As a control, amplification was also performed using pNL4.3 plasmid as a template (lane 8). Bands corresponding to vif-2, vpr-3 and env-1 mRNAs are indicated by arrows. In the mutant virus, all amplicons were shortened by 34 nt, due to the SL1 deletion. This result is representative of at least three independent experiments

Dr. Terrezza began his manufacturing in an 1,100 square foot room in one of his offices in Pensacola with the help of a colleague, Rolando Domingo, who would become the lab manager. Dr. Terrezza and Rolando saw great returns when he began manufacturing lenses for his patients and continued to see more returns when he gained more accounts from his colleagues in the area of Pensacola.     Music production in the FL Studio software in leisure activities

    Much of today's teenage generation identifies with rap music. Each rap is accompanied by a beat, which is usually created in music software called a DAW. FL Studio is one of these DAWs. The aim of this thesis is to create a methodology for teaching FL Studio in hobby groups or ZUŠ. Another aim is to verify whether continuous work with FL Studio develops the student's musicality similarly to playing instrument or singing. The thesis also explores the impact of learning to work with FL Studio on learner wellbeing. Most recently, this thesis investigates the effect of the use of DAWs in primary school music education on pupils' motivation. The method of research in the thesis is case studies, the method of data collection is observation and interviews with students. The author of the paper concluded that continuous work with FL Studio really develops the pupil's musicality, it can even be a way for pupils with reduced motor skills but musical talent. The impact on pupils' wellbeing was positive for all participants observed. In music education classes, where the author applied the work with FL Studio, there was the expected revival of teaching and two pupils subsequently enrolled in the electronic music club.     This paper is the product of long studies on the specific character of FL teachers’ work. It comprises the latest achievements of language teaching methodology. The author considers it to be especially important since her intention is to combine such procedures as diary writing and lesson observations into one model that concentrates on FL teachers’ creativity. The first level of this model is designed for student teachers. Later, participant observations and self observations are expected to contribute to the development of FL teacher’s autonomy.     Are SLA Research Reports of Value for FL Teachers? On the Need for Researcher-Teacher CollaborationThe paper discusses the issue of second language acquisition SLA research reports andthe application of their findings in practice. On the basis of interviews with researchers and practitioners as well as the author&rsquo;s own experience, the reasons for the gap between theory and practice are explained. Some solutions are also offered to bridge the gap, such as teachers becoming researchers or collaboration between SLA researchers and practitioners. Finally, the author attempts to answer the question posed in the title of the paper. Since SLA research reports are not appreciated by many foreign language FL teachers, some solutions are proposed, such as eliminating research jargon, complex statistics and theoretical frameworks and introducing topics of relevance to FL teachers. Každý rap je doprovázen tzv. beatem, který bývá vytvořený v hudebním software zvaném DAW. Jedním z takových DAW je i FL Studio. Tato práce si klade za cíl vytvořit metodiku výuky práce s FL Studiem v zájmových kroužcích či ZUŠ. Dalším cílem je ověřit, zda soustavná práce s FL Studiem rozvíjí hudebnost žáka podobně jako hra na nástroj či zpěv. Práce se také zabývá tím, jaký vliv má studium práce s FL Studiem na wellbeing žáka. Naposledy práce zkoumá, jaký vliv na motivaci žáků má využití DAW v hudební výchově základní školy. Metodou výzkumu v diplomové práci jsou případové studie, metodou sběru dat je pozorování a rozhovory se žáky. Autor práce dospěl k závěru, že soustavná práce s FL Studiem hudebnost žáka opravdu rozvíjí, může být dokonce cestou pro žáky se sníženými motorickými schopnostmi, ale přesto s hudebním nadáním. Vliv na wellbeing žáků byl u všech pozorovaných účastníků pozitivní. V hodinách hudební výchovy, kde autor aplikoval práci s FL Studiem, došlo k očekávanému oživení výuky a dva žáci se následně přihlásili na kroužek elektronické hudby. Nakonec autor práce prohlašuje, že výuka hudební produkce by mohla být jednou z cest, kterou by se současná hudební pedagogika měla zabývat.Katedra hudební výchovyFaculty of EducationPedagogická fakult
