A comprehensive review of synthetic strategies and SAR studies for the discovery of PfDHODH inhibitors as antimalarial agents. Part 1: triazolopyrimidine, isoxazolopyrimidine and pyrrole-based (DSM) compounds
One of the deadliest infectious diseases, malaria, still has a significant impact on global morbidity and mortality. Plasmodium falciparum dihydroorotate dehydrogenase (PfDHODH) catalyzes the fourth step in de novo pyrimidine nucleotide biosynthesis and has been clinically validated as an innovative and promising target for the development of novel targeted antimalarial drugs. PfDHODH inhibitors have the potential to significantly slow down parasite growth at the blood and liver stages. Several PfDHODH inhibitors based on various scaffolds have been explored over the past two decades. Among them, triazolopyrimidines, isoxazolopyrimidines, and pyrrole-based derivatives known as DSM compounds showed tremendous potential as novel antimalarial agents, and one of the triazolopyrimidine-based compounds (DSM265) was able to reach phase IIa clinical trials. DSM compounds were synthesized as PfDHODH inhibitors with various substitutions based on structure-guided medicinal chemistry approaches and further optimised as well. For the first time, this review provides an overview of all the synthetic approaches used for the synthesis, alternative synthetic routes, and novel strategies involving various catalysts and chemical reagents that have been used to synthesize DSM compounds. We have also summarized SAR study of all these PfDHODH inhibitors. In an attempt to assist readers, scientists, and researchers involved in the development of new PfDHODH inhibitors as antimalarials, this review provides accessibility of all synthetic techniques and SAR studies of the most promising triazolopyrimidines, isoxazolopyrimidines, and pyrrole-based PfDHODH inhibitors

Instance Level Affinity-Based Transfer for Unsupervised Domain Adaptation
Domain adaptation deals with training models using large scale labeled data from a specific source domain and then adapting the knowledge to certain target domains that have few or no labels. Many prior works learn domain agnostic feature representations for this purpose using a global distribution alignment objective which does not take into account the finer class specific structure in the source and target domains. We address this issue in our work and propose an instance affinity based criterion for source to target transfer during adaptation, called ILA-DA. We first propose a reliable and efficient method to extract similar and dissimilar samples across source and target, and utilize a multi-sample contrastive loss to drive the domain alignment process. ILA-DA simultaneously accounts for intra-class clustering as well as inter-class separation among the categories, resulting in less noisy classifier boundaries, improved transferability and increased accuracy. We verify the effectiveness of ILA-DA by observing consistent improvements in accuracy over popular domain adaptation approaches on a variety of benchmark datasets and provide insights into the proposed alignment approach
Effect of different surfactants on the efficacy of triclosan against bacillus subtilis
Master of Science (Chemistry)Drugs designed for oral dosage forms have major challenge of their bioavailability. Any drug must be present in the form of an aqueous solution at the site of absorption. Triclosan, an effective antibacterial agent, inhibits an essential enzyme present in most of the bacteria (Fab I) thereby blocking their fatty acid biosynthesis. However, the molecule is poorly soluble in aqueous solutions.
A study was carried out to understand the effect of six different surfactants on the IC50 value of triclosan against Bacillus Subtilis. Four of the six surfactants contained long alkyl or alkoxy chains namely, Tween-80, Tween-20, TritonX-100 and CTAB. The other two surfactants were carbohydrate and steroid based namely γ-cyclodextrin and sodiumdeoxycholate. Tween-80 and Tween-20 enhanced the growth of bacteria in presence of triclosan thereby inhibiting the effect of drug. TritonX-100 marginally inhibited the growth of bacteria around its CMC values there by suggesting it to be the best surfactant among six to dissolve triclosan. All other surfactants themselves inhibited the growth of bacteria and there by masked the effect of triclosan.School of Chemistry and Biochemistry, Thapar University, Patial
Synthesis and Characterization of Benzophenone Based Phenyl Ether Derivatives
Master of Science- ChemistryBenzophenone and diphenyl ether derivatives are important compounds because of their biological activities and materialistic applications. Such compounds have also been isolated from a number of natural products. Individually, both these categories of compounds have found numerous applications in medicinal and material world. Therefore, the work presented here describes synthesis of benzophenone derivatives of diphenyl ethers. Three compounds D, E and F were synthesized using same benzophenone precursor, (2-hydroxy-4-methoxyphenyl)(phenyl) methanone, and 2- bromo-5-nitrobenzaldehyde to get 2-(2-benzoyl-5-methoxyphenoxy)-5-nitrobenzaldehyde (D). Using different concentrations of the same reducing agent (NaBH4) two different products were obtained. In one case only aldehyde was reduced (Compound E) while in other case both aldehyde and ketone present in the molecule (Compound F) were reduced. Characterization was done using 1H and 13C NMR spectroscopy

Ultra Wideband Wearable Sensors for Motion Tracking Applications
The increasing interest and advancements in wearable electronics, biomedical applications and digital signal processing techniques have led to the unceasing progress and research in novel implementations of wireless communications technology. Human motion tracking and localisation are some of the numerous promising applications that have emerged from this interest. Ultra-wideband (UWB) technology is particularly seen as a very attractive solution for microwave-based localisation due to the fine time resolution capabilities of the UWB pulses. However, to prove the viability of utilizing UWB technology for high precision localisation applications, a considerable amount of research work is still needed. The impact of the presence of the human body on localisation accuracy needs to be investigated. In addition, for guaranteeing accurate data retrieval in an impulse-radio based system, the study of pulse distortion becomes indispensable. The objective of the research work presented in this thesis is to study and carry out experimental investigations to formulate new techniques for the development of an Impulse-radio UWB sensor based localisation system for human motion tracking applications. This research work initiates a new approach for human motion tracking by making use of pulsed UWB technology which will allow the development of advanced tracking solutions with the capacity to meet the needs of professional users.
Extensive experimental studies involving several ranging and three dimensional localisation investigations have been undertaken, and the potential of achieving high precision localisation using ultra-wideband technology has been demonstrated. Making use of the upper portion of the UWB band, a novel miniature antenna designed for integration in the UWB localisation system is presented and its performance has been examined. The key findings and contributions of this research work include UWB antenna characterisation for pulse based transmission, evaluation of comprehensive antenna fidelity patterns, impact of pulse fidelity on the communication performance of a UWB radio system, along with studies regarding the effect of the human body on received pulse quality and localisation accuracy. In addition, an innovative approach of making use of antenna phase centre information for improving the localisation accuracy has been presented
