Widespread occurrence of an emerging fungal pathogen in heavily traded Chinese urodelan species
Understanding introduction routes for wildlife pathogens is vital for the development of threat abatement plans. The chytrid fungus Batrachochytrium salamandrivorans (Bsal) has recently emerged in Europe, where it is considered to be a serious threat for urodelan conservation. If the highly diverse Chinese urodelans were to constitute a Bsal reservoir, then the significant international trade in these species may vector Bsal into naïve urodelan communities. Here, we analyzed a total of 1,143 samples, representing 36 Chinese salamander species from 51 localities across southern China for the presence of Bsal. We found Bsal was present across a wide taxonomic, geographical, and environmental range. In particular, Bsal DNA was detected in 33 samples from the genera Cynops, Pachytriton, Paramesotriton, Tylototriton, and Andrias, including the heavily traded species Paramesotriton hongkongensis and Cynops orientalis. The true Bsal prevalence across our data set was estimated between 2% and 4%, with a maximum of 50% in a population of P. hongkongensis. Even at this overall relatively low Bsal prevalence, the exportation of millions of animals renders Bsal introduction in naïve, importing countries a near certainty, which calls for the urgent implementation of proper biosecurity in the international wildlife trade

An Adaptive Unscented Kalman Filter for the Estimation of the Vehicle Velocity Components, Slip Angles, and Slip Ratios in Extreme Driving Manoeuvres
This paper presents a novel unscented Kalman filter (UKF) implementation with adaptive covariance matrices (ACMs), to accurately estimate the longitudinal and lateral components of vehicle velocity, and thus the sideslip angle, tire slip angles, and tire slip ratios, also in extreme driving conditions, including tyre-road friction variations. The adaptation strategies are implemented on both the process noise and measurement noise covariances. The resulting UKF ACM is compared against a well-tuned baseline UKF with fixed covariances. Experimental test results in high tyre-road friction conditions show the good performance of both filters, with only a very marginal benefit of the ACM version. However, the simulated extreme tests in variable and low-friction conditions highlight the superior performance and robustness provided by the adaptation mechanism

Spatiotemporal evolution of early innate immune responses triggered by neural stem cell grafting
INTRODUCTION: Transplantation of neural stem cells (NSCs) is increasingly suggested to become part of future therapeutic approaches to improve functional outcome of various central nervous system disorders. However, recently it has become clear that only a small fraction of grafted NSCs display long-term survival in the (injured) adult mouse brain. Given the clinical invasiveness of NSC grafting into brain tissue, profound characterisation and understanding of early post-transplantation events is imperative to claim safety and efficacy of cell-based interventions.
METHODS: Here, we applied in vivo bioluminescence imaging (BLI) and post-mortem quantitative histological analysis to determine the localisation and survival of grafted NSCs at early time points post-transplantation.
RESULTS: An initial dramatic cell loss (up to 80% of grafted cells) due to apoptosis could be observed within the first 24 hours post-implantation, coinciding with a highly hypoxic NSC graft environment. Subsequently, strong spatiotemporal microglial and astroglial cell responses were initiated, which stabilised by day 5 post-implantation and remained present during the whole observation period. Moreover, the increase in astrocyte density was associated with a high degree of astroglial scarring within and surrounding the graft site. During the two-week follow up in this study, the NSC graft site underwent extensive remodelling with NSC graft survival further declining to around 1% of the initial number of grafted cells.
CONCLUSIONS: The present study quantitatively describes the early post-transplantation events following NSC grafting in the adult mouse brain and warrants that such intervention is directly associated with a high degree of cell loss, subsequently followed by strong glial cell responses
invloedrijke schilderijen ooit gemaakt.
meest uiteenlopende disciplines.
Nailfold capillaroscopy for day-to-day clinical use: construction of a simple scoring modality as a clinical prognostic index for digital trophic lesions
Objective Construction of a simple nailfold videocapillaroscopic (NVC) scoring modality as a prognostic index for digital trophic lesions for day-to-day clinical use.
Methods An association with a single simple (semi)quantitatively scored NVC parameter, mean score of capillary loss, was explored in 71 consecutive patients with systemic sclerosis (SSc), and reliable reduction in the number of investigated fields (F32-F16-F8-F4). The cut-off value of the prognostic index (mean score of capillary loss calculated over a reduced number of fields) for present/future digital trophic lesions was selected by receiver operating curve (ROC) analysis.
Results Reduction in the number of fields for mean score of capillary loss was reliable from F32 to F8 (intraclass correlation coefficient of F16/F32: 0.97; F8/F32: 0.90). Based on ROC analysis, a prognostic index (mean score of capillary loss as calculated over F8) with a cut-off value of 1.67 is proposed. This value has a sensitivity of 72.22/70.00, specificity of 70.59/69.77, positive likelihood ratio of 2.46/2.32 and a negative likelihood ratio of 0.39/0.43 for present/future digital trophic lesions.
Conclusions A simple prognostic index for digital trophic lesions for daily use in SSc clinics is proposed, limited to the mean score of capillary loss as calculated over eight fields (8 fingers, 1 field per finger)
Diagnosis of complement alternative pathway disorders
Kidney diseases resulting from abnormal control of the complement alternative pathway include atypical hemolytic uremic syndrome, C3 glomerulonephritis, and dense-deposit disease, as well as atypical postinfectious glomerulonephritis. Although clinically diverse, they all result from loss of surface or fluid-phase complement control, caused by acquired or genetic defects in the complement alternative pathway. As such, the diagnostic approach is similar and includes a comprehensive biochemical, genetic, and pathologic analysis of the complement pathway. The biochemical test battery includes functional activity measurements of the entire complement pathway, functional and quantitative analysis of individual components and regulators, and quantification of activation products. In patients with a thrombotic microangiopathy, ADAMTS-13 activity should be determined to exclude a thrombotic thrombocytopenic purpura. The spectrum of genes currently known to be involved in the pathogenesis of alternative pathway disorders is rapidly expanding. Pathologic analysis of a kidney biopsy specimen is sophisticated with ad hoc immunofluorescence studies and laser microdissection with mass spectrometry. The identification of the underlying defect in the alternative pathway based on this comprehensive analysis will allow treatment to be directed to the site of dysregulation. © 2015 International Society of Nephrology
A retrospective, multicentre study on the efficacy, durability, and tolerability of bictegravir/emtricitabine/tenofovir alafenamide (BIC/FTC/TAF) for the treatment of HIV in a real-world setting in Belgium
Conclusion • Several RCTs have demonstrated BIC/FTC/TAF to be a first-line option for treatment-naïve and experienced PLWH [1-4]. • Real-world studies provide complementary information to RCTs and ensure that those results can be generalized to broader populations seen in daily practice. • The aim of this study was to describe the Belgian HIV population treated with BIC/FTC/TAF and to evaluate its efficacy, durability, and tolerability in a real-world setting. We would like to thank everyone who contributed to this study including the study participants and investigators. This study was supported by a grant from Gilead International
