No full textTussentaal is een overbodige, negatieve, tendentieuze term met een willekeurige inhoud. Zoveel blijkt uit de vakliteratuur, maar ook uit de Vlaamse schoolboeken Nederlands voor het secundair onderwijs. Wie bovendien discussieert over tussentaal, discussieert ook over standaardtaal. De begrenzing van het ene begrip impliceert de begrenzing van het andere. Op de vraag wat nu in Vlaanderen tot die standaardtaal behoort, is het finale antwoord nog lang niet gegeven. Het Vlaamse taalbeleid is bijzonder versnipperd, met normenchaos als gevolg. Er is dan ook nood aan een beleid dat de taalgebruiker centraal stelt, zoals dat door de Taalunie in feite nu al wordt gepropageerd: een niet-normatief geïnspireerd beleid dat de gelijkwaardigheid van de Nederlandse en de Vlaamse taalgemeenschap erkent en het inzicht in en een positieve attitude tegenover taalvariatie bevordert. Met het oog daarop worden in het tweede deel voorgesteld: een algemene aanvaarding van de Vlaamse taalemancipatie, de taalmengtafel als een model voor positief en realistisch denken over taalvariatie, en een minimaal concept van standaardtaal.status: Publishe

Op 3 mei 2022 publiceerden we een Story @ UGent @ Work omtrent inactiviteit (i.e. personen die noch werken, noch werk zoeken) in België, op basis van de nieuwste Eurostat-cijfers. Dit kreeg toen brede aandacht in de media en in de communicatie van verschillende beleidsmensen, zoals parlementairen, partijvoorzitters en regeringsleden.
Een vraag die bleef terugkeren, was hoe de hoge inactiviteit onder personen met een migratieachtergrond te verklaren. Hoeveel procent van de Belgische inactiviteit maken deze migranten precies uit? Wat betekent dit in absolute aantallen? Hoe zit het met de verdeling naar geslacht en naar de Belgische gewesten? Maar ook: wat zijn de laatste cijfers van het opleidingsniveau van deze migranten, i.e. dé voorspeller van arbeidsmarktkansen, in vergelijking met het buitenland?

An estimation of lameness in Sable Island horses using radiographic evaluation of the distal phalanx and hoof capsule
No full textLaminitis is a painful condition that causes lameness in horses. However, monitoring this condition in feral horses is logistically difficult. Laminitis can be detected postmortem, as inflammation of the sensitive laminae of the hoof changes the relative location of the bony structures within the hoof capsule. Thus, evaluation of cadavers may be used to estimate laminitis prevalence in feral populations of horses. The objective of this study was to estimate the prevalence of laminitis in feral horses inhabiting the Sable Island National Park Reserve, Canada, using radiographic imagery on the hooves of naturally deceased horses. Radiographic indicators evaluated included coronary band extensor process distance (CE), capsular rotation (CR), palmar angle (PA), ratio of the dorsal hoof wall thickness to the length of the distal phalanx (DHWT:P3) and sole depth (SLD). Each value measured indicated the following prevalence of laminitis: CE 0% (97.5% Confidence Interval (CI) 0–4.51%), 8.75% CR (95%CI 3.59–17.2%), PA 25% (95% CI 15.99–35.94%), DHWT:P3 6.25% (95%CI 2.06–14.0%) and 65% SLD (95% CI 53.52–75.33%). The majority of indices measured for laminitis were low. These findings suggest that laminitis is fairly infrequent in the Sable Island horses evaluated in this study

Intravesicular Genomic DNA Enriched by Size Exclusion Chromatography Can Enhance Lung Cancer Oncogene Mutation Detection Sensitivity
No full textExtracellular vesicles (EVs) are cell-derived structures surrounded by a lipid bilayer that carry RNA and DNA as potential templates for molecular diagnostics, e.g., in cancer genotyping. While it has been established that DNA templates appear on the outside of EVs, no consensus exists on which nucleic acid species inside small EVs (<200 nm, sEVs) are sufficiently abundant and accessible for developing genotyping protocols. We investigated this by extracting total intravesicular nucleic acid content from sEVs isolated from the conditioned cell medium of the human NCI-H1975 cell line containing the epidermal growth factor (EGFR) gene mutation T790M as a model system for non-small cell lung cancer. We observed that mainly short genomic DNA (<35–100 bp) present in the sEVs served as a template. Using qEV size exclusion chromatography (SEC), significantly lower yield and higher purity of isolated sEV fractions were obtained as compared to exoEasy membrane affinity purification and ultracentrifugation. Nevertheless, we detected the EGFR T790M mutation in the sEVs’ lumen with similar sensitivity using digital PCR. When applying SEC-based sEV separation prior to cell-free DNA extraction on spiked human plasma samples, we found significantly higher mutant allele frequencies as compared to standard cell-free DNA extraction, which in part was due to co-purification of circulating tumor DNA. We conclude that intravesicular genomic DNA can be exploited next to ctDNA to enhance EGFR T790M mutation detection sensitivity by adding a fast and easy-to-use sEV separation method, such as SEC, upstream of standard clinical cell-free DNA workflows
