    Genome-wide association study of adolescent idiopathic scoliosis in Southern Chinese and significant improvement in detection of missing heritability and genetic risk prediction in genome-wide association studies by 1000 genomes-based imputation

    Adolescent idiopathic scoliosis (AIS) is a lateral curvature of the spine affect school age children. Although many studies have been conducted to investigate the candidate loci and genes for AIS, the underlying mechanisms of the pathogenesis is still poorly understood. In this study, a genome-wide association study on a cohort consisting of 189 AIS patients and 313 controls was performed to investigate the genetic variants associated with AIS in the southern Chinese population. The most promising association region is 12p13.32, in which a significant linkage signal was found in previous study. The three most significant SNPs (rs10849195, P = 6.84 × 10-7, OR = 0.51, 95% CI 0.39-0.67; rs11063476, P = 9.82 × 10-6, OR = 0.55, 95% CI 0.42-0.72; rs2109185, P = 3.83 × 10-5, OR = 0.57, 95% CI 0.44-0.75) in this region were in the same linkage disequilibrium (LD) block of about 2 kb. The most significant SNPs rs10849195 on chromosome 12 was successfully replicated in two independent replication studies. One replication study with 1271 individuals consist of 568 cases and 703 controls (P = 0.037, OR = 0.84, 95% CI 0.72-0.99), and the other replication study with 536 individuals consist of 286 cases and 250 controls (P = 0.026, OR = 0.76, 95% CI 0.60-0.97). The combined two stage P-value is 9.1 × 10-7, OR = 0.74. SNP rs11190870 near LBX1 (ladybird homeobox 1), which was associated with Japanese adolescent idiopathic scoliosis patients by a genome-wide association study, was also validated in an independent southern Chinese population. The results suggested that SNP rs11190870 is statistically significant associated with adolescent idiopathic scoliosis in southern Chinese (P = 9.1 10-10; odds ratio = 1.85; 95% confidence interval = 1.52 - 2.25). Quality control (QC) by removing individuals or markers with low genotyping quality is of utmost importance to minimize potential false positive associations. IPGWAS was developed to facilitate the identification of the rational thresholds in QC of GWAS datasets, association analysis, Manhattan plot, quantile-quantile (QQ) plot, and format conversion for genetic analyses, such as meta-analysis, genotype phasing, and imputation. IPGWAS is freely available at http://sourceforge.net/projects/ipgwas/. Missing heritability is a common problem in genome-wide association study. The Wellcome Trust Case Control Consortium Phase I genotype data were imputed using 1000 genomes as reference. Then phenotypic variance explained by SNPs passed certain p-value thresholds was estimated. The results suggested that the proportions of phenotypic variance explained by genetic variants increased significantly when the new association variants identified through 1000 genome based imputation were included for most complex diseases. The obtained results were consistent with the larger number of variants that are yet to be identified as potential sources of missing heritability. Most GWAS results cannot achieve a reliable genetic risk prediction performance. After genotype imputation, both prediction sensitivity and specificity were increased to a relative reliable level (>0.7).     Effect of coordination number of particle contact force on rutting resistance of asphalt mixture

    Optimizing asphalt mix design at the indoor stage is of significant importance for enhancing the rutting resistance of asphalt mixture, which is affected by its structural characteristics. In this work, the coordination number of particle contact force (CNpcf) was proposed as an indicator to represent contact characteristics of skeleton structure aggregates in asphalt mixture. Nine asphalt mixtures with different gradations were designed, and the relationship of CNpcf with the number of aggregate contact zones (CZ) was established by combining rutting tests and digital image processing technique (DIP). The Mann-Whitney U test was implemented to analyze the distribution properties of inter-particle contacts before and after the rutting test. In addition, the resistance to the further expansion of rutting was analyzed. The results revealed a significant positive correlation (PCCs = 0.843, R2 = 0.711) between CNpcf and CZ. The content of coarse aggregates in the dominant structure did not exhibit monotonic related to anti-rutting performance of the asphalt mixture. Therefore, an optimum aggregate content of 57% was utilized. The Mann-Whitney U test revealed that the mesoscale skeleton structure of the asphalt mixes before and after rutting exhibited excellent stability.     National Blood Pressure Reference for Chinese Han Children and Adolescents Aged 7 to 17 Years

    We sought to develop and validate a national blood pressure (BP) reference based on age, sex, and height for Chinese children. Data were obtained on 197 430 children aged 7 to 17 who participated in the Chinese National Survey on Students&apos; Constitution and Health in 2010. BP percentiles were estimated and fitted using the lambda, mu, and sigma method and then compared with a US reference and China existing reference. In an external independent validation sample of 59 653 children aged 7 to 18 from 7 Chinese provinces in 2013, the prevalence of elevated BP was compared applying the 3 references. BP values were similar for boys and girls at the younger ages (7-13 years) and lower height percentiles, whereas higher at the older ages (14-17 years) for boys than girls. At medial height in boys and girls aged 7 to 13, the 50th, 90th, 95th, and 99th percentiles of BP for the new national reference were consistent with US reference and lower than current Chinese reference. In the independent sample, elevated BP prevalence, based on the new national reference, ranged from 7.8% to 18.5% among children aged 7 to 17, which was higher than the US reference values (4.3%-14.5%) and lower than the current Chinese reference (12.9%-25.5%) in each age group.     The change in temporal trend and spatial distribution of CO<sub>2</sub> emissions of China's public and commercial buildings

    The CO2 emission mitigation of the commercial and public building sector (P&amp;C) is critical for achieving China's carbon peak and carbon neutrality. Analyzing changes in CO2 emissions and their driving factors from temporal and spatial perspective provides insights for developing equitable and effective decarbonization strategies. This study investigated the change in temporal trend and spatial distribution of CO2 emissions of China's P&amp;C during period of 2005–2018 according to the Kaya identify and Gravity Center model. Meanwhile, combined with the Logarithmic Mean Divisia Index, this study proposed a decomposed method to identify the driving factor of the movement of the gravity center. The results showed that: 1) in the temporal dimension, China's P&amp;C has still not achieved its CO2 emissions peak, arriving 820.68 MtCO2. The most positive and negative driving factors were per capita add value of tertiary industry and energy efficiency, respectively; 2) in the spatial dimension, during the 13th Five Year Plan period, the gravity center moved southwestward, and the most positive and negative driving factors were energy consumption unit area and energy efficiency, respectively; Besides, to accelerate the decarbonization of China's P&amp;C, this study reviewed the main decarbonization strategies, divided them into six categories and provided policy implications.
