    Green Energy Ansatz für die anaerobe Behandlung und Analyse von HTC-Prozesswasser

    Die vorliegende Arbeit befasst sich mit der innovativen Nutzung von Aufbereitungswasser aus der hydrothermalen Karbonisierung (HTC) als Quelle für die Erzeugung von Biogas, einem Schlüsselelement im Bereich der erneuerbaren Energien. Durch den Einsatz fortschrittlicher Analysemethoden, darunter Gaschromatographie-Massenspektrometrie (GC-MS) und Hochleistungsflüssigkeitschromatographie (HPLC), sowie durch Batch-Experimente, Hemmstoffanalysen und weitere Versuchsreihen, wurde die Auswirkungen des HTC-Prozesswassers (HTC-PW) als Substrat in anaeroben Vergärungsprozessen zur Biogasgewinnung erforscht. In der Untersuchung kamen zwei Arten von Bioreaktoren zum Einsatz: ein kontinuierlicher Rührkesselreaktor und ein Festbettreaktor, die beide über einen Zeitraum von rund 180 Tagen auf ihre Eignung für die Verarbeitung von HTC-PW hin untersucht wurden. Die Studie offenbart eine deutliche Variabilität in der Zusammensetzung der HTC-PW-Proben, die maßgeblich von den eingesetzten Rohstoffen und den spezifischen Bedingungen des HTC-Prozesses abhängt. Insbesondere die hohe Präsenz von kurzkettigen Fettsäuren und stickstoffhaltigen Verbindungen in HTC-PW stellt sowohl eine Chance als auch eine Herausforderung für die Biogasproduktion dar, wobei insbesondere aromatische Verbindungen wie Phenole und Benzolderivate potenziell hemmende Effekte auf die mikrobielle Aktivität haben können. Die Erzeugung von Biogas aus HTC-PW zeigte vielversprechende Methanausbeuten und unterstreicht das Potenzial zur Energierückgewinnung aus dieser unkonventionellen Abwasserfraktion. Dennoch stehen einer großtechnischen Umsetzung noch Herausforderungen wie die Prozessoptimierung, die Variabilität der Substrate und der energieintensive HTC-Prozess entgegen. Die Integration der anaeroben Gärung in die Aufbereitung von HTC-PW erscheint als aussichtsreiche Strategie, um die Energiegewinnung zu steigern und gleichzeitig Umweltrisiken, die mit der Einleitung unbehandelter Abwässer verbunden sind, zu minimieren. Diese Arbeit betont die Notwendigkeit weiterführender Forschungs- und Entwicklungsarbeiten zur Optimierung der Nutzung von HTC-PW für die Biogasproduktion und leistet einen Beitrag zur Einbindung in Modelle der Kreislaufwirtschaft sowie zur globalen Energiewende hin zu erneuerbaren Energien und Ressourcenrückgewinnung.This research investigates the use of process water from hydrothermal carbonization (HTC PW) as a potential source for producing biogas, a renewable energy. Through a series of gas chromatography-mass spectrometry (GCMS) analyses, high performance liquid chromatography (HPLC) analyses, batch trials, inhibition studies, and other trials, this work explores the impact of HTC PW as a substrate for anaerobic digestion to generate biogas. Two reactor types, a continuous stirred tank reactor and a packed bed reactor, were tested for around 180 days to assess the long-term suitability of HTC-PW for digestion. The results revealed significant variability in the composition and properties of HTC-PW samples, influenced by feedstock characteristics and HTC process parameters. While HTC-PW exhibited high levels of short-chain fatty acids and nitrogenous compounds, its suitability for biogas production varied depending on the specific components present. Notably, GC-MS analysis identified aromatic compounds such as phenols and benzene derivatives, which can exert inhibitory effects on microbial activity in AD systems. Biogas fermentation trials using HTC-PW substrates demonstrated promising methane yields, indicating the potential for energy recovery from this unconventional wastewater fraction. However, challenges such as process optimization, substrate variability, and energy-intensive HTC processing remain significant hurdles to overcome for large-scale implementation. Integration of AD into the HTC-PW treatment process emerges as a promising strategy to enhance energy recovery while reducing environmental risks associated with untreated wastewater discharge. This study underscores the importance of further research and development efforts to optimize HTC-PW utilization for biogas production, paving the way for its integration into circular economy models and contributing to the global transition towards renewable energy and resource recovery

    Aufreinigung von rekombinanten Protein aus Escherichia coli Homogenaten

