    The role of mitochondrial dysfunction in acute pancreatitis

    Acute pancreatitis is a serious and often lethal inflammatory disease. Its causes are diverse and incompletely understood; however, gallstones and alcohol abuse are the principal triggers. Oxidative stress has been proposed as a determinant of acute pancreatitis (AP) severity, and has been the subject of recent clinical trials. The major AP precipitants, alcohol, alcohol metabolites and bile salts, were investigated for their potential role in the production of reactive oxygen species (ROS), and their effects upon cell fate. Application of the bile salt taurolithocholic acid sulphate (TLC-S) to isolated human and murine pancreatic acinar cells generated significant Ca2+-dependent mitochondrial ROS which were inhibited with the antioxidant N-acetyl-L-cysteine (NAC), and promoted with dimethoxy-2-methylnaphthalene (DMN), an inhibitor of the antioxidant enzyme NAD(P)H quinone oxidoreductase (NQO1). Elevations of ROS mediated by bile salts were crucial in the determination of cell fate, producing apoptosis rather than necrosis. In contrast, ethanol and its metabolites, both oxidative (acetaldehyde) and non-oxidative (fatty acid ethyl esters: FAEEs), were shown to produce no significant ROS in similar circumstances. Assessment of ethanol and its metabolites revealed that ethanol and acetaldehyde showed little effect on cell fate. Low concentrations of ethanol with fatty acid, however, induced toxic elevations of [Ca2+]C, mitochondrial dysfunction and necrosis when oxidative metabolism was compromised. This effect was reversed by inhibition of FAEE synthase, suggesting important deleterious actions of non-oxidative alcohol metabolism in the pancreas

    Cult: A Composite Novel

    Cult (redacted) The first component of the thesis is a composite novel called Cult which falls into two parts with seven narratives in each. Part 1 tracks the protagonist, Ellen, from her first involvement with the cult through to her eventually leaving it. Although fiction, the first half of the book answers the kinds of questions the author is asked when people discover that she was once a sannyasin (a follower of the guru Bhagwan Shree Rajneesh). While the experiences of meditation, group therapy and communal living are all faithfully rendered within the stories, the need for strong characters, narrative drive and a lightness of touch takes precedence. Part 2 picks up Ellen’s story some twenty or so years later and explores what becomes of her in middle age. It also looks at other groups in society, such as academia, the law and the internet dating community which each have their own jargon, hierarchies, rituals and rules but are not considered to be cults. The book examines the question raised in the Epigraph, ‘how do we be together when we feel so alone’ with a focus on relationships other than the familial and the romantic. Collisions, Chasms and Connections: a Performative Exploration of the Composite Novel Form The second part of the thesis is both a critical and creative response to three contemporary American books: Olive Kitteridge by Elizabeth Strout; A Visit from the Goon Squad by Jennifer Egan; and Legend of a Suicide by David Vann. The critical element comprises a close reading of the three books; a chronological reconstruction of their overarching storylines; and a consideration of what their authors have said about writing the books. It concludes that, in the composite novel, the simultaneous presentation of multiple views and storylines operate much like a 3D image to give the impression of depth to the characters and situations rendered. The creative element of the essay is a playful and personal response to the texts

    Yogic breathing instruction in patients with treatment‑resistant generalized anxiety disorder: Pilot study

    Aim: This study aims to evaluate the feasibility and effects of instruction in yogic breathing techniques (Pranayama) in patients with treatment-resistant generalized anxiety disorder (GAD) in UK secondary mental health services settings. Materials and Methods: Participants were adult primary or secondary care patients with a primary diagnosis of GAD (with or without comorbidity) and persistent anxiety symptoms of at least moderate intensity, despite prior treatment with two or more medications of proven efficacy. Patients participated in group-delivered yogic breathing training and practice for 12 weeks. Structured assessments were performed at baseline, after 1, 2, and 6 weeks of instruction, and at end-point. Participants also completed the antisaccade (emotional variant) task and startle response task at baseline and end-point. Results: At baseline, participating patients (n = 9) had moderate-to-severe anxiety symptoms and mild-to-moderate depressive symptoms, they attended 84% of offered sessions and provided positive feedback on the content and delivery of treatment. Symptom severity reduced significantly from baseline to end-point. There were greater errors on negative trials compared to neutral trials in the antisaccade task at baseline, and a significant reduction in antisaccade errors for negative stimuli as compared to neutral stimuli between baseline and end-point: but there were no significant differences in either mean heart rate or startle response between baseline and end-point. Limitations: The absence of a control group and small sample size. Conclusion: Yogic breathing techniques proved simple to learn and may be beneficial in reducing anxiety and depressive symptoms in patients with treatment-resistant GAD. Yogic breathing had no effect on autonomic arousal, but the reduction in errors to negative stimuli in the antisaccade task suggests an improvement in attention control during the intervention accompanying the reduction in symptoms

