Insulin resistance, obesity and the metabolic syndrome. Is there a therapeutic role for endothelin-1 antagonists?
There is increasing evidence to suggest that chronic activation of the endothelin-1 system can lead to heterologous desensitization of the glucose-regulatory and mitogenic actions of insulin with subsequent development of glucose intolerance, hyperinsulinemia, impaired endothelial function and exacerbation of cardiovascular disease. Effects are mediated through a variety of mechanisms that include attenuation of key insulin signalling pathways and decreased tyrosine phosphorylation of insulin receptor substrates IRS-1, SHC and G alpha q/11. Other actions involve hemodynamic changes leading to reduced delivery of insulin and glucose to peripheral tissues as well as enhanced hepatic glycogenolysis, decreased glucose-transporter translocation and modulation of various adipokines that regulate insulin action. Overall the data suggest that ET-1 antagonists may provide an effective means of improving cardiac dysfunction and favourably influencing glucose tolerance in obese humans and patients with early insulin sensitivity where there is clear evidence for activation of the ET-1 system. Although most effects of ET-1 that modulate mechanisms leading to glucose intolerance appear to involve the ETA receptor subtype recent data indicates that combined ETA/ETB receptor antagonists may function as effectively as selective ETA blockers. Prospective trials are needed to assess whether ET-1 antagonists, either alone or in combination, are superior to other more conventional therapies such as insulin sensitizers and to evaluate effects of combined treatments on the development of insulin resistance and the progression of diabetes. Early screening of patients at risk for evidence of ET-1 activation would help to identify subjects who may benefit most from such treatment

A 0.7-V 0.43-pJ/cycle Wakeup Timer based on a Bang-bang Digital-Intensive frequency-Locked-Loop for IoT Applications
A 40-nm CMOS wakeup timer employing a bang-bang digital-intensive frequency-locked loop for Internet-of-Things applications is presented. A self-biased ΣΔ digitally controlled oscillator (DCO) is locked to an RC time constant via a single-bit chopped comparator and a digital loop filter. Such highly digitized architecture fully exploits the advantages of advanced CMOS processes, thus enabling operation down to 0.7 V and a small area (0.07 mm 2 ). Most circuitry operates at 32× lower frequency than the DCO in order to reduce the total power consumption down to 181 nW. High frequency accuracy and a 10× enhancement of long-term stability is achieved by the adoption of chopping to reduce the effect of comparator offset and 1/f noise and by the use of ΣΔ modulation to improve the DCO resolution. The proposed timer achieves the best energy efficiency (0.43 pJ/cycle at 417 kHz) over prior art while keeping excellent on-par long-term stability (Allan deviation floor <;20 ppm) and temperature stability (106 ppm/°C).Accepted Author Manuscript(OLD)Applied Quantum Architecture

A Versatile and Efficient 0.1-to-11 Gb/s CML Transmitter in 40-nm CMOS
We present a wireline transmitter (TX) for re-configurable chip-to-chip links. The proposed design features a frequency-adaptive clock chain, a fast 16:1 clocked-CMOS multiplexer (C2MOS MUX) tree, and a full-rate synchronous current-mode logic (CML) clock driver. A prototype realized in 40-nm CMOS accomplishes a wide 0.1-to-11 Gb/s operation range (fmax/fmin = 110×). At 11 Gb/s, the prototype achieves 3.98 pJ/bit for a bit error rate (BER) < 10-12 with a 60.9-ps eye width.Accepted author manuscriptElectronic
Computing fluid-fluid interface evolution in Hele-Shaw cells using rational approximation
We deploy a rational solver to compute moving boundary problems in Hele-Shaw cells and porous media. At each time, a quasi-stationary Laplace problem for the pressure is solved using rational approximation. This furnishes the interfacial velocity from which the moving boundary is updated. Two approaches for representing the moving boundary are presented: orthonormal basis functions (such as Chebyshev polynomials) and level-set functions. For fluid domains with corners that are stationary or translate in a simple fashion, poles are exponentially clustered exterior to the fluid domain in the vicinity of the corner and moved in step with the respective vertex. In the case of more complicated flows, we use the adaptive Antoulas–Anderson (AAA) algorithm to select the location of poles at each time, based adaptively upon the interface shape and boundary conditions. The accuracy of the numerical method is verified by comparison with various exact solutions. The method has wide applicability, which we demonstrate through examples including porous gravity currents, one- and two-phase instabilities and flow in doubly connected regions. Key advantages of our method are its speed, relative simplicity of implementation and ability to handle interfaces with corners and regions of high curvature at low computational cost
