A coalgebraic perspective on probabilistic logic programming

    No full text
Probabilistic logic programming is increasingly important in artificial intelligence and related fields as a formalism to reason about uncertainty. It generalises logic programming with the possibility of annotating clauses with probabilities. This paper proposes a coalgebraic perspective on probabilistic logic programming. Programs are modelled as coalgebras for a certain functor F, and two semantics are given in terms of cofree coalgebras. First, the cofree F-coalgebra yields a semantics in terms of derivation trees. Second, by embedding F into another type G, as cofree G-coalgebra we obtain a “possible worlds” interpretation of programs, from which one may recover the usual distribution semantics of probabilistic logic programming

Probabilistic logic programming is increasingly important in artificial intelligence and related fields as a formalism to reason about uncertainty. It generalises logic programming with the possibility of annotating clauses with probabilities. This paper proposes a coalgebraic semantics on probabilistic logic programming. Programs are modelled as coalgebras for a certain functor F, and two semantics are given in terms of cofree coalgebras. First, the cofree F-coalgebra yields a semantics in terms of derivation trees. Second, by embedding F into another type G, as cofree G-coalgebra we obtain a 'possible worlds' interpretation of programs, from which one may recover the usual distribution semantics of probabilistic logic programming. Furthermore, we show that a similar approach can be used to provide a coalgebraic semantics to weighted logic programming

Functorial semantics as a unifying perspective on logic programming

Logic programming and its variations are widely used for formal reasoning in various areas of Computer Science, most notably Artificial Intelligence. In this paper we develop a systematic and unifying perspective for (ground) classical, probabilistic, weighted logic programs, based on categorical algebra. Our departure point is a formal distinction between the syntax and the semantics of programs, now regarded as separate categories. Then, we are able to characterise the various variants of logic program as different models for the same syntax category, i.e. structure-preserving functors in the spirit of Lawvere’s functorial semantics. As a first consequence of our approach, we showcase a series of semantic constructs for logic programming pictorially as certain string diagrams in the syntax category. Secondly, we describe the correspondence between probabilistic logic programs and Bayesian networks in terms of the associated models. Our analysis reveals that the correspondence can be phrased in purely syntactical terms, without resorting to the probabilistic domain of interpretation

Tingyu Liang,* Yixuan Song,* Lingui Gu, Yu Wang, Wenbin Ma Department of Neurosurgery, Peking Union Medical College Hospital, Chinese Academy of Medical Sciences and Peking Union Medical College, Beijing, People’s Republic of China*These authors contributed equally to this workCorrespondence: Wenbin Ma; Yu Wang, Email [email protected]; [email protected]: Glioblastoma (GBM) is the most common malignant primary brain cancer in adults. It is always resistant to existing treatments, including surgical resection, postoperative radiotherapy, and chemotherapy, which leads to a dismal prognosis and a high relapse rate. Therefore, novel curative therapies are urgently needed for GBM. Chimeric antigen receptor T (CAR-T) cell therapy has significantly improved life expectancy for hematological malignancies patients, and thus it increases the interest in applying CAR-T cell therapy for solid tumors. In the recently published research, it is indicated that there are numerous obstacles to achieve clinical benefits for solid tumors, especially for GBM, because of GBM anatomical characteristics (the blood–brain barrier and suppressive tumor microenvironment) and the tumor heterogeneity. CAR-T cells are difficult to penetrate blood–brain barrier, and immunosuppressive tumor microenvironment (TME), which induces CAR-T cell exhaustion, impairs CAR-T cell therapy response. Moreover, under the pressure of CAR-T cell therapy, the tumor heterogeneity and tumor plasticity drive tumor evolution and therapy resistance, such as antigen escape. Nonetheless, scientists strive for strategies to overcome these hurdles, including novel CAR-T cell designs and regional delivery. For instance, the structure of multi-antigen-targeted CAR-T cells can enrich CAR-T accumulation in tumor TME and eliminate abundant tumor cells to avoid tumor antigen heterogeneity. Additionally, paired with an immune modifier and one or more stimulating domains, different generation of innovations in the structure and manufacturing of CAR-T cells have improved efficacy and persistence. While single CAR-T cell therapy receives limited clinical survival benefit. Compared with single CAR-T cell therapy, the combination therapies have supplemented the treatment paradigm. Combinatorial treatment methods consolidate the CAR-T cells efficacy by regulating the tumor microenvironment, optimizing the CAR structure, targeting the CAR-T cells to the tumor cells, reversing the tumor-immune escape mechanisms, and represent a promising avenue against GBM, based on multiple impressive research. Moreover, exciting results are also reported to be realized through combining effective therapies with CAR-T cells in preclinical and clinical trials samples, have aroused inspiration to explore the antitumor function of combination therapies. In summary, this study aims to summarize the limitation of CAR-T cell therapies and introduces novel strategies to enhance CAR-T cell function as well as prospect the potential of the therapeutic combination. Keywords: CAR-T, GBM, novel strategies, therapeutic combinatio

