Ulusal Tasarım Kongresi İstanbul, 8 - 9 Ekim 2009.“Kriz”, Türk Dil Kurumu sözlüğünde bunalım anlamına gelmesine rağmen Çince’deki karşılığı “tehlike” ve “fırsat” olarak tanımlanmaktadır (TDK, 2009; Keown-McMullan, 1997). Birbirine zıt görünen bu iki kelime aslında olaylarla birlikte düşünüldüğünde birbirinden türeyebilecek kavramlar olarak da görülebilir. Tarih boyunca pek çok krize rastlanmış olmasına rağmen akılda en kalıcı olanı 1929’daki “Kara Perşembe” adıyla da anılan Büyük Buhran olmuştur. Bunun sebebi Büyük Buhran’ın uzun yıllar boyunca birçok ülkede etkisinin hissedilmiş olmasıdır. Bugün de yine bir kriz dönemi içinde bulunmaktayız, bu durumu televizyon programlarından ve reklamlardan dahi gözlemlemek mümkündür. Özellikle “Krizdeyiz” gibi düşük bütçeyle evde yemek yapmayı teşvik eden ya da Singer’in sponsorluğunu yaptığı “Deryalı Günler” gibi evde dikiş dikmeyi, kullanılmayan malzemeleri değerlendirmeyi öğreten programlar bunlara birkaç örnektir. Bu çalışmada krizde evkadınlarının sosyal hayatlarındaki değişikliğe yön vermek amacıyla yapılan programlardan yola çıkılarak, piyasada bir zamanlar zirvedeyken kaybolan ve günümüzde tekrar gündeme gelen Singer dikiş makineleri incelenecektir. Bu araştırmada dikiş makinesinin tarihi, kadınlar için önemi ve Singer’in hayatımıza belirli dönemlerde nasıl girdiği ele alınacaktır

    Effector Th1 cells under <scp>PD</scp> ‐1 and <scp>CTLA</scp> ‐4 checkpoint blockade abrogate the upregulation of multiple inhibitory receptors and by‐pass exhaustion

    Immune checkpoint inhibitor (ICI) immunotherapy relies on the restoration of T-cell functions. The ICI receptors are not only found on exhausted T cells but also upregulated upon activation and reach high levels on effector T cells. In an ex vivo model, this study explored the consequences of PD-1 and cytotoxic T-lymphocyte antigen (CTLA-4) blockade applied during specific time frames of T-cell stimulation that coincide with distinct functional phases in type 1 helper T (Th1) cells. When applied at an early stimulation stage, the checkpoint blockade interfered with the upregulation of multiple inhibitory receptors such as PD-1, LAG3, TIM-3 and CTLA-4. Moreover, extension of the blockade period restricted the hyporesponsiveness in T cells. Alternatively, a short-term ICI treatment was advantageous when applied at late time frames of Th1 cell stimulation. Here, a transition phase from effector to exhausted state, which coincided with the late time frames of Th1 stimulation, was clearly determined together with the transcriptomics data demonstrating the initiation of significant alterations in metabolic pathways, genetic information processes, effector and exhaustion specific pathways. Applied in this transition phase, PD-1 and/or CTLA-4 blockade downregulated the inhibitory receptors which were already present on the effector Th1 cells, potentially through endocytic pathways. Therefore, the efficacy of ICI therapy was modulated by the functional status of T cells and can be improved by modifying the timing and duration of PD-1 and CTLA-4 blockade. In conclusion, the ICI therapy not only supports the reactivation of T cells but can also constrain de novo exhaustion

    Retina and Optic Disc Characteristics in Amblyopic and Non-amblyopic Eyes of Patients with Myopic or Hyperopic Anisometropia

    Objectives: To compare retinal and optic disc characteristics between amblyopic and non-amblyopic eyes in patients with myopic and hyperopic anisometropia measured with optical coherence tomography (OCT). Materials and Methods: Patients with myopic (25 patients: 17 female, 8 male; median age 27 years, range 16-40 years) and hyperopic (31 patients: 19 female, 12 male; median age 20 years, range 13-41 years) anisometropic amblyopia were included. Eye examination included determination of best-corrected visual acuity (BCVA) with a Snellen chart, measurement of manifest and cycloplegic refraction after pupillary dilation, alternate cover testing, globe movement evaluation, A-scan biometry for axial length, biomicroscopy, fundus examination, and OCT scanning. Main outcome measures were spherical equivalence, BCVA, axial length, retinal nerve fiber layer (RNFL) thickness, macular thickness, macular volume, and optic disc area. Results: In both myopic and hyperopic patients, the absolute value of the mean spherical equivalence was significantly greater in the amblyopic than non-amblyopic eyes, and the mean BCVA was significantly less in the amblyopic than the non-amblyopic eyes. In both myopic and hyperopic patients, there were no significant differences in mean RNFL thickness, macular thickness, macular volume, axial length, or optic disc area between amblyopic and non-amblyopic eyes. Conclusion: The amblyopic process may have no significant effect on the RNFL, macula, or optic disc. Further studies with more patients, including postmortem studies, may clarify the retinal, histopathologic, and anatomic differences between amblyopic and non-amblyopic eyes
