    The production of polyphonic manuscripts in thirteenth-century Paris: new evidence for standardised procedures

    Modern understanding of the production and dissemination of thirteenth-century polyphony is constrained by the paucity of manuscript sources that have been preserved in their entirety; the panorama of sources of medieval polyphony is essentially fragmentary. Some of the surviving fragments, however, were torn from lost books of polyphony that were to some extent comparable to well-known extant codices. The fragment of polyphony preserved in the binding of manuscript 6528 of the Biblioteca Nacional de Madrid is illustrative in this respect. This fragment displays a number of codicological and musical features that are strikingly similar to those of the Florence manuscript (F).     Genome-wide transcriptome study in skin biopsies reveals an association of E2F4 with cadasil and cognitive impairment

    Demència; Mal de cap; IctusDemencia; Dolor de cabeza; IctusDementia; Headache; StrokeCADASIL is a small vessel disease caused by mutations in NOTCH3 that lead to an odd number of cysteines in the EGF-like repeat domain, causing protein misfolding and aggregation. The main symptoms are migraine, psychiatric disturbances, recurrent strokes and dementia, being executive function characteristically impaired. The molecular pathways altered by this receptor aggregation need to be studied further. A genome-wide transcriptome study (four cases paired with three healthy siblings) was carried out, in addition to a qRT-PCR for validation purposes (ten new cases and eight new controls). To study the expression profile by cell type of the significant mRNAs found, we performed an in situ hybridization (ISH) (nine cases and eight controls) and a research in the Single-nuclei Brain RNA-seq expression browser (SNBREB). Pathway analysis enrichment was carried out with Gene Ontology and Reactome. Neuropsychological tests were performed in five of the qRT-PCR cases. The two most significant differentially expressed mRNAs (BANP, p-value = 7.23 × 10–4 and PDCD6IP, p-value = 8.36 × 10–4) were selected for the validation study by qRT-PCR. Additionally, we selected two more mRNAs (CAMK2G, p-value = 4.52 × 10–3 and E2F4, p-value = 4.77 × 10–3) due to their association with ischemic neuronal death. E2F4 showed differential expression in the genome-wide transcriptome study and in the qRT-PCR (p = 1.23 × 10–3), and it was upregulated in CADASIL cases. Furthermore, higher E2F4 expression was associated with worse executive function (p = 2.04 × 10–2) and attention and information processing speed (IPS) (p = 8.73 × 10–2). In situ hibridization showed E2F4 expression in endothelial and vascular smooth vessel cells. In silico studies indicated that E2F4 is also expressed in brain endothelial cells. Among the most significant pathways analyzed, there was an enrichment of vascular development, cell adhesion and vesicular machinery terms and autophagy process. E2F4 is more highly expressed in the skin biopsy of CADASIL patients compared to controls, and its expression is present in endothelial cells and VSMCs. En lo que sigue nos serviremos de este prefacio para tomar de allí los principales ejes temáticos con los que discuten críticamente los artículos que conforman esta edición.

    [ES] La corrosión de las armaduras es una de las principales razones de la reducción de la durabilidad de las estructuras de hormigón armado. Por otra parte, existen diversos mecanismos de corrosión que dan lugar a la generación de diferentes defectos con geometrías diferentes. El presente estudio se centra en evaluar el comportamiento mecánico de armaduras con diferentes tipos de defectos, desde corrosión generalizada, por picaduras hasta corrosión bajo tensión y fragilización por hidrógeno. Los resultados obtenidos muestran que la resistencia de los aceros no depende únicamente de la pérdida de material, sino que es muy importante evaluar la geometría de los defectos. Estos resultados permiten explicar la variación en el comportamiento mecánico de armaduras corroídas por diferentes métodos.[EN] Corrosion of reinforcement is one of the main reasons for reducing the durability of structures. Corrosion results in the appearance of different defects with different geometries. This study focuses on evaluating the mechanical behaviour of rebars with different types of defects, from generalised corrosion, pitting to stress corrosion and hydrogen embrittlement.     Long-read sequencing reveals increased isoform diversity in key transcription factor effectors of intercellular signalling at the invertebrate-vertebrate transition

    Several intercellular signalling pathways (namely wingless - Wnt, Hedgehog - Hh, and Bone Morphogenetic Protein - BMP) are used repeatedly in animals throughout development and evolution, and are also frequent targets for disease-associated disruptions. We have previously shown that the major transcriptional effectors of βcatenin-dependent Wnt signalling, the TCF/LEF proteins, in contrast to other pathway components, have a higher gene number and isoform diversity in vertebrates versus invertebrates, but this increased diversity has only been poorly quantified. Considering that isoform diversity correlates with organism complexity, any increase in major signalling effectors is likely to have made a significant contribution to vertebrate evolution. Using de novo long-read transcriptomes, we compared isoform number per gene for the chordates Ciona intestinalis, Lampetra planeri and Xenopus tropicalis, thus encompassing the invertebrate sister group to vertebrates, as well as a cyclostome and a gnathostome vertebrate. Our results implicate an increase in isoform diversity of the transcription factors of major intercellular signalling pathways as having a disproportionate role in the evolutionary origin and diversification of vertebrates
