediction than 24 hours after trauma. Our study also suggests that the startle attenuation immediate after intense stress may serve as an objective measurement of peritraumatic dissociation in rats

    Lesions of the posterior paraventricular nucleus of the thalamus attenuate fear expression

    The paraventricular nucleus of the thalamus (PVT) has generated interest because of its strong projections to areas of the brain associated with the regulation of emotional behaviors. The posterior aspect of the PVT (pPVT) is notable for its projection to the central nucleus of the amygdala which is essential for the expression of a conditioned fear response. The present study was done to determine if the pPVT is involved in the expression of fear by examining the effect of post-conditioning lesions of the pPVT. Male rats were trained to bar press for food pellets on a variable ratio schedule. Fear conditioning was done using auditory tones (30 s) that co-terminate with footschocks (0.65 mA, 1.0 s). Rats were anesthetized 24 hours later and small bilateral electrolytic lesions of the pPVT were made. Fear expression to the tone was assessed using suppression of bar-pressing and freezing after one week of recovery from the surgical procedure. Small bilateral lesions of the pPVT increased bar-pressing for food and decreased freezing during the presentation of the conditioned tone. Lesions of the pPVT had no effect on fear extinction, fear conditioning to a novel tone, or the motivation for food as assessed using a progressive ratio schedule. The results of the experiment support a role for the pPVT in fear expression. In contrast, the pPVT does not appear to be involved in fear learning or extinction nor does it appear to play a role in the motivation of rats to bar press for food

    Identification of microRNAs and their targets associated with fruit-bagging and subsequent sunlight re-exposure in the ‘Granny Smith’ apple exocarp using high-throughput sequencing

    Bagged fruits of green apple cultivar ‘Granny Smith’ have been found to turn cardinal red after debagging during fruit-ripening in the Loess Plateau region of China. To understand such phenomenon at post-transcriptional level, we have investigated the roles of microRNAs (miRNAs) in response to debagging. Three small RNA libraries were primarily constructed from peels of ‘Granny Smith’ apples subjected to bagging followed by sunlight re-exposure treatments (0h, 6h, 1d) (debagging), and from peels of apples without any bagging treatments (0h, 6h, 1d). 201 known miRNAs belonging to 43 miRNA families and 220 novel miRNAs were identified via high-throughput sequencing. Some miRNAs were found to be differentially expressed after debagging, which indicated that miRNAs affected anthocyanin accumulation through their target genes in mature apple. To further explore the effect of debagging on miRNAs regulating the expression of anthocyanin regulatory genes, four miRNAs and their target genes regulating anthocyanin accumulation, miR156, miR828, miR858 and miR5072, were compared between green cultivar ‘Granny Smith’ and red cultivar ‘Starkrimson’. Results showed that mdm-miR828 and mdm-miR858 regulated anthocyanin contents in both apple cultivars, while mdm-miR156 only affected anthocyanin accumulation in ‘Granny Smith’, and miR5072 affected anthocyanin accumulation in ‘Starkrimson’. Additional analysis of gene ontology for the differentially expressed miRNAs after debagging treatments and their predicted target genes showed that they were involved in photo-protective response after debagging from 0h to 1d; they might play important roles in fruit development and adaptation to high light stress

    Sample preparation for mass spectrometry imaging of plant tissues: a review

    Mass spectrometry imaging (MSI) is a mass spectrometry based molecular ion imaging technique. It provides the means for ascertaining the spatial distribution of a large variety of analytes directly on tissue sample surfaces without any labeling or staining steps. These advantages make it an attractive molecular histology tool in medical, pharmaceutical and biological research. Likewise, MSI has started gaining popularity in plant sciences; yet, information regarding sample preparation methods specific for plant tissues is still limited. Sample preparation is a crucial step that is directly associated with the quality and authenticity of the imaging results, it therefore demands in-depth studies based on the characteristics of plant samples. In this review, a sample preparation pipeline is discussed in detail and illustrated through selected practical examples. In particular, special concerns regarding sample preparation for plant imaging are critically evaluated. Finally, the applications of various MSI techniques in plants are reviewed according to different classes of plant metabolites

    Frequency-dependent changes in the amplitude of low-frequency fluctuations in Internet gaming disorder

    Neuroimaging studies have revealed that the task-related functional brain activities are impaired in Internet gaming disorder (IGD) subjects. However, little is known about the alternations in spontaneous brain activities about them. Recent studies have proposed that the brain activities of different frequency ranges are generated by different nervous activities and have different physiological and psychological functions. Thus, in this study, we set to explore the spontaneous brain activities in IGD subjects by measuring the fractional amplitude of low-frequency fluctuation (fALFF), to investigate band-specific changes of resting-state fALFF. We subdivided the frequency range into five bands based on literatures. Comparing to healthy controls, the IGD group showed decreased fALFF values in the cerebellum posterior lobe and increased fALFF values in superior temporal gyrus. Significant interactions between frequency bands and groups were found in the cerebellum, the anterior cingulate, the lingual gyrus, the middle temporal gyrus and the middle frontal gyrus. Those brain regions are proved related to the executive function and decision-making. These results revealed the changed spontaneous brain activity of IGD, which contributed to understanding the underlying pathophysiology of IGD

    How the risky features of previous selection affect subsequent decision-making: Evidence from behavioral and fMRI measures

    Human decision making is rarely conducted in temporal isolation. It is often biased and affected by environmental variables, particularly prior selections. In this study, we used a task that simulates a real gambling process to explore the effect of the risky features of a previous selection on subsequent decision making. Compared with decision making after an advantageous risk-taking situation (Risk_Adv), that after a disadvantageous risk-taking situation (Risk_Disadv) is associated with a longer response time (RT, the time spent in making decisions) and higher brain activations in the caudate and the dorsolateral prefrontal cortex. Compared with decisions after Risk_Adv, those after Risk_Disadv in loss trials are associated with higher brain activations in the left superior temporal gyrus and the precuneus. Brain activity and relevant RTs significantly correlated. Overall, people who experience disadvantageous risk-taking selections tend to focus on current decision making and engage cognitive endeavors in value evaluation and in the regulation of their risk-taking behaviors during decision making

    Similarities and differences in decision-making impairments between autism spectrum disorder and schizophrenia

    Although individuals with autism spectrum disorders (ASD) and schizophrenia (SCH) share overlapping characteristics and may perform similarly on many cognitive tasks, cognitive dysfunctions common to both disorders do not necessarily share the same underlying mechanisms. Decision making is currently a major research interest for both ASD and SCH. The aim of the present study was to make direct comparisons of decision making and disorder-specific underlying neuropsychological mechanisms between the two disorders. Thirty-seven participants with ASD, 46 patients with SCH and 80 healthy controls (HC) were assessed with the Iowa Gambling Task (IGT), which measures decision-making under ambiguity, and the Game of Dice Task (GDT), which measures decision-making under risk. The results revealed that both the ASD and SCH groups had deficits for both the IGT and GDT compared with the HC. More importantly, in the IGT, participants with ASD displayed a preference for deck A, indicating that they had more sensitivity to the magnitude of loss than to the frequency of loss, whereas patients with SCH displayed a preference for deck B, indicating that they showed more sensitivity to the frequency of loss than to the magnitude of loss. In the GDT, the impaired performance might be due to the deficits in executive functions in patients with SCH, whereas the impaired performance might be due to the deficits in feedback processing in participants with ASD. These findings demonstrate that there are similar impairments in decision-making tasks between ASD and SCH; however, these two disorders may have different impairment mechanisms