    The term Quality by Design defines a concept for processing biologics in pharmaceutical biotechnology. In short it means that quality is built into the product rather than prove it afterwards. Knowing the product and even more a fully understanding of the process is mandatory. This work helps to gain process knowledgment of industrial downstream processing of E.coli homogenates. Escherichsa coli (E. coli) is a well-known microorganism, easy to manipulate and popular in biotechnology processes. The fermentation process is fast, cheap and simple. Due to the lack of secretion properties isolation and purification of recombinant proteins is challenging. To characterize the unit operations and their mutual influence, the E. coli strain HMS 174 (DE3) with the plasmid pET11a-GFPmut3.1 was used to establish a downstream model process. gfp was chosen because of its easy quantification. The recently established BIO INDUSTRIAL PILOT PLANT provided the possibility to work on a pilot scale level. First part of the investigation was the homogenization and its effectiveness in dependence of cell density, number of passages and operating pressure. Over the whole range of operating pressure (20MPa to 90MPa) cell density had a negligible effect on the breakage of the cells. Above 60MPa over 90% protein release was achieved with one passage. The disruption efficiency was highly dependent on operating pressure. It was also possibly to establish a correlation between solid content in the solution and its viscosity. It provides a useful estimate of solid content with a short measurement. This is particularly useful for design of centrifuge separations. Furthermore, the influence of the homogenization on the centrifugation behavior was studied. It could be shown that, the higher the operating pressure and the more passages had been performed the clearance efficiency was dramatically decreasing. Another important point is the reduction factor of the pellet volume. At high cell density the pellet volume reduction was lower, leading to more discharges when the homogenate was clarified in a disc stack centrifuge. In turn, this led to significant product losses because 0.4 L of liquid is discharged during every ejection. The chromatographic capture step using anion exchange chromatography was investigated with the resins CaptoQ and Q-Sepharose FF in detail for crude and diafiltrated homogenate. Adsorption isotherms revealed a displacement effect of gfp by other components at high feed concentrations. The displacement can be modeled by the extended Langmuir isotherm, which takes into account the variable qi. It represents the available binding sites for each component and it is suggested to be in correlation with their molecular size. Additionally, was investigated the adsorption kinetics of the diafiltrated and crude homogenate. Kinetic measurements showed that an overshoot above equilibrium capacity took place. The higher the feed concentration the more pronounced was the overshoot. According to these data, it is suggested that a low residence time and a high feed concentration is required to obtain the maximum binding capacity. In general, CaptoQ had a higher binding capacity than Q-Sepharose FF. Also diafiltrating the homogenate enhanced binding capacity. Furthermore break through curves (BTC) were performed with both resins. Remarkably, changes in the residence time had no effect on the dynamic binding capacity (DBC) for the crude homogenate. Additionally, a higher DBC was exhibited at a higher feed concentration. However, the recovery in the elution step was quite low due to the fact that a high amount of product was lost during the wash out phase. BTC's with the diafiltrated revealed a higher binding capacity which was positively influenced by enhancing the residence time and lowering the feed concentration

