PREVALENZA DEI PROCESSI METABOLICI NEL TRASPORTO DELLA RIBOFLAVINA IN ENTEROCITI ISOLATI DI RATTO
In isolated rat enterocytes, both normal (normoenergized) and rotenone-deenergized, riboflavin intracellular metabolic processes, operating in association with membrane specific transport mechanism, were investigated. The time course contents of unlabeled (endogenous) and labeled (exogenous) riboflavin compounds (riboflavin, RF; flavin mononucleotide, FMN; and flavin-adenine dinucleotide, FAD) were determined by a HPLC analytical procedure before and after incubation with tritium labeled riboflavin. In normoenergized enterocytes total labeled riboflavin content, which is the sum of riboflavin membrane transported and intracellular metabolized, steadily increased to a plateau up to 20 min incubation; FMN content reached a plateau after 3-20 min, while FAD and free RF increased constantly. The phosphorylated forms prevailed over the free RF form, being about 60% of total flavins. In deenergized enterocytes total riboflavin content was significantly lower than in normoenergized enterocytes and reached a plateau after only 3 min incubation; FMN and FAD contents were significantly lower than in normoenergized enterocytes and free RF represented the prevailing form (about 70% of total RF content). In both normoenergized and deenergized enterocytes the contents of unlabeled total RF, FMN and FAD significantly decreased after 20 min incubation, whereas free RF significantly increased only in normoenergized enterocytes

Preliminary analysis of indoor pollution from nitrogen dioxide in an area of Northern Italy
OBJECTIVE: The aims of this work are to describe the annual mean concentration of NO2 and to identify the home features and "habits" of its occupants related to NO2 levels. Both indoor and outdoor NO2 concentrations were measured in 114 dwellings in Pavia (Northern Italy).
DESIGN: The study is in the frame of European Community Respiratory Health Survey II. Data were collected in the period 2001-2002 during home visits by trained fieldworkers.
PARTICIPANTS: 116 subjects gave the consent to home visits.
MAIN OUTCOME MEASURE: NO2 concentration measured using passive samplers.RESULTS: Indoor NO2 concentration is in average 47.1 microg/m3 (+/- 24.5 microg/m3) and outdoor NO2 concentration is 44.3 microg/m3 (+/- 14.5 microg/m3). The gas oven is the only domestic gas appliance associate with high levels of NO2 when measuring in homes. The outdoor pollution affects NO2 indoor concentrations and contribute to store NO2 in homes.
CONCLUSION: the study is the first step in the evaluation of NO2 exposure effect on respiratory health

Microvillous membrane vesicles prepare from the cortex of rats, were suspended in different media, controlling the composition of the intravesicular fluid by prolonged equilibration at 4°C in preincubation buffers of desired composition. Until used they were held on ice. For the uptake experiments the vesicles were incubated at 25°C with tritiated thiamin for suitable times. Then, they were separated with a rapid filtration procedure for the radiometric measusement of the radioactivity taken up. Thiamin uptake was Na+-independent and 53% membrane bound. The concentration curve had a biphasic course which was hyperbolic (saturable) at physiological concentrations (<1.25 uM) and linear at higher ones. Thiamin uptake was three times increased by an outwardly directed H+ gradient (pHin 6 : pHout 7.5), which enhanced only thiamin translocation and maintained biphasic the concentration curve. However, the saturable component displayed kinetic costants 12-fold higher than those in the absence of gradient. Moreover, the saturable component remained unmodified by typical organic cations transported by renal microvillous vesicles, but inhibited by inhibitors of intestinal thiamin/H+, renal guanidine/H+ and Na+/H+ antiporters
