No full textAcanthamoeba spp. are free-living amoebae that are ubiquitous in natural environments. They can cause cutaneous, nasopharyngeal, and disseminated infection, leading to granulomatous amebic encephalitis (GAE) in immunocompromised individuals. In addition, they can cause amoebic keratitis in contact lens wearers. Acanthamoeba GAE is almost always fatal because of difficulty and delay in diagnosis and lack of optimal antimicrobial therapy. Here, we report the description of an unusual strain isolated from skin and brain of a GAE patient. The amoebae displayed large trophozoites and star-shaped cysts, characteristics for acanthamoebas belonging to morphology Group 1. However, its unique morphology and growth characteristics differentiated this new strain from other Group 1 species. DNA sequence analysis, secondary structure prediction, and phylogenetic analysis of the 18S rRNA gene confirmed that this new strain belonged to Group 1, but that it was distinct from the other sequence types within that group. Thus, we hereby propose the establishment of a new species, Acanthamoeba byersi n. sp. as well as a new sequence type, T18, for this new strain. No full textVe své diplomové práci se autor zabývá institutem přechodu práv a povinností z pracovněprávních vztahů. Úprava přechodu práv a povinností je v českém právním řádu upravena z podstatné části zákonem č. 262/2006 Sb., zákoník práce a dále v dalších právních předpisech. Úprava v českém právním řádu vychází z harmonizované úpravy na úrovni práva Evropské unie, která je zakotvena ve směrnici Rady 2001/23/ES ze dne 12. března 2001 o sbližování právních předpisů členských států týkajících se zachování práv zaměstnanců v případě převodů podniků, závodů nebo částí podniků nebo závodů. Ve své práci autor zkoumá současnou úpravu v českém právním řádu a rozhodovací praxí českých soudů a srovnává je zejm. s rozhodovací praxí Soudního dvora Evropské unie. Autor se také zabývá změnami úpravy přechodu práv a povinností provedenými novelou č. 285/2020 Sb., kterou se mění zákon č. 262/2006 Sb., zákoník práce, ve znění pozdějších předpisů a některé další související zákony, a porovnává platné znění s předcházejícím. Diplomová práce se dále věnuje historii úpravy přechodu práv a povinností na území České republiky, následně se zabývá obecným vymezením pracovněprávních vztahů. Práce popisuje úpravu přechodu práv a povinností v právu Evropské unie a rozebírá koncept hospodářské jednotky tak, jak tuto definuje Soudní dvůr...In his thesis, the author deals with the transfer of rights and obligations arising from employment relationships. In the Czech law, the regulation of transfer of rights and obligations is governed in substantial part by the Act No. 262/2006 Coll., the Labour Code, as amended, and in other statutes. The Czech legislation is based on harmonised regulation in the law of the European Union, which is provided for in the Council Directive 2001/23/EC of 12 March 2001 on the approximation of the laws of the Member States relating to the safeguarding of employees' rights in the event of transfers of undertakings, businesses or parts of undertakings or businesses. In his thesis, the author examines the current regulation in the Czech law and the decision- making practice of the Czech courts and compares it with the decision-making practice of the Court of Justice of the European Union. The author also deals with the amendment of transfer of rights and obligations implemented by the Act No. 285/2020 Coll., amending the Act No. 262/2006 Coll., the Labour Code, as amended, and several other related acts, and compares the applicable law with the preceding. Method for the Simultaneous Establishment of Many Axenic Cultures of Paramecium * †
No full textA method is described for the simultaneous treatment of 42 (or more) stocks of Paramecium , and their adaptation to growth in axenic culture. Samples of dense cultures of these ciliates growing with Enterobacter aerogenes are rendered bacteria-free by migration through 2 sets of tubes containing Adaptation Medium (Peters' salts solution, stigmasterol, vitamins, and autoclaved E. aerogenes ). The 2nd set of tubes contains Adaptation Medium plus antibiotics. Bacteria-free samples containing ∼ 100 animals are then transferred to test tubes containing Adaptation Medium without antibiotics. This medium also serves as a growth medium. It supports indefinite growth of all Paramecium stocks tested. After adaptation to this medium, the ciliates can be grown in the axenic medium developed by Soldo, Godoy & van Wagtendonk. On a single trial at least half of the stocks can be expected to produce axenic cultures within 5 to 10 days by these procedures. The method has been applied successfully to several of the species of the Paramecium aurelia complex, to all syngens of Paramecium multimicronucleatum , to several stocks of Paramecium jenningsi , and to 1 stock each of Paramecium caudatum and Paramecium calkinsi.