    The ascomycete Trichoderma reesei is an industrial producer of technical enzymes, especially for cellulases and hemicellulases. As such it is an interesting target for genetic engineering and optimisation as microbial production host. The development demands a growing genetic toolbox to optimise product yields and process parameters. The first approach was the design of an artificial promoter. The promoter of the uncharacterized gene cdna1 (Pcdna1) is one of the strongest in T. reesei active during growth on glucose. Although stronger promoters exist, they are repressed by glucose or require activation. In order to design a synthetic core promoter with defined characteristics, high strength and no repression effects on glucose, we analysed 50 highly expressed genes towards their core promoter structure. A box structure system was established, a box is a conserved 5-45 bp long sequence within the promoter. Two sequences based on native core promoters and one completely de novo designed one were used as basis to arrange the boxes, ultimately generating 200 bp long artificial core promoters, controlling the expression of the reporter gene cellulase CEL12A. The design of the artificial core promoters included known sequences like TATA-boxes, GC-boxes or CT-rich regions, but also newly found boxes from the sequence analysis. Two of the synthetic core promoters surpassed the expression strength of the native reference core promoter of cdna1 by about 175%. To gain a full-length promotor, these two superior synthetic core promoters were elongated with different distal promoter regions. Surprisingly, all combinations resulted in different activity values, which suggests that the core promoters were not effective with each of the distal promoters. The results form the basis for a deeper analysis or the relation of different promoter elements. The second object of interest was the morphology of the fungus. As most of the energy consumption is used to generate biomass, it is interesting for industrial processes to develop a system in which the growth of T. reesei can be regulated. Aim of this study was to optimise product output by limitation of growth during fermentation by downregulation of specific chitin synthases and one glucan synthase. Therefore, the native promoters of chs1, chs2, chs3, chs6 and fks1 were replaced by the L-methionine repressible promoter of tauD1. Transcript levels showed that the repressible promoter of tauD1 was effectively repressed by the addition of 0.1 mM L-methionine, but the addition of the amino acid also led to an increase of biomass formation and a decline in specific cellulase activity. The overexpression of 1,3-β-glucan synthase FKS1, caused by the tauD1 promoter in an unrepressed stage, led to a compact pellet growth and less biomass formation in liquid culture in QM9414 based strains and in the industrial ancestor strain RUT-C30. Interestingly, the altered phenotype was accompanied by an increased cellulase activity in the supernatant in QM9414 strains but led to a complete abolishment of cellulase activity in RUT-C30. The results of the study opened a new opportunity for morphology engineering of T. reesei and model for similar experiments in other filamentous fungi. Another approach to further increase protein production was the introduction of hyperbranching. Since most of exocytosis is located at the hyphal apical tip, more branches could result in more protein secretion. RAC plays a crucial role in actin dynamics and is involved in polarisation of the cell during germination and apical extension of the hyphae. Rac1 deletion caused a hyperbranching phenotype with dichotomous branching, shorter total hyphae length and a larger hyphal diameter. While extracellular cellulase activities remained at parental strain levels on D-glucose and cellulose, the specific activity on lactose cultures was increased up to three times at 72 hours accompanied by an upregulation of transcription of the main cellulases. Although the morphology of the Δrac1 strains was considerably altered, the viscosity of the culture broth in a fed-batch fermentation was not significantly different in comparison to the parental strain. Our study demonstrates that enhanced protein secretion is not per se related to the number of tips. The thesis contributes to strain improvement of T. reesei and broadened our understanding in the experimental field of synthetic biology and morphology engineering.Der Ascomycet Trichoderma reesei ist ein in der Industrie etablierter Mikroorganismus für die Produktion von technischen Enzymen, besonders von Cellulasen und Hemicellulasen. Als mikrobieller Produktionswirt ist T. reesei ein interessantes Studienobjekt zur genetischen Stammverbesserung, um seine Proteinausbeute und Fermentationseigenschaften zu optimieren. Ein Ansatz zur Stammverbesserung ist die Entwicklung synthetischer Promotoren. Der Promotor des uncharakterisierten Gens cdna1 ist einer der stärksten in T. reesei während dem Wachstum auf Glucose. Obwohl prinzipiell stärkere Promotoren existieren, sind diese auf Glucose entweder reprimiert oder benötigen