    Specific growth rate observer for the growing phase of a Polyhydroxybutyrate production process

    This paper focuses on the specific growth rate estimation problem in a Polyhydroxybutyrate bioplastic production process by industrial fermentation. The kinetics of the process are unknown and there are uncertainties in the model parameters and inputs. During the first hours of the growth phase of the process, biomass concentration can be measured online by an optical density sensor, but as cell density increases this method becomes ineffective and biomass measurement is lost. An asymptotic observer is developed to estimate the growth rate for the case without biomass measurement based on corrections made by a pH control loop. Furthermore, an exponential observer based on the biomass measurement is developed to estimate the growth rate during the first hours, which gives the initial condition to the asymptotic observer. Error bounds and robustness to uncertainties in the models and in the inputs are found. The estimation is independent of the kinetic models of the microorganism. The characteristic features of the observer are illustrated by numerical simulations and validated by experimental results

    Whole-cell biocatalysis in biphasic systems permits the synthesis of inhibiting chiral alcohols if appropriate non-water miscible ionic liquids are used. Taking the asymmetric reduction of 2-octanone to R-2-octanol by a recombinant Escherichia coli as a model reaction, a detailed characterisation of the biocatalytic reaction was performed with [HMPL][NTF] as ionic liquid. This made the asymmetric synthesis of R-2-octanol in a simple batch at a conversion > 99 % and at an enantiomeric excess > 99.5 % possible. The recyclability of the ionic liquid over a large number of cycles including the biotransformation, the separation of the ionic liquid, the product isolation, as well as the solvent’s subsequent reuse is of large importance for industrial applications. It could be demonstrated for the first time that the ionic liquid was recyclable over at least 25 process cycles without any consequences on the batch biotransformation.Ganzzell-Biokatalyse in Zwei-Phasen Systemen ermöglicht die Synthese von inhibierenden chiralen Alkoholen, wenn geeignete nicht mit Wasser mischbare Ionische Flüssigkeiten eingesetzt werden. Am Beispiel der asymmetrischen Reduktion von 2-Oktanon zu R-2-Oktanol durch rekombinante Escherichia coli erfolgte eine umfassende reaktionstechnische Charakterisierung der biokatalytischen Umsetzung mit [HMPL] [NTF] als ionische Flüssigkeit. Die asymmetrische Synthese von R-2-Oktanol konnte dadurch mit einem Umsatz > 99 % und einem Enantiomerenüberschuss > 99,5 % im einfachen Satzverfahren ermöglicht werden. Die für industrielle Anwendungen wichtige Wiederverwendbarkeit der ionischen Flüssigkeit über mindestens 25 Prozesszyklen bestehend aus Biotransformation, Abtrennung der ionischen Flüssigkeit, Produktisolierung und Rückführung der ionischen Flüssigkeit konnte ohne Rückwirkung auf das biokatalytische Satzverfahren erstmalig gezeigt werden

    Mit Hilfe der scaffolded DNA Origami Technologie können aus einzelsträngiger DNA (ssDNA) dreidimensionale Nanostrukturen gebildet werden. Zur skalierbaren Produktion von ssDNA wurden verschiedene Verfahren zur Herstellung des Bakteriophagen M13 mit geeigneten Escherichia coli Stämmen im Rührkesselreaktor untersucht. Höchste Konzentrationen von bis zu 590 mg L-1 aus Bakteriophagen isolierter ssDNA konnten durch Hochzelldichtekultivierung mit Escherichia coli im Zulaufverfahren erreicht werden.Scaffolded DNA origami enables the fabrication of three dimensional nanostructures using single-stranded DNA (ssDNA).     Anodenatmung könnte eine Möglichkeit darstellen, die Produktbildung bei Mikroorganismen gezielt zu verändern. Hierzu wurde ein Rührkesselreaktor entsprechend modifiziert, so dass ein ausreichender konvektiver Stofftransport bei der Mediator vermittelten Anodenatmung mit rekombinanten P. putida erreicht werden konnte. Durch Redoxpotentialregelung im Medium konnte damit die Produktion von para-Hydroxybenzoesäure aus Citronensäure mit P. putida um 70 % gegenüber der Sauerstoffatmung erhöht werden.Anodic respiration could represent a possibility to selectively affect product formation by microorganisms. A stirred-tank bioreactor was modified so that sufficient convective mass transport in mediated anodic respiration could be achieved with a recombinant P. putida. By controlling the redox potential in the medium the production of para-hydroxybenzoic acid from citric acid could be increased by 70 % compared to aerobic respiration

