Desiring the east: a comparative study of Middle English romance and modern popular sheikh romance
This thesis comparatively examines a selection of twenty-first century sheikh romances and Middle English romances from the fourteenth and fifteenth centuries that imagine an erotic relationship occurring between east and west. They do so against a background of conflict, articulated in military confrontation and binary religious and ethnic division. The thesis explores the strategies used to facilitate the cross-cultural relationship across such a gulf of difference and considers what a comparison of medieval and modern romance can reveal about attitudes towards otherness in popular romance.
In Chapter 1, I analyse the construction of the east in each genre, investigating how the homogenisation of the romance east in sheikh romance distances it from the geopolitical reality of those parts of the Middle East seen, by the west, to be "other". Chapter 2 examines the articulation of gender identity and the ways in which these romances subvert and reassert binary gender difference to uphold normative heterosexual relations. Chapter 3 considers how ethnic and religious difference is nuanced, in particular through the use of fabric, breaking down the disjunction between east and west. Chapter 4 investigates the way ethnicity, religion and gender affect hierarchies of power in the abduction motif, enabling undesirable aspects of the east to be recast.
The key finding of this thesis is that both romance genres facilitate the cross-cultural erotic relationship by rewriting apparently binary differences of religion and ethnicity to create sameness. While the east is figured differently in Middle English and modern sheikh romance, the strategies they use to facilitate the cross-cultural erotic relationship are similar. The thesis concludes that the constancy of certain attitudes towards the east in both medieval and modern romance reveals a persistence of conservative values in representations of the east in romance
MOLECULAR STRATIFICATION AND PROGNOSTIC DETERMINANTS OF ADULT AND PEDIATRIC CROHN’S DISEASE
Benjamin P. Keith: Molecular stratification and prognostic determinants of adult and pediatric Crohn’s disease (Under the direction of Terry Furey, Praveen Sethupathy, and Shehzad Sheikh) Crohn’s disease (CD) is a chronic relapsing gastrointestinal inflammatory disorder with heterogeneous clinical presentation. Current clinical diagnostic methods involve invasive procedures and are ineffective towards predicting disease progression and response to therapy. High-throughput sequencing technologies have been utilized in various complex disorders to identify disease subtypes based on molecular signatures that are associated with clinical parameters. Given the success of molecular subtyping, particularly within the cancer field, there is substantive interest in implementing this methodology to predict disease progression and treatment response in CD. By analyzing differences in colonic gene expression between CD patients, our group revealed 2 subsets of patients—a group characterized by genes more highly expressed in the colon (colon-like CD) and a group with increased expression of ileum marker genes (ileum-like CD). Building on these initial findings, we aimed to validate molecular subtypes associated with CD through analysis of RNA-sequencing and small RNA-sequencing data in adult and pediatric cohorts. In the following chapters, I show that CD molecular subtypes can be detected using genome-wide expression of microRNAs, a class of small non-coding RNAs that post-transcriptionally regulate gene expression. Further, I show that microRNA-31 (miR-31) is a molecular driver of CD subtypes in both adult and pediatric patient cohorts and, through association analyses of clinical patient phenotypes, show that miR-31 expression levels are associated with the development of distinct disease outcomes. Using gene expression data from a large cohort of adult colon-like patients, I identified heterogeneous expression signatures that suggests an additional molecular subtype of CD associated with the expression of Paneth cell markers. Characterization of single-nucleotide polymorphisms implicated in the pathogenesis of Crohn's disease
The inflammatory bowel diseases (IBD) are a group of chronic inflammatory conditions
involving the gastrointestinal tract. IBD affects between 1 and 2 million Americans, and its incidence is increasing for unknown reasons. One of the predominant forms of IBD is Crohn’s disease (CD). Genome-wide association studies (GWAS) have linked many single-nucleotide polymorphisms (SNPs) within genes of the immune system to CD pathogenesis, but it is unknown which of these SNPs are causative. In genetically susceptible individuals, CD can result from a disruption in the balance between pro-inflammatory and anti-inflammatory gene expression in intestinal immune cells. Gene expression is partially regulated by the binding of transcription factors to DNA to either activate or repress target genes. The ability of transcription factors to bind to DNA may be influenced by chromatin accessibility. To determine the location of accessible, or “open” chromatin regions in the genome, we used Formaldehyde-Assisted Isolation of Regulatory Elements (FAIRE). Many of these open chromatin regions were located near genes involved in immune system function. A subset of these open chromatin regions
overlapped with CD-associated SNPs. I used luciferase reporter assays to determine the potential regulatory function of a CD-linked SNP found in a region of open chromatin. DEMONSTRATING MECHANISMS BY WHICH VITAMIN D REGULATES INFLAMMATION PATHWAYS IN SYSTEMIC LUPUS ERYTHEMATOSUS
Systemic Lupus Erythematosus (SLE, lupus) is a chronic autoimmune disease where healthy tissue is attacked due to a failure to differentiate self from non-self. While the etiology is unclear, a combination of genetic predispositions and environmental triggers are the likely cause. Lupus patients are prone to vitamin D deficiency. This is likely in part due to photosensitivity and, avoidance of the sun, a major source of vitamin D. Vitamin D supplementation has been explored as an intervention to reduce inflammation in patients but the mechanism of action has not been determined. Here, I review the current literature to investigate three mechanisms by which vitamin D likely plays a role in lupus etiology: lymphocyte regulation, epigenetic regulation of immune function, and intestinal barrier function. Providing evidence of the role of vitamin D in these three mechanisms that underlie lupus symptoms will help clarify important concepts in our understanding of disease pathogenesis.Master of Scienc