eine Aktivierung. Um einen synthetischen Kernpromoter mit definierten Eigenschaften designen zu können, nämlich hohe Expressionsstärke und keine Reprimierung bei Anzucht auf Glukose, wurden 50 hoch exprimierte Gene bezüglich ihrer Kernpromoterstruktur untersucht. Aus dieser Analyse wurde ein Boxensystem abgeleitet, wobei eine Box eine konservierte Sequenz von 5 bis 45 Basenpaaren darstellt. Die Boxen wurden auf zwei natürlichen und einer zuvor völlig de novo designten synthetischen Basissequenzen arrangiert, um so 200 bp lange Kernpromotoren zu formen, die die Expression der Reporterzellulase CEL12A kontrollieren. Das synthetische Design umfasste bekannte Motive wie TATA-Boxen, GC-Boxen oder CT-reiche Regionen, aber auch in dieser Studie neu gefundene Sequenzen. Zwei dieser synthetischen Konstrukte überstiegen die Aktivität des Referenzpromotors von cnda1 deutlich, um 175 %. Um einen Promotor mit voller Länge zu generieren, wurden diese zwei Kernpromotoren mit verschiedenen distalen Promotorteilen verlängert. Alle diese Kombinationen führten zu unterschiedlichen Aktivitäten, was darauf hindeutet, dass die Kernpromotoren nicht effektiv mit jedem beliebigen distalen Promotorteilen kombiniert werden kann. Diese Ergebnisse bilden eine solide Basis für weitere Analysen zum komplexen Zusammenspiel verschiedener Promotorteile. Der zweite Teil der Arbeit behandelte die Optimierung der Morphologie des Pilzes. Da viel Energie in den Aufbau von Biomasse fließt, ist es für industrielle Prozesse interessant ein System zu entwickeln, mit dem das Wachstum von T. reesei genetisch reguliert werden kann. Um eine Optimierung der Produktmenge durch Wachstumslimitierung während einer Fermentation zu erreichen, wurden ausgewählte Chitinsynthasen und einer Glucansynthase unter einen regulierbaren Promotor gestellt. Dazu wurden die natürlichen Promotoren der chs1, chs2, chs3, chs6 und fks1 Gene durch einen mit L-Methionin reprimierbaren Promotor des Gens tauD1 ersetzt. Die Transkriptdaten zeigten eine effektive Reprimierung bei Zugabe von 0,1 mM L-Methionin, allerdings führte der Zusatz der Aminosäure zu einer erhöhten Biomassebildung und einem Abfall der Zellulaseaktivität im Überstand. Die durch den tauD1-Promoter erzeugte Überexpression der β-1,3-Glucansynthase fks1 führte im unreprimierten Zustand zur einem kompakten Pelletwachstum und einer verminderten Biomassebildung in Flüssigkulturen, sowohl in QM9414 als auch im Industriestamm RUT-C30. Interessanterweise war dieser Phänotyp in QM9414 von einer erhöhten Zellulaseaktivität begleitet, führte bei RUT-C30 jedoch zu einer vollständigen Reduktion ebendieser. Die Ergebnisse dieser Studie eröffneten eine neue Möglichkeit der Morphologieoptimierung in T. reesei und kann als Model für ähnliche Studien in anderen filamentösen Pilzen verwendet werden. Eine weitere Möglichkeit um die Protein Produktion zu steuern ist das Erzeugen eines Hyperbranchers. Da die Hyphenspitze viel Proteine sekretiert, könnten mehr Verzweigungen mehr Proteinsekretion bedeuten. Die kleine GTPase RAC spielt eine wichtige Rolle in der Actindynamik und ist auch in die Polarisierung der Zelle während der Sporulation und dem vegetativen Wachstum involviert. Die Deletion von rac1 in T. reesei führte zu einem Hyperbrancher-Phänotyp mit dichotomen Verzweigen, kürzerem Hyphen und dickeren Hyphendurchschnitten. Die Cellulaseproduktion auf Glukose und Zellulose der rac1 deletierten Stämme entsprach der des Ausgangsstammes, doch auf Laktose konnte nach 72 Stunden ein Anstieg in der Aktivität und der Geamtproteinmenge auf das dreifache Niveau beobachtet werden zusammen mit einem Anstieg der Transkriptlevel der Hauptzellulase. Trotz der starken Morphologieveränderung hat sich die Viskosität der Deletionsstämme im Fed-Batch Reaktor im Vergleich zum Referenzstamm nicht signifikant verändert. Die Studie hat gezeigt, dass die Menge an sekretiertem Protein nicht direkt mit der Anzahl der Hyphenspitzen zusammenhängt. Diese Arbeit hat insgesamt zu einem besseren Verständnis verschiedener Möglichkeiten zur Stammverbesserung von T. reesei beigetragen, sowie das Wissen im Bereich der synthetischen Biologie und Morphologieoptimierung erhöht

    Pantothensäureabbau in Trichoderma reesei