    Die mittlere Lichtverfügbarkeit wurde für ausgewählte Scenedesmus Stämme modellgestützt in Flachplattenphotobioreaktoren mit LED-Beleuchtung bestimmt und zur Identifikation geeigneter kinetischer Modelle des Licht abhängigen Wachstums dieser Mikroalgen genutzt. Durch Erhöhung der Eingangsphotonenflussdichten entsprechend des Algenwachstums ließen sich so optimale mittlere Lichtverfügbarkeiten im Reaktor länger aufrechterhalten und damit die Prozesszeiten im Satzverfahren deutlich verkürzen.The average availability of light was determined model-based for selected Scenedesmus spec. in flat-plate photobioreactors with a LED-lighting system in order to identify appropriate kinetic models for the light dependent growth of these microalgae. By increasing the incident photon flux density as a function of algal growth, optimal average light availabilities could be kept constant in the reactor resulting in significantly shortened process times of the batch processes

    Die zweistufige Reduktion von Dehydrocholsäure zu 12-Keto-Ursodesoxycholsäure ist der biokatalytische Schritt in der chemo-enzymatischen Synthese von Ursodesoxycholsäure. Hydroxysteroiddehydrogenasen exprimierende E. coli-Ganzzellbiokatalysatoren wurden für ein einfaches Eintopfverfahren entwickelt. Durch Modellierung wurde der Prozess optimiert. Im Litermaßstab konnten dadurch selbst bei geringen Biokatalysatormengen 28 g L-1 Dehydrocholsäure bei hohen Raum-Zeit-Ausbeuten von 120 g L-1 d-1 vollständig zum reinen chiralen Produkt umgewandelt werden.The two-step reduction of dehydrocholic acid to 12-keto-ursodeoxycholic acid is the biocatalytic step in the chemo-enzymatic synthesis of ursodeoxycholic acid. E. coli whole-cell biocatalysts expressing hydroxysteroid dehydrogenases were created to enable a simple biocatalytic one-pot process. The process was analyzed and optimized through modeling. When applying even low amounts of the whole cell biocatalyst on a liter-scale, 28 g L-1 dehydrocholic acid could be completely converted to the desired pure chiral product and high space-time yields of 120 g L-1 d-1 were achieved

    Spinnenseidenproteine lassen sich rekombinant mit Bakterien herstellen. Ziel dieser Arbeit war die reaktionstechnische Analyse der intrazellulären Herstellung eines hydrophoben Spinnenseidenproteins in Rührkesselreaktoren, um mögliche Wege zur Prozessintensivierung aufzuzeigen. Durch Minimierung von Einsatzstoffen, Vereinfachung der Substratdosierung und eine geeignete Induktionsstrategie konnten E. coli Zellen im Hochzelldichteprozess maximal mit Spinnenseidenprotein gefüllt und damit die Produktkonzentration im Reaktor mehr als verdoppelt werden.Spider silk proteins can be produced with recombinant bacteria. The aim of this thesis was the reaction engineering analysis of the intracellular production of a hydrophobic spider silk protein in stirred tank reactors to reveal possible ways for process intensification. Using minimized amounts of raw materials, a simplified substrate dosing and an appropriate induction strategy, the E. coli cells could be maximally filled with the spider silk protein applying a high cell density fermentation process. Thus, the product concentration in the reactor was more than doubled