    Dynamics and folding of single two-stranded coiled-coil peptides studied by fluorescent energy transfer confocal microscopy

    We report single-molecule measurements on the folding and unfolding conformational equilibrium distributions and dynamics of a disulfide crosslinked version of the two-stranded coiled coil from GCN4. The peptide has a fluorescent donor and acceptor at the N termini of its two chains and a Cys disulfide near its C terminus. Thus, folding brings the two N termini of the two chains close together, resulting in an enhancement of fluorescent resonant energy transfer. End-to-end distance distributions have thus been characterized under conditions where the peptide is nearly fully folded (0 M urea), unfolded (7.4 M urea), and in dynamic exchange between folded and unfolded states (3.0 M urea). The distributions have been compared for the peptide freely diffusing in solution and deposited onto aminopropyl silanized glass. As the urea concentration is increased, the mean end-to-end distance shifts to longer distances both in free solution and on the modified surface. The widths of these distributions indicate that the molecules are undergoing millisecond conformational fluctuations. Under all three conditions, these fluctuations gave nonexponential correlations on 1- to 100-ms time scale. A component of the correlation decay that was sensitive to the concentration of urea corresponded to that measured by bulk relaxation kinetics. Thetrajectories provided effective intramolecular diffusion coefficients as a function of the end-to-end distances for the folded and unfolded states. Single-molecule folding studies provide information concerning the distributions of conformational states in the folded, unfolded, and dynamically interconverting states.Author manuscript.     An Impact Study of the Economic Partnership Agreements (EPAs) in the Six ACP Regions

    This article intends to present a very detailed analysis of the trade-related aspects of Economic Partnership Agreements (EPAs) negotiations. We use a dynamic partial equilibrium model – focusing on the demand side – at the HS6 level (covering 5,113 HS6 products). Two alternative lists of sensitive products are constructed, one giving priority to the agricultural sectors, the other focusing on tariff revenue preservation. In order to be WTO compatible, EPAs must translate into 90 percent of bilateral trade fully liberalised. We use this criterion to simulate EPAs for each negotiating regional block. ACP exports to the EU are forecast to be 10 percent higher with the EPAs than under the GSP/EBA option. On average ACP countries are forecast to lose 70 percent of tariff revenues on EU imports in the long run. Yet imports from other regions of the world will continue to provide tariff revenues. Thus when tariff revenue losses are computed on total ACP imports, losses are limited to 26 percent on average in the long run and even 19 percent when the product lists are optimised. The final impact on the economy depends on the importance of tariffs in government revenue and on potential compensatory effects. However this long term and less visible effect will mainly depend on the capacity of each ACP country to reorganise its fiscal base.Preferential Trade Agreements, Africa, EPAs, Partial Equilibrium Simulations, International Relations/Trade,

    Ammonia (NH3) emissions mainly originate from agricultural practices and can have multiple adverse impacts on the environment. With the substantial increase of synthetic fertilizer use over the past decades, volatilization of NH3 has become a major loss of N applied to land. Since NH3 can be strongly influenced by both environmental conditions and local management practices, a better estimate of NH3 emissions from fertilizer use requires improved understanding of the relevant processes. This study describes a new process-based model, AMmonia–CLIMate (AMCLIM), for quantifying agricultural NH3 emissions. More specifically, the present paper focuses on the development of a module (AMCLIM–Land) that is used for simulating NH3 emissions from synthetic fertilizer use. (Other modules, together termed as AMCLIM-Livestock, simulate NH3 emissions from agricultural livestock, are described in Part 2). AMCLIM–Land dynamically models the evolution of N species in soils by incorporating the effects of both environmental factors and management practices to determine the NH3 emissions released from the land to the atmosphere. Based on simulations for 2010, NH3 emissions resulting from the synthetic fertilizer use are estimated at 15.0 Tg N yr-1, accounting for around 17 % of applied fertilizer N.     Partial discharge on-line monitoring for HV cable systems using electro-optic modulators

    A technique for the remote inspection and monitoring of partial discharge (PD) activity using an optical network is described. The network uses a LiNbO3 modulator to modulate the intensity of the transmitted laser light approximately proportional to the voltage applied across the modulator. The laser light is transmitted along an optical fibre and measured remotely by a high-speed optical receiver. A capacitive coupler has been used to detect partial discharge activity and act as the modulator’s input voltage source. The electro-optic modulator is passive and does not require a power supply at the site of the PD detection sensor. The system has the additional advantages of being immune to electromagnetic interference, having very little signal transmission attenuation, with good sensitivity, compact size, as well as being convenient to use and safe. Both simulation experiments and practical PD tests on two cable systems have indicated that the optical system provides a feasible remote PD monitoring technique for high voltage cable systems.<br/