    Pantothensäure bzw Vitamin B5 ist ein wichtiger Precursor für die Biosynthese von Coenzym A (CoA), das als Acylgruppenträger fungiert und u.a. am Auf- und Abbau von Kohlenhydraten und Fetten beteiligt ist. Im Gegensatz zum Aufbau der Pantothensäure ist über deren Abbau wenig bekannt. Im T. reesei Genom sind die für den Pantothensäureabbau notwendigen Gene in einem Cluster zusammengefasst, der auch noch einen Zn(II)2Cys6 Transkriptionsfaktor PAA1 und eine Permease PAP1 umfasst. Um den Einfluss des Transkriptionsfaktors PAA1 auf die Expression des Pantothensäureclusters aufzuzeigen, wurden die Expression der anderen 5 Gene in einem Stamm, der im Regulator deletiert ist, untersucht. Es zeigte sich, dass die Induktion dieser Gene durch Pantothensäure in Abwesenheit des PAA1 Regulators verloren geht, aber in einem Stamm in dem der intakte Regulator wieder eingeführt wurde, wiederhergestellt werden konnte. Die Funktion der Permease PAP1 wurde mittels einer Komplementationsassays der Saccharomyces cerevisiae Pantothensäure-Permease-Mutante fen2 untersucht. Die Einbringung des T. reesei paa1 in die fen2 Mutante, führte zur Aufhebung des Wachstumsdefektes des fen2 Stammes und bestätigte somit die Funktion der PAP1 als Pantothensäurepermease. Weiters wurden die 2 Enzyme der ersten beiden Schritte des Pantothensäureabbaus in E. coli exprimiert und über einen His Tag gereinigt. Die Pantothenase PAN1, welche für den Abbau der Pantothensäure zu Pantoat und ß-Alanin verantwortlich ist, wurde in Inclusion Bodies produziert und konnte erst nach Optimierung der Wachtsumsbedingungen in löslichen Zustand produziert werden. Die Pantoat 4-dehydrogenase PAN2, welche die Oxidation von Pantoat zu R-4-dehydropantoat katalysiert, hat ein Molekulargewicht von 27 kDa, KM(Pantoate) von 19,51 mM und eine vmax von 3,33 U/mg. In der vorliegenden Arbeit wurden somit weitere Bestandteile des Pantothensäureabbaus in T. reesei funktionell charakterisiert.Pantothenic acid (Vitamin B5) is essential for life as a precursor for coenzyme A (CoA) biosynthesis. CoA functions as an acyl group carrier required for carbohydrate and fat metabolism. The biosynthesis pathway of pantothenate is already well documented in many bacteria, fungi and plants whereas the pantothenate catabolism is largely unknown. Genes involved in pantothenate catabolism are clustered in the Trichoderma reesei genome and include beside four metabolic enzyme encoding genes, a putative pantothenate permease PAP1 and a Zn(II)2Cys6 transcription factor PAA1. In this thesis the effect of the deletion of paa1 on the expression of the other genes of the cluster was studied. It was shown that in the absence of paa1 no induction could be observed. Induction of the genes was, however, restored in strains in which paa1 was reintroduced. The function of the putative pantothenate permease PAP1 was tested in a complementation assay. Therefore, the T. reesei pap1 was introduced into the Saccharomyces cerevisiae pantothenate permease mutant fen2. In these strains the growth defect of the fen2 mutants was abolished which showed that pap1 indeed encodes a pantothenate permease. The first two enzymes of pantothenate catabolism were expressed in E. coli and purified as His-Tag proteins. Pantothenase PAN1, which is responsible for the degradation of pantothenic acid to pantoate and ß-alanine, was mainly produced in inclusion bodies and could only be produced in soluble form after optimization of the growth conditions. Pantoat 4-dehydrogenase PAN2 which oxidizes pantoate to R-4-dehydropantoate, was produced in soluble form with a Mw of 27 kDa, KM (pantoate) of 19,51 mM and a vmax of 3,33 U/mg.     Neue Spieler im Biomasseabbau in Trichoderma reesei

    The ability of the filamentous ascomycete Trichoderma reesei to efficiently degrade lignocellulose was already discovered over 70 years ago. Since then, research on the organism was mainly propelled by the vision, to use its secreted enzymes, to generate biofuels or other products from plant waste material as a sustainable alternative to refined fossil oil. However, currently available enzyme formulations are still not cost efficient and research efforts are ongoing to develop new ways to make these biorefinery concepts economically more efficient. Technological advances including -omics technologies, have led to the identification of several potential novel players in biomass degradation. But the presence of ¿omics data is only powerful, if genes and proteins identified by these technologies are already described. However, this is a clear bottleneck as, in contrast to long-serving ¿old bugs¿ like S. cerevisiae and E. coli, filamentous fungi such as T. reesei still have a plethora of genes with unknown function. To unravel this dormant potential, the study of single genes and their encoding proteins plays a key role in the establishment of novel players in lignocellulose

