No full textIl problema della contaminazione degli alimenti liquidi con fotoiniziatori, una classe di molecole utilizzate nella fissazione degli inchiostri-UV sui cartoni di materiale poliaccoppiato per imballaggio, ha avuto origine nel novembre 2005, quando il ritrovamento del fotoiniziatore ITX (2-isopropil thioxanthone) in alcuni campioni di latte per l’infanzia causo’ il ritiro dal mercato di una quantità pari a circa 30 milioni di litri. Successivamente, l’EFSA (Autorità Europea per la Sicurezza Alimentare) si pronuncio’ affermando che, nonostante in alcuni campioni di latte e succhi di frutta erano stati rinvenuti livelli di ITX compresi tra 27 e 440 g l-1 (latte) e tra 5 e 249 g l-1 (succhi di frutta), la presenza di tale contaminante negli alimenti non costituiva un pericolo per la salute del consumatore, data la bassa tossicità della molecola [1]. Tuttavia, in letteratura a tutt’oggi non sono disponibili convincenti dati che possano escludere la genotossicità della molecola [2]. In questi ultimi due anni sono stati pubblicati vari lavori che propongono differenti metodi analitici per monitorare l’ITX nel packaging e nei liquidi alimentari in esso contenuti: si va dall’estrazione in fase solida (SPE) all’estrazione liquida pressurizzata (PLE) [3], dall’HPLC-DAD-FLD alla LC/MS e alla GC/MS [4].
Lo scopo di questo lavoro è stato quello di sviluppare un nuovo metodo analitico che permetta l’analisi contemporanea di 5 fotoiniziatori (ITX, EHDAB, Benzofenone, IRGACURE 184 ed EDAB) nel packaging e nei relativi alimenti liquidi contenuti, mediante l’utilizzo della GC/MS e dell’HPLC/MS/MS. Il metodo sviluppato è stato applicato all’analisi di 40 campioni di latte, succhi di frutta e vino contenuti in cartoni di materiale poliaccoppiato. Il benzofenone è stato trovato in tutti i cartoni e in tutti gli alimenti analizzati, in un range di concentrazione che va da 0.2 a 387 g dm-2 (cartoni) e da 5 a 217 g l-1 (alimenti). Meno estesa è risultata la contaminazione da ITX, rinvenuto in 9 cartoni (0.01-0.08 g dm-2) e in soli 3 alimenti (0.2-0.24 g l-1). Tutti i risultati relativi allo screening dei 40 campioni, ottenuti mediante analisi in GC/MS sono stati confermati in HPLC/MS/MS (ion trap)

Improving MHC-I ligand identifications from LC-MS/MS data by incorporating allelic peptide motifs
No full textMHC class I (MHC-I)-bound ligands play a pivotal role in CD8 T cell immunity and are hence of major interest in understanding and designing immunotherapies. One of the most commonly utilized approaches for detecting MHC ligands is LC-MS/MS. Unfortunately, the effectiveness of current algorithms to identify MHC ligands from LC-MS/MS data is limited because the search algorithms used were originally developed for proteomics approaches detecting tryptic peptides. Consequently, the analysis often results in inflated false discovery rate (FDR) statistics and an overall decrease in the number of peptides that pass FDR filters. Andreatta et al. describe a new scoring tool (MS-rescue) for peptides from MHC-I immunopeptidome datasets. MS-rescue incorporates the existence of MHC-I peptide motifs to rescore peptides from ligandome data. The tool is demonstrated here using peptides assigned from LC-MS/MS data with PEAKs software but can be deployed on data from any search algorithm. This new approach increased the number of peptides identified by up to 20-30% and promises to aid the discovery of novel MHC-I ligands with immunotherapeutic potential
