    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Promoter elements for gene therapy of regenerating hepatic tissue.

    We have identified and characterized two transcriptional modules derived from the promoters of liver genes whose expression is specific for proliferating hepatocytes: A) An enhancer module from the HNF1β promoter. HNF1α and β are two trans-acting factors that play a central role in the expression of liver-specific genes. We reported that, during mouse and rat development, HNF1β expression precedes that of HNF1α and is inducible by retinoic acid treatment. Moreover HNF1β (but not HNF1α) is expressed in undifferentiated hepatocytes. A combined strategy of deletions and site-specific mutagenesis allowed us to identify the cis-acting elements of the HNF1β promoter. In our transient expression assay, the -435/+33 region of the promoter is sufficient to reproduce the expression pattern of the endogenous gene. This region can be further subdivided into a tissue-specific enhancer and a proximal promoter element. The transcriptional activity of the enhancer element, as measured by co-transfection assays and band-shift experiments, depends on the binding of two transcriptional activators: the liver-specific HNF-4 factor (-255/-235), and the RARα/RXRα heterodimeric protein, that recognize a RARE element partially overlapping with the HNF-4 binding site. B) The proximal element of the alpha-foetoprotein promoter. The alpha-fetoprotein (AFP) gene is expressed in the prenatal life in the yolk sac and in the foetal liver. At birth, the AFP gene is "shut down" at the transcriptional level, and it can be re-expressed in the adult liver only in pathologic conditions, like hepatic parenchyma regeneration or liver tumours. Therefore, the AFP gene must contain cis acting regulatory elements able to activate transcription only in proliferating hepatocytes. We have isolated and individually analysed the key elements of the AFP transcriptional regulation located in the proximal promoter region, where two partially overlapping AP1 and GRE elements dictate the activation or repression of the gene, respectively. Our results suggest that, while two adjacent HNF1α/β binding sites restrict the AFP promoter activity to liver cell, changes in the GR/AP1 ratio can account for the AFP reactivation in the regenerating hepatocyte. We have constructed a series of artificial promoters by assembling different combinations of these transcriptional modules. The most efficient combinations have been used to construct SV40-derived episomal vectors, that we plan to use for gene therapy of proliferating hepatocytes.     Jersey Homesteads (later the Borough of Roosevelt) was established in the 1930s as an agro-industrial cooperative community. It was established specifically for urban Jewish garment workers, many of whom had emigrated from Europe. President John F. Kennedy sent a telegram to the citizens of Roosevelt, New Jersey, apologizing for not being able to attend the memorial dedication in honor of former President Franklin Delano Roosevelt. (Jersey Homesteads became Roosevelt in 1945 in honor of the president.) President Kennedy expressed his gratitude to the people of Roosevelt for constructing the memorial, and commented that it will serve as a constant reminder of Roosevelt's good works

    The excavation of the Gallo-Roman sanctuary of Les Bouchauds, in the commune of Saint-Cybardeaux (Charente), executed in 1974-75, 1978-80 and 1986-88, has shown the existence of two sacred precincts : the widest, on the west, is enclosing two squre fana with galleries meanwhile an octogonal temple and a rectangular temple with pronaos are in the eastern and smaller precinct. The construction of this complex has known two periods : the former is very likely dating from the Flavians ; the second one is undoubtedly the end of the lid century, of which specially architectural ornaments, studied by D. Tardy (annexe 1), give evidence. An earliest frequenting of the site during the Augustean period is revealed by an important lot of Gallic and Roman coins, investigated by P. Tronche (annexe 2). Situated on the top of a hill, the sanctuary is flanked, on its northern side, by one of the greatest theaters of Gaul, already studied by M. Fincker and F. Thierry (Aquitania , 3, 1985) and probably linked to the Imperial cult ; and in the other hand, at the bottom of the hill, in southern part, a small town" — which was conjectured because of finds at the beginning of the century and in the seventies — is now attested by J. Dassié's photographies (annexe 3).La fouille du sanctuaire gallo-romain des Bouchauds, commune de Saint-Cybardeaux (Charente) effectuée en 1974-75, 1978-80, 1986-88, a révélé l'existence de deux enceintes sacrées ; la plus vaste, à l'ouest, enferme deux fana carrés avec galeries ; l'enceinte orientale, plus restreinte, abrite un temple octogonal et un temple rectangulaire avec pronaos. La construction de cet ensemble a connu deux époques : la première remonte vraisemblablement aux Flaviens ; la seconde est assurément datée de la fin du Ile siècle, comme en témoignent en particulier les décors d'architecture étudiés par D. Tardy (annexe 1). Une première fréquentation du site à l'époque augustéenne est révélée par un important lot de monnaies gauloises et romaines, analysé par P. Tronche (annexe 2).     Logarithmic variance profiles and the corresponding f-1 spectra of temperature fluctuations in turbulent Rayleigh-Bénard convection

    We report experimental results for the temperature variance 2(z) and the corresponding frequency spectra P(f) in turbulent Rayleigh-Bénard convection (RBC) in a cylindrical sample of aspect ratioT= D/L = 1:00 (D = 1:12 m is the diameter and L = 1:12 m the height). The measurements were conducted in the Rayleigh-number range 1011 < Ra < 1:35 1014 and Pr ' 0:8. For Ra = 1:35x1014, 2(z) could be described well by a logarithmic dependence on the vertical position z in a range of z 1 < z < z 2 with z 1 ' 70 and z 2 = 0:1L. Here L=(2Nu) is the thickness of a thin thermal sublayer adjacent to the horizontal plate where the heat flux (denoted by the Nusselt number Nu) is carried mostly by thermal diffusion. In the log layer, we found that the temperature spectra had a significant frequency range over which P(f) f with close to 1. As Ra decreased, increased so that the log layer became thinner. At Ra = 2:05 1011, z 2 < z 1 and therefore there was no range for a log layer. Correspondingly, the temperature spectrum near the horizontal plate did not have the f1 scaling form either