    Eine neue Rolle für Toll-like-Rezeptoren in Hepatozyten bei der Kontrolle der Eisenrestriktion

    Iron homeostasis is crucial for human health. During infection, iron is restricted as a host-defense mechanism, resulting in the development of hypoferremia and, after prolonged iron restriction, to anemia of inflammation. Pathogens are recognized by macrophages which leads to cytokine secretion, such as interleukin (IL)–6 and IL–1β. Consequently, hepatocytes are activated and produce the key iron regulator peptide hepcidin. Hepcidin promotes the internalization and degradation of the only known iron exporter ferroportin resulting in decreased serum iron levels. However, whether Toll-like receptors (TLRs) expressed on hepatocytes directly recognize pathogen-associated molecular patterns (PAMPs) and contribute to hepcidin upregulation has not been resolved. To answer this question, primary murine hepatocytes were stimulated with TLR1-9 ligands for 4 h. Hepatocytes responded with an upregulation of pro-inflammatory genes and secreted cytokines. Stimulation with the TLR5 ligand flagellin and TLR2/6 ligand FSL1 could specifically upregulate hepcidin expression in hepatocytes. Early FSL1-mediated hepcidin upregulation was independent of autocrine upregulation by IL-1β and IL-6 and was further shown to be independent of macrophage secreted cytokines in a macrophage:hepatocyte co-culture. Hepcidin expression in response to FSL1, flagellin, heat inactivated Staphylococcus aureus and Brucella abortus was mediated by TLR2/6 and the downstream mediators c–Jun-N-terminal kinases (JNK) and extracellular–signal regulated kinases (ERK). An ex vivo perfusion experiment demonstrated that bacteria could pass the liver sinusoidal endothelial cell barrier and stimulate hepcidin upregulation in hepatocytes. This study highlights hepatocytes as active immunological players and provides insight into novel regulators of hepcidin, and consequently of iron homeostasis.Die Eisenhomöostase spielt eine entscheidende Rolle für die menschliche Gesundheit. Im Verlauf einer Infektion wird durch einen angeborenen Abwehrmechanismus die verfügbare Serumeisenkonzentration des Wirts vermindert, was bei längerer Entzündungsdauer zur Entwicklung einer entzündungsbedingten Anämie führt. Pathogene werden durch Makrophagen erkannt, die Zytokinen wie Interleukin (IL)-6 und IL-1β ausschütten. Infolgedessen werden Hepatozyten aktiviert und produzieren das eisenregulierende Peptidhormon Hepcidin, welches den Abbau des einzigen bekannten Eisenexporters, Ferroportin einleitet. Dies führt zu einem verringertem Serumeisenspiegel. Ob Hepatozyten mit Hilfe ihrer Toll-like Rezeptoren (TLRs) Pathogene erkennen und infolgedessen zur Hepcidinantwort beitragen können ist bisher ungelöst.Um diese Frage zu beantworten, wurden primäre Hepatozyten der Maus für 4 Stunden mit TLR1-9-Liganden stimuliert. Sie reagierten mit einer erhöhten Expression von proinflammatorischen Genen und sekretierten Zytokine. Die Stimulation mit dem TLR5-Liganden FLA-ST und dem TLR2/6-Liganden FSL1 erhöhte die spezifische Expression von Hepcidin in Hepatozyten. Die anfängliche Hepcidinexpression als Antwort auf FSL1 ist unabhängig von autokriner Regulierung mit IL-1β und IL-6. Ein Makrophagen:Hepatozyten Ko-Kultursystem zeigte weiters, dass die frühe Hepcidinantwort nicht von Makrophagen und deren Zytokinen abhängt. Weiters wurde gezeigt, dass die Hepcidinantwort auf FSL1, Flagellin, sowie auf hitzeinaktivierte Staphylococcus aureus und Brucella abortus von der Aktivierung von TLR2/6 und der Signaltransduktionsmoleküle c-Jun-N-terminale Kinasen (JNK) und extrazellulär-signalregulierte Kinasen (ERK) abhängig ist. Ein ex vivo Perfusionsversuch zeigte, dass Bakterien die Barriere der sinusoidalen Endothelzellen der Leber passieren und die Expression von Hepcidin in Hepatozyten stimulieren können.     Die Charakterisierung einer neuen PEGylierten FVIII Präparation auf potentiell immunogene Eigenschaften