    Metabolic Control Analysis of microbial fed-batch production of L-phenylalanine

    Um den Stoffwechsel rekombinanter E. coli-Zellen aus dem L-Phenylalanin-Produktionsprozess durch Zugabe unterschiedlicher Substrate anzuregen, wurden parallele Kurzzeitexperimente über 20 Minuten in Rührkesselreaktoren durchgeführt und die zelluläre Antwort über Stofffluss- und Metabolomanalysen charakterisiert. Mit Hilfe eines kinetischen Stoffwechselmodells konnte damit die metabolische Kontrolle der L-Phenylalanin-Produktion aus Glycerin zu verschiedenen Prozesszeitpunkten ermittelt werden.Metabolic disturbances were induced in recombinant E. coli cells producing L-phenylalanine by using different substrates in parallelized stationary short-term (20 minutes) perturbation experiments in stirred-tank bioreactors. Cellular response and deflection of metabolism were monitored by analyses of fluxome and metabolome. Identification of metabolic control of L-phenylalanine production with glycerol was enabled at several points of the production process by applying kinetic network models

    A bottleneck for the production of recombinant proteins in Escherichia coli is often the limited in vivo solubility. Refolding of the target protein becomes necessary and complicates downstream processing. Individual refolding conditions have to be determined empirically with a large experimental effort. Thus, a novel stochastic optimization strategy was developed on the basis of database information. This strategy allows an optimization of protein refolding conditions with a minimum number of experiments. The suitability of this new approach was demonstrated with 6 different proteins achieving an up to 30-fold increase of refolded activities compared to literature. In addition, it was shown that bagged decision trees were able to model the refolding conditions provided that the dataset of the optimization was large enough.Ein Problem bei der Herstellung von rekombinanten Proteinen in Escherichia coli ist oftmals deren begrenzte Löslichkeit in der Zelle. Dies macht eine Rückfaltung der Proteine bei der Aufarbeitung erforderlich. Individuelle Rückfaltungsbedingungen müssen hierzu aufwendig empirisch ermittelt werden. Daher wurde auf der Basis von Datenbankinformationen eine stochastische Versuchsplanungsstrategie erarbeitet, die eine Optimierung von Rückfaltungsbedingungen mit einem Minimum an Experimenten ermöglicht. Diese neue Optimierungsstrategie wurde erfolgreich für 6 unterschiedliche Proteine angewendet. Im Vergleich zur Literatur konnte hierbei eine bis zu 30-fache Verbesserung der Rückfaltungsaktivität erreicht werden. Darüber hinaus konnte gezeigt werden, dass eine Modellierung von Rückfaltungsbedingungen mit Hilfe von „bagged decision tree“ Ansätzen möglich ist, wenn genügend Datensätze aus der Optimierung zur Verfügung stehen

    One strategy to reduce the costs of the production of chemicals from renewable resources consists in combining fermentation and catalysis to avoid the expensive purification of the bio-derived intermediate. This process option was evaluated using succinic acid produced from fermentation as an example. For the aqueous hydrogenation of succinic acid, the use of different homogeneous and immobilized metal complexes as well as heterogeneous clay catalysts was analyzed. For the esterification of succinic acid, a reactive extraction process was investigated with different biological and chemical catalysts. After the optimization of the reaction conditions, succinic acid could be directly esterified in an aqueous fermentation broth with the homogeneous catalyst dodecylbenzenesulfonic acid with yields up to 94 % using 1-octanol. Finally, the produced diesters could be easily separated from the reaction system.Ein Ansatz zur Reduktion der Produktionskosten bei der Herstellung von Chemikalien aus nachwachsenden Rohstoffen ist die direkte Kombination von Fermentation und Katalyse ohne aufwändige Aufreinigung des biotechnologisch hergestellten Zwischenproduktes. Diese Option wurde am Beispiel von fermentativ hergestellter Bernsteinsäure untersucht. Für die wässrige Hydrierung von Bernsteinsäure wurden die Einsatzmöglichkeiten verschiedener homogener und immobilisierter Metallkomplexe, sowie heterogener Tonkatalysatoren analysiert. Für die Veresterung von Bernsteinsäure wurden Reaktivextraktionen mit verschiedenen biologischen und chemischen Katalysatoren reaktionstechnisch untersucht. Nach Optimierung der Reaktionsbedingungen konnte Bernsteinsäure mit dem Homogenkatalysator Dodecylbenzensulfonsäure direkt aus der wässrigen Fermentationslösung mit Ausbeuten von 94 % mit 1-Oktanol verestert und einfach abgetrennt werden