Expanding Ethical Horizons: Rethinking the Ethics of De 德 and Guṇa in the Laozi 老子 and Bhagavad-Gītā

    No full text
This paper aims to engage in an ethical discussion of de in the Laozi and guṇa in the Bhagavad-Gītā to expand the horizon of our ethical understanding of Chinese and Indian philosophy. First, this paper will explore the different ethical levels of de and guṇa and discuss how these levels operate and are bound together. From an ethical perspective, this paper points out that de and guṇa can each be divided into two parts—higher de and lower de, and higher guṇa and lower guṇa. It will further explain the ethical significance of de and guṇa, arguing that they offer a unique contribution to developing our contemporary understanding of ethics. We will also seek to discover the interplay of de and/with the dao in the Laozi and that among the three guṇas in the Gītā from the perspective of human behaviors. In the conclusion, we will provide a comparative discussion of our findings on the ethics of de and guṇa in the Laozi and the Gītā. These findings suggest that de and guṇa promote a shared philosophical outlook within their respective traditions and thereby make a contribution to post-comparative ethical approaches

DataSheet1_Protective effect of compatible herbs in Jin-Gu-Lian formula against Alangium chinense-induced neurotoxicity via oxidative stress, neurotransmitter metabolisms, and pharmacokinetics.DOCX

Background:A. chinense frequently used in Miao medicine to treat rheumatic diseases. However, as a famous toxic herb, Alangium chinense and its representative components exhibit ineluctable neurotoxicity, thus creating significant challenges for clinical application. The combined application with compatible herbs in Jin-Gu-Lian formula attenuates such neurotoxicity according to the compatible principle of traditional Chinese medicines.Purpose: We aimed to investigate the detoxification of the compatible herbs in Jin-Gu-Lian formula on A. chinense-induced neurotoxicity and investigate its mechanism. Methods: Neurobehavioral and pathohistological analysis were used to determine the neurotoxicity in rats administered with A. chinense extract (AC), extract of compatible herbs in Jin-Gu-Lian formula (CH) and combination of AC with CH for 14 days. The mechanism underlying the reduction of toxicity by combination with CH was assessed by enzyme-linked immunosorbent assays, spectrophotometric assays, liquid chromatography tandem-mass spectrometry and real-time reverse transcription-quantitative polymerase chain reaction. Results: Compatible herbs attenuated the AC-induced neurotoxicity as evidenced by increased locomotor activity, enhanced grip strength, the decreased frequency of AC-induced morphological damage in neurons, as well as a reduction of neuron-specific enolase (NSE) and neurofilament light chain (NEFL) levels. The combination of AC and CH ameliorated AC-induced oxidative damage by modulating the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and total antioxidant capacity (T-AOC). AC treatment significantly reduced the levels of monoamine and acetylcholine neurotransmitters in the brains of rats, including acetylcholine (Ach), dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC), homovanillic acid (HVA), norepinephrine (NE), and serotonin (5-HT). Combined AC and CH treatment regulated the abnormal concentrations and metabolisms of neurotransmitters. Pharmacokinetic studies showed that the co-administration of AC and CH significantly decreased plasma exposure levels of two main components of AC, as evidenced by the reduction of maximum plasma concentration (Cmax), area under the plasma concentration-time curve (AUC) compared to AC. In addition, the AC-induced downregulation in mRNA expression of cytochrome P450 enzymes was significantly reduced in response to combined AC and CH treatment. Conclusion: Compatible herbs in Jin-Gu-Lian formula alleviated the neurotoxicity induced by A. chinense by ameliorating oxidative damage, preventing abnormality of neurotransmitters and modulating pharmacokinetics.</p
