    Multiple deletions of the osmolyte transporters BetL, Gbu, and OpuC of Listeria monocytogenes affect virulence and growth at high osmolarity

    The success of Listeria monocytogenes as a food-borne pathogen owes much to its ability to survive a variety of stresses, both in the food environment and, after ingestion, within the animal host. Growth at high salt concentrations is attributed mainly to the accumulation of organic solutes such as glycine betaine and carnitine. We characterized L. monocytogenes LO28 strains with single, double, and triple deletions in the osmolyte transport systems BetL, Gbu, and OpuC. When single deletion mutants were tested, Gbu was found to have the most drastic effect on the rate of growth in brain heart infusion (BHI) broth with 6dded NaCl. The highest reduction in growth rate was found for the triple mutant LO28BCG (ΔbetL ΔopuC Δgbu), although the mutant was still capable of growth under these adverse conditions. In addition, we analyzed the growth and survival of this triple mutant in an animal (murine) model. LO28BCG showed a significant reduction in its ability to cause systemic infection following peroral coinoculation with the wild-type parent. Altering OpuC alone resulted in similar effects (R. D. Sleator, J. Wouters, C. G. M. Gahan, T. Abee, and C. Hill, Appl. Environ. Microbiol. 67:2692-2698, 2001), leading to the assumption that OpuC may play an important role in listerial pathogenesis. Analysis of the accumulation of osmolytes revealed that betaine is accumulated up to 300 μmol/g (dry weight) when grown in BHI broth plus 6␗aCl whereas no carnitine accumulation could be detected. Radiolabeled-betaine uptake studies revealed an inability of BGSOE (ΔbetL Δgbu) and LO28BCG to transport betaine. Indeed, for LO28BCG, no accumulated betaine was found, but carnitine was accumulated in this strain up to 600 μmol/g (dry weight) of cells, indicating the presence of a possible fourth osmolyte transporter

    Noise due to unsteady flow past trailing edges

    This paper presents two-dimensional direct numerical simulations (DNS) of noise generated at trailing edges (TE) with zero thickness. The simulations are conducted specifying either no-slip or slip walls in order to investigate viscous effects. In both cases, small amplitude disturbances are introduced close to the inflow boundary that serve as pressure disturbances at the TE. DNS data reveals that the unsteady Kutta condition is not satisfied, irrespective of the wall boundary condition. However, it appears that the validity of the unsteady Kutta condition is not essential for making an accurate prediction of the far field noise. The far field pressure is predicted as a function of the surface pressure difference using a 2-D modification of Amiet's classical theory, and compared with the far field pressure computed directly. Directivity plots provide evidence that the presence of boundary layers and noise generated by an unsteady wake in the no-slip cases lead to smearing of individual lobes, and that the downstream pointing lobes in no-slip wall cases are probably due to nonlinear noise generation in the wake. The simulations are conducted using a high-order accurate numerical method which is free of upwinding, artificial dissipation or any form of explicit filtering, and employs a novel boundary treatment

    Characteristic distribution and scale interaction of turbulence in a boundary layer

    This work revisits the concept of turbulent boundary layers from a novel perspective on scale transfer. Turbulence production and dissipation together with the energy budgets are analyzed in the velocity gradient invariant phase space. In combination with filtering, the mechanism of scale coupling is investigated and illustrated for different characteristic flow topologies. The understanding of the scale coupling is important to model turbulence. Turbulence models describe the complex interaction of the scales of motion in a simplified form. The essential task of turbulence modeling is to capture the coupling of the modeled and unmodeled scales as well as the evolution of the modeled scales within the unmodeled flow. This work characterizes the scale coupling by focusing on the interfaces between modeled and unmodeled flow such as production and dissipation. The mechanisms that govern the evolution of the modeled quantities are investigated for their core properties and universal features. Direct numerical simulation (DNS) is carried out to obtain data of a compressible zero pressure-gradient flat plate turbulent boundary layer flow. This flow topology allows to unveil the effect of a wall on the coupling of scales and evolution of turbulence

    The radical psychiatrist R.D. Laing (1927-1989) was an accomplished author with an extensive philosophical knowledge that informed his ideas on reading, writing, and interpretation. Laing argues that psychiatry should be modeled on skilful textual exegesis rather than scientific explanation. The exegesis of a psychotic’s words and actions is difficult, he infers, because the impoverishment of our experience cuts us off from the sense that lies within seeming madness. Like philosophers such as Edmund Husserl, Laing therefore criticizes the way in which the natural sciences have invalidated subjective experience. He consequently employs a rhetoric designed to disclose with renewed vigor its complexity, variety and reality. Laing fails, however, to find an alternative to scientific reason: "experience", in his weakest work, is an irrational realm of mystical and self-validating certainty that closely parallels Heidegger’s later accounts of "Being"

    Genetic and phenotypic trends for carcass traits in Irish beef cattle

    Animals subjected to intensive genetic selection require continuous monitoring to observe changes in economically important traits. The focus of this research was to estimate genetic and phenotypic trends for carcass traits for three genotypes: dairy dam and dairy sire (D×D), dairy dam and beef sire (D×B) and beef dam and beef sire (B×B). Linear regression models were used to generate genetic and phenotypic trends for carcass weight, conformation and fat score for both cows and prime animals (i.e. never parented an animal). Phenotypic trends for age at slaughter were also estimated in prime animals. Results indicated that carcass weight increased genetically by 1.1 kg per year for B×B animals, but decreased by 0.14 and 0.39 kg per year for D×B and D×D animals, respectively. Phenotypic trends reflected the genetic trends for carcass weight for D×B and B×B animals, but the decline in genetic trends for carcass weight for D×D animals was not replicated in their phenotypic trends. Carcass conformation declined genetically by 0.02 and 0.05 units per year for D×D and D×B animals, respectively, and increased by 0.05 units per year for B×B animals. Phenotypic trends for conformation were similar to genetic trends. Carcass fat showed a decrease genetically for D×D and B×B, but remained constant for D×B animals, although this was not reflected in the phenotypic trends. Age at slaughter decreased phenotypically for all genotypes. Current breeding programmes have a favourable impact on carcass traits for B×B animals, but an unfavourable trend was observed for D×D and D×B animals
