    The Blue Jackal

    How surprising that I lived in Berkeley for four years and never ran across this book or this publisher while I was there! A Google search shows that Vidya Books still exists in Berkeley, though apparently with a post office box number, not a street address. This twenty-two page large-format children's book often combines text and colored illustrations on its paired pages. Sharma narrates the traditional story as an expanded fable, with pleasing development of the situation (drought and heat), scene (a small village near the jungle), and time of day (siesta). About the middle of the story, a "has" needs to be a "had" in the phrase "He has finished dyeing a load of muslin blue that morning." Perhaps the strongest illustration in the book is that of the blue jackal after he emerges from the tub. This same illustration is used on the book's dust-jacket and cover. In this telling, the jackal goes too far when he tells the other animals "Lord Krishna has made me blue in his image. He has also given me some of his special powers." This version then nicely brings the story to its climax with the long-awaited monsoon rains, which of course wash off the jackal's blue dye. "The free ride was over." When the other animals pursue the fleeing jackal, he lets out the "yowl" that is the usual give-away as to his true identity. There is an afterword on Punchtuntra stories. To my surprise, Sharma dates Dabshalim to the fourth century BCE; she says that Dabshalim "defeated the Greek Governor left behind by Alexander." There is also this statement to consider: "Some scholars connect these tales to the African Aesop…." She comments pointedly on the meaning of this story: "The story of The Blue Jackal is told in India to instill the idea of a color-blind society, as opposed to superiority of one race based on skin color." I had previously found a discontinued library copy of this book.     <p><span>For individuals all over the world, the Corona or Covid-19 virus has posed many difficulties. The emergence of the virus in December 2019 was centered in the Wet Marketplaces in Wuhan, China. The World Health Organization proclaimed this virus a global epidemic on March 11, 2020. As the epidemic and its manifestations remained undiagnosed and uncertain, the virus spread not just in neighboring countries but internationally, creating various constraints and difficulties in people's everyday lives. With more than 8,06 million reported cases worldwide and more than 3,65 lakh patients in India, the uproar of this virus will not end soon or until no vaccine is found for diagnosis at any time. For the individuals brought to hospitals for the COVID-19 scans and treatment, this has also raised a number of difficulties. The overcharging from the patients who are fighting against the Corona by private hospitals for research, treatment and PPE protection kits to stop its transmission was the most significant challenge that required immediate attention. Aging population and ill patients around the globe have been causing havoc with the current pandemic. In designated hospitals in Indore Region, the rush for COVID-19 patients was very strong and sometimes impossible to manage. The cyclical spike in cases of infection offers little to no space for hospital administrators to establish successful treatment-pricing SOPs. The epidemic did not effectively enforce the directives issued by the administration.     Waste-to-Energy as a Method of Refuse Disposal: An Analysis of Sustainable Technologies and Their Environmental Impact

    Nowadays, waste-to-energy (WtE) is considered the most effective disposal system for nonrecyclable materials. There is general concern with regard to potential environmental and health risks connected with WtE plants. Data from epidemiological studies, conducted since the 1980s, had shown that old-generation plants have a negative environmental impact. Owing to innovations in both technology and methodology involved in waste processing, new-generation WtE plants have considerably mitigated such negative impact. The aim of the study is to assess how legislation, technological innovation, and mitigation strategies can help and reduce the repercussions that waste-to-energy might have on both the environment and people’s health. The first step in this study consisted in investigating the current European and Italian legislation, as well as the operating mechanisms and technologies available to mitigate environmental impacts. Successively, the second step consisted in verifying their actual effectiveness when applied to the most recent plants. In particular, ten modern incinerations, new or restored in the last decade, were examined in detail. The case studies’ analysis and their comparison showed that, when adequately submitted to a health impact assessment (HIA) and in presence of valid monitoring plans, the most advanced facilities are able to keep emissions way below the limits prescribed by the law, mitigating environmental and health impacts. In addition, the study highlighted some areas of possible future interventions and actions. Finally, the outcome of the research is to propose a set of guidelines, legal tools, and appropriate technologies, as exportable and scalable healthy strategies in several contexts

    The small-scale approach in wastewater treatment

    The wastewater treatment is an essential practice and is one of the forefront new worldwide challenges. Indeed, the today aims in wastewater management are profoundly changing. The sanitization processes are of course essential, but now some more efforts are required not only to reduce the environmental impact of the effluent regarding the receiving water body, for example in terms of organic load, but also to provide management practices to allow water recovery, recycle and reuse. This means that treated wastewater are new resources that may be reused according to their final level of quality. New technological facilities are available about, some more are expected in the near future, and innovative scaling approaches are strongly encouraged too. Actually, sometimes extensive sewage collection systems are no more cost-effective, thus decentralisation treatment processes considering small-scale wastewater treatment plants may represent a potential solution in both developed and developing countries. Furthermore, this kind of approach will strengthen those areas that are historically affected by drought phenomena or are expected to suffer from water scarcities in the near future as a consequence of climate change

    A nanny model for intrinsically disordered proteins

    Proteins without a well-defined tertiary structure are intrinsically unstable and are prone to degradation by the 20S proteasome. In my thesis, I investigate the protection mechanisms of intrinsically disordered (ID) protein regions via protein interactions using the AP-1 complex as a model system. AP-1 is composed of c-Fos and c-Jun proteins, out of which c-Fos has a shorter half-life than c-Jun. Interactions by c-Jun were shown to prolong the lifetime of c-Fos, leading to the proposal of the nanny model. This mechanism, where weak protein interactions protect unstructured regions without an induced folding, however, has never been probed directly. Here I investigate the nature of the interactions of c-Fos with c-Jun and how changes in disordered regions contribute to changes in half-life. I use mutational analysis to provide insight into changes in degradation rate as a function of the binding affinity in the bound form with c-Jun. I designed five mutants at the structured regions of c-Fos affecting specific contact sites (L165V, L172V) or charge separation (E175D, E189D, K190R) with c-Jun of which both modulate c-Fos turnover, proportionally to their impact on binding affinity. Interestingly, removal of the disordered region in the complex beyond the structured domain is observed to decrease c-Fos half-life indicating their role in the stability of the complex. The finding suggests that the protein turnover by the 20S proteasome can be fine-tuned by both structured and unstructured regions between c-Fos and c-Jun, consistent with the proposed 'nanny' model. These results highlight a novel aspect of disordered regions present in the bound form (fuzziness) in regulating protein half-life via fine-tuning the association rates between the two proteins. First, it demonstrates that the protection of disordered regions from degradation could be achieved without inducing a stable structure as confirmed by ECD spectroscopy. Binding to a partner generates a fuzzy complex, where fuzzy regions in protein complexes can serve as a nonspecific transient anchor. Second, the protection of disordered regions can be achieved with many binding configurations in the bound state without decreasing the conformational entropy. Thus, the protective role of fuzzy interactions from the 20S proteasome could also provide a possible explanation for how low-complexity sequence motifs involved in higher-order protein structures might serve as selective inhibitors of proteolysis.d