    Hämophilie A ist eine erblich bedingte Blutgerinnungsstörung, bei der es durch ein Fehlen von funktionellem Gerinnungsfaktor VIII (FVIII) zu einem Defekt in der Koagulationskaskade kommt. Die derzeitige Therapie besteht in der Substitution des fehlenden Gerinnungsfaktors mit plasmatischen oder rekombinanten FVIII Konzentraten. BAX 855 ist ein chemisch modifiziertes FVIII Präparat, das durch eine verlängerte Halbwertszeit eine reduzierte Behandlungsfrequenz ermöglicht. Eine der schwersten Komplikationen im Patienten ist die Entstehung von neutralisierenden Antikörpern gegen FVIII, welche die Therapie unwirksam machen. Da der zugrundeliegende Mechanismus noch nicht entschlüsselt worden ist, muss BAX 855, wie alle neuen FVIII Präparate, im Zuge der präklinischen und klinischen Entwicklung ausführlich auf die Immunogenität getestet werden. In der vorliegenden Thesis wurden verschieden Aspekte zur möglichen Induktion einer Immunantwort von BAX 855 im Patienten beleuchtet. Der erste Teil dieser Arbeit beschäftigt sich mit der potentiellen Immunogenität von Proteinaggregaten. Dabei wurden die Quantität und Qualität dieser Proteinaggregate von BAX 855 und ADVATETM mittels einer neu etablierten durchflusszytometrischen Methode verglichen. ADVATETM ist das Basismolekül für BAX 855 mit einer langjährigen, therapeutischen Sicherheit. In unserer Studie war kein Unterschied zwischen beiden Präparaten festzustellen. Dadurch ist zu erwarten, dass ein möglicher Einfluss von Proteinaggregaten auf die Immunogenität für BAX 855 und ADVATETM vergleichbar groß ist. Der zweite Teil dieser Arbeit beschäftigt sich mit der potentiellen Immunogenität von Polyethylenglykol (PEG), einem Bestandteil von BAX 855. Unsere Resultate zeigen, dass ein Teil der gesunden Bevölkerung IgG und IgM Antikörper entwickeln, die aber keine offensichtliche Symptomatik verursachen. Außerdem deuten unsere Daten darauf hin, dass PEG an ein immunogenes Protein gekoppelt sein muss, um eine Immunantwort auszulösen. Daher würde man in jenen Patienten, die mit BAX 855 behandelt werden, die Entstehung von PEG Antikörpern erwarten, die wiederum eine Immunantwort gegen FVIII entwickeln. Unsere Erfahrungen mit PEG Antikörpern in gesunden Spendern sowie weiterführende Studien, die gezeigt haben, dass diese Antikörper nicht mit endogenen Antigenen kreuzreagieren, weisen aber auf ein vernachlässigbares Risiko hin. Zusammenfassend zeigen die gefundenen Daten, dass für BAX 855 ein vergleichbares Sicherheitsprofil wie für ADVATETM im Patienten zu erwarten ist.Hemophilia A is a congenital bleeding disorder characterized by the lack or insufficient amount of functional factor VIII (FVIII) leading to a defect in the coagulation cascade. Patients are currently treated with a replacement therapy using plasma-derived or recombinant FVIII. BAX 855 is a novel PEGylated FVIII preparation with a prolonged half-life of FVIII in the circulation of patients. The formation of neutralizing antibodies against FVIII, so called FVIII inhibitors, remains the major challenge in current hemophilia A care. Why some patients develop FVIII inhibitors while others do not is far from being understood. Therefore it is important that BAX 855, like all novel FVIII preparations, need to be tested for its immunogenic potential during non-clinical and clinical development. In the present thesis we established suitable technologies to address different aspects of the potential of BAX 855 to induce unwanted immune responses and to analyze different components of the immunogenicity of BAX 855. In the first part we focused on sub-visible particles, which were recently linked to enhance the immunogenicity of protein therapeutics. We established a flow-cytometry-based approach for the analysis of sub-visible particles and compared the content and quality of sub-visible particles within BAX 855 and ADVATETM, the base molecule of BAX 855 with a proven safety record. In our study we could not identify any differences between both molecules and concluded that the potential impact of sub-visible particles on the immunogenicity is expected to be similar for both, BAX 855 and ADVATETM. In the second part we investigated the potential antibody responses against polyethylene glycol (PEG), which is a major compound of BAX 855, and the possible mechanisms on their induction. Our study indicates a development of IgM and IgG antibodies against PEG in healthy individuals without any pathology. The induction of anti-PEG antibodies seems to require the attachment of PEG to immunogenic peptides or proteins. Therefore, we expect that patients who recognize the protein part of BAX 855 as immunogenic protein are at risk to develop antibodies against PEG. Our experience with anti-PEG antibodies in healthy individuals and the negative outcome of a tissue crossreactivity study however indicate that these anti-PEG antibodies are unlikely to be of any safety concern. Based on the findings in this thesis, BAX 855 can be expected to have a safety profile in patients similar to that of ADVATETM

