In vitro phospholipid biosynthesis for growing and dividing minimal cells
Natural life is extraordinarily complex, which by definition means that it has many interconnected and functioning parts. The goal of synthetic biology is to engineer living systems, though due to their very complexity they remain recalcitrant to engineering. What if it were possible to reduce the complexity to a finite amount of parts that are well understood and therefore possible to manipulate. That is the motivation for constructing a so called minimal cell
Attention-deficit/hyperactivity disorder: a systems perspective. A comprehensive qualitative analysis of the literature
Plan BAttention-Deficit/Hyperactivity Disorder (ADHD) is a neurological syndrome resulting in problems with self-regulation. ADHD is characterized by inattention and/or hyperactivity-impulsivity which causes impairment in at least two settings. These characteristics often arise in childhood and were previously thought to diminish during adolescence and disappear by adulthood. However, studies over the past decade indicate that although some symptoms may decrease in severity, ADHD does continue throughout adulthood for many individuals. The impact of this disorder is felt not only by the individuals with ADHD, but also by the various systems in which these individuals live, work, and socialize. In this comprehensive analysis of the literature ADHD is discussed from a systemic perspective. Initially the symptoms and characteristics that define ADHD are reviewed along with other disorders that commonly coexist with ADHD, and what research tells us does and does not cause ADHD and the development of the co-morbid disorders. Prevalence of ADHD and co-morbidity is also reviewed. The term “system” is defined and the various systems presented as related to this topic, from the micro-system of the individual to the macro-system of society at large. As the characteristics of ADHD are exhibited inconsistently according to the age of the individual and the context of the interaction, these are addressed, as appropriate, in the categories of child, adolescent/teen, and adult for each system. The first system discussed is the internal system of the individual in which ADHD resides. This micro-system consists of the biological, intellectual, and psychological components. This is followed by examination of this micro-system of the individual interacting in the family system, both nuclear and extended. The common effects on other family members in the system and the system as a whole are discussed. With understanding of the individual and his/her family with ADHD as a foundation, the view is broadened to other systems in which both the individual and his/her family operate. The discussion includes the reciprocal effects of ADHD and the educational, employment, social/recreational, and legal systems, as well as the influence of interactions between these systems. The presentation of the pervasive impact of ADHD on all systems is followed by analysis of the different treatment options and effectiveness. Included are interventions in the areas of interpersonal relationships within the family, social relationships, leisure/recreation, school, work, and in the legal arena. Since the acceptance and attitude about ADHD are important factors in interaction and treatment effectiveness, these are also investigated throughout. A summary of the analysis of the literature is presented in the second chapter. This is followed by conclusions and a discussion of implications for marriage and family therapists

A Historical Research Study of the Evolution of Attention Deficit Disorder
Across the United States of America, schools and social service agencies have become increasingly aware of a disability called Attention Deficit Disorder (A.D.D.). Families and employers are learning the name for this confusing, frustration, seemingly uncontrolled problem. The focus of this thesis will be to trace the origin and historical development of this disorder. It examines the work of those who were involved in identifying, defining and treating the disorder. The author has attempted to draw connections between history, discovery, research and implications for treatment, practice and public policy
Banana marketing performance in Blantyre Agricultural Development Division, Malawi
1990 Fall.Includes bibliographical references. This study provides insight into the complexities underlying the marketing of bananas in Blantyre Agricultural Development Division (A.D.D.), in southern Malawi. While farmers formed the core of the study, varying numbers of wholesalers/dealers, retailers and truckers were also contacted. A dualistic system of marketing operated in the area; that is, farmers sold their bananas both directly to consumers and also through middlemen. Sales, in order of importance based on patronage, were directly to the urban markets by the farmers themselves, to the middlemen/traders, to the village or roadside markets and lastly to the local markets. The most important problem was that of transportation, especially in getting the bananas to the urban markets. The main issue was the condition of the outlet road system. It appears that there is a lot of risk taking on the part of the truckers who opt to operate in the area. A detailed market performance analysis was not possible due to the paucity of relevant data. However, imperfect indicators of performance were highlighted. These included physical losses as bananas passed through various channels; price setting by market authorities; erratic supplies and inconsistent volume measures. Retail prices for a period of 69 days showed highly significant differences among mean prices of the three grades of bananas - large, medium and small. There appeared to be no relationship at all between the price movements for the large and medium size bananas. However, those for large and small, as well as medium and small, showed considerably higher degrees of positive association
Cell-Free Phospholipid Biosynthesis by Gene-Encoded Enzymes Reconstituted in Liposomes
The goal of bottom-up synthetic biology culminates in the assembly of an entire cell from separate biological building blocks. One major challenge resides in the in vitro production and implementation of complex genetic and metabolic pathways that can support essential cellular functions. Here, we show that phospholipid biosynthesis, a multiple-step process involved in cell membrane homeostasis, can be reconstituted starting from the genes encoding for all necessary proteins. A total of eight E. coli enzymes for acyl transfer and headgroup modifications were produced in a cell-free gene expression system and were co-translationally reconstituted in liposomes. Acyl-coenzyme A and glycerol-3-phosphate were used as canonical precursors to generate a variety of important bacterial lipids. Moreover, this study demonstrates that two-step acyl transfer can occur from enzymes synthesized inside vesicles. Besides clear implications for growth and potentially division of a synthetic cell, we postulate that gene-based lipid biosynthesis can become instrumental for ex vivo and protein purification-free production of natural and non-natural lipids
A framework for evaluating the chemical knowledge and reasoning abilities of large language models against the expertise of chemists
Large language models LLMs have gained widespread interest owing to their ability to process human language and perform tasks on which they have not been explicitly trained. However, we possess only a limited systematic understanding of the chemical capabilities of LLMs, which would be required to improve models and mitigate potential harm. Here we introduce ChemBench, an automated framework for evaluating the chemical knowledge and reasoning abilities of state of the art LLMs against the expertise of chemists. We curated more than 2,700 question answer pairs, evaluated leading open and closed source LLMs and found that the best models, on average, outperformed the best human chemists in our study. However, the models struggle with some basic tasks and provide overconfident predictions. These findings reveal LLMs impressive chemical capabilities while emphasizing the need for further research to improve their safety and usefulness. They also suggest adapting chemistry education and show the value of benchmarking frameworks for evaluating LLMs in specific domain
