Yeast linear plasmid with T(2)AG(3) telomeres: TEL+CEN antagonisms and genetic and molecula stability
No full textA linear plasmid containing ARS1, CEN4, and 48 bp of vertebrate (T(2)AG(3)) telomeric sequences at each end was used to transform Saccharomyces cerevisiae. Only circular plasmids that had lost the centromere and had retained the T(2)AG(3) sequences were obtained, indicating that the vertebrate T(2)AG(3) sequences and the yeast CEN4 could not be simultaneously present in this vector. This hypothesis was verified by removing the CEN4 sequence from the construct. In fact, the resulting transformants contained two classes of efficiently replicating linear plasmids: one of the expected size and one about twice as large. During subsequent growth, plasmids of the former, but not latter, class were subjected to concatemer formation. This can best be explained by recombination events involving the T(2)AG(3) sequences al the ends of the molecule, since very similar centric and acentric linear plasmids bearing Tetrahymena telomeric ends replicated faithfully

No full textIl contributo analizza il d.l. n. 113/2018, convertito in L. n. 132/2018, focalizzandosi sulle specifiche disposizioni in materia di sicurezza urbana: dalla previsione di forme di coordinamento tra le forze di polizia statali e locali, con particolare riferimento all’accesso al CED del Ministero dell’interno, fino all’ampliamento dei soggetti potenziali destinatari del divieto di accesso a determinati luoghi, passando per la sperimentazione del taser da parte della Polizia municipale e all’inasprimento delle misure di contrasto e delle sanzioni in materia di spaccio di sostanze stupefacenti, mendicità e esercizio abusivo dell’attività di parcheggiatore. Viene proposto un quadro problematico dei più recenti interventi normativi in materia di “sicurezza urbana”, nel tentativo di ricostruire gli esatti confini, le finalità e i diversi protagonisti coinvolti nella gestione integrata di una politica che rappresenta, ormai da anni, uno dei fronti più caldi e contrastati dell’agire pubblico

Heterogeneous and tissue-specific regulation of effector T cell responses by IFN-gamma during Plasmodium berghei ANKA infection.
No full textIFN-γ and T cells are both required for the development of experimental cerebral malaria during Plasmodium berghei ANKA infection. Surprisingly, however, the role of IFN-γ in shaping the effector CD4(+) and CD8(+) T cell response during this infection has not been examined in detail. To address this, we have compared the effector T cell responses in wild-type and IFN-γ(-/-) mice during P. berghei ANKA infection. The expansion of splenic CD4(+) and CD8(+) T cells during P. berghei ANKA infection was unaffected by the absence of IFN-γ, but the contraction phase of the T cell response was significantly attenuated. Splenic T cell activation and effector function were essentially normal in IFN-γ(-/-) mice; however, the migration to, and accumulation of, effector CD4(+) and CD8(+) T cells in the lung, liver, and brain was altered in IFN-γ(-/-) mice. Interestingly, activation and accumulation of T cells in various nonlymphoid organs was differently affected by lack of IFN-γ, suggesting that IFN-γ influences T cell effector function to varying levels in different anatomical locations. Importantly, control of splenic T cell numbers during P. berghei ANKA infection depended on active IFN-γ-dependent environmental signals--leading to T cell apoptosis--rather than upon intrinsic alterations in T cell programming. To our knowledge, this is the first study to fully investigate the role of IFN-γ in modulating T cell function during P. berghei ANKA infection and reveals that IFN-γ is required for efficient contraction of the pool of activated T cells