    Enhancement of anti-tumor immune reactivity by targeting signaling checkpoint blockade molecules

    Krebsimmuntherapie stellt ein wichtiges Werkzeug zur Bekämpfung verschiedener Krebsarten dar. Durch Ausschalten von immunsuppressiven Signalwegen in TZellen, führt Checkpoint Blockade Therapie zu verstärkter Anti-Tumor Immunantwort. Casitas B-lineage proto-oncogene-b (Cbl-b) ist ein wichtiges Protein im Signalweg der T-Zellen. Hierbei handelt es sich um ein intrazelluläres Molekül, welches mehrere Signalwege beeinflusst. Es spielt eine große Rolle bei der T-Zell Aktivierung und somit der Regulierung des Immunsystems. Es wurden 80.000 niedermolekulare Substanzen bezüglich ihrer Auswirkungen auf den Cbl-b Signalweg untersucht. Dabei ist die orale Verabreichung eines Wirkstoffs gegen Krebs das Ziel. Um die Beeinflussung der intrazellulären Signalwege durch die LMW-Substanzen zu untersuchen, wurden in-vitro und in-vivo Studien mit humanen und murinen primären Immunzellen durchgeführt. Des weiteren wurden Kontakt-Assays mit T-Zellen und Tumorzellen ausgeführt, um die Stimulation der T-Zellen physiologischer zu gestalten. Immunologische Mediatoren und Marker wurde quantitativ bestimmt. Die Ergebnisse zeigten gesteigerte T-Zell Aktivierung nach Zugabe der wirksamen Substanzen. Inflammatorische Zytokine und Oberflächenaktivierungsmarker wurden vermehrt exprimiert. Die zugegebenen Substanzen hatten jedoch keine Wirkung auf unstimulierte Zellen oder Tumorzellen. Durch in-vivo Versuche konnten die Ergebnisse bestätigt werden. Compound vermittelte T-Zell Aktivierung steht auch in Zusammenhang mit Tumorzellvernichtung. Cbl-b mRNA Silencing wurde durchgeführt, um zu untersuchen, ob Compounds den Cbl-b Signalweg beeinflussen. Die Wirkungsweise der Compounds und der Cbl-b Deaktivierung sind unterschiedlich. Jedoch wird die Cbl-b Aktivität durch Compound Zugabe verringert, was zu dem Rückschluss führt, dass Compound upstream von Cbl-b in den Signalweg eingreifen. Diese Arbeit bietet Einblicke in Signalwege in T-Zellen, beeinflusst durch Cbl-b und die verwendeten Compounds. Die Wirksamkeit der Compounds in T-Zellen wurde sowohl in-vitro als auch in-vivo gezeigt. Zur Bestimmung des Compound Targets und genauerer Effekte in Tumorgewebe sind weitere Untersuchungen nötig.Immunotherapy of cancer is presenting an important tool for combating various cancer types. Checkpoint blockade therapy leads to enhancement of anti-tumor immune responses in T-cells by removing inhibitory signaling pathways. An important molecule in T-cell signaling is Casitas B-lineage proto-oncogene-b (Cblb). Cbl-b is an intracellular molecule, regulating multiple signaling pathways. It plays a crucial role in establishing the threshold for T-cell activation and therefore maintaining homeostasis of the immune system. 80.000 maximal divers LMW compounds were analyzed concerning their ability to interfere with Cbl-b pathways with the aim of developing an orally available drug against cancer. In vitro studies with primary human and murine immune cells were performed to understand intracellular signaling pathways affected by the compounds. Further, contact assays with T-cells and tumor cells were performed to achieve a more physiologic stimulation. Immunologic mediators and markers were quantitatively analyzed. Results showed enhanced T-cell activation after compound application. The expression of inflammatory cytokines and surface activation markers increased. Compounds have no effect on unstimulated immune cells or on tumor cells only. In vivo studies could confirm these results. Compound mediated T-cell activation correlates with tumor cell destruction. To check whether compounds act over Cbl-b pathways, Cbl-b mRNA was silenced. Results showed that compound mode of action and silencing of Cbl-b expression are not similar. However, compound application reduces Cbl-b activity and leads to the assumption that the compound target is upstream of Cbl-b. This study displays insights in T-cell signaling pathways influenced by Cbl-b and influenced by developed compounds. The efficacy of the compounds in T-cell activation was shown, both in vitro and in vivo. Further research is needed to identify the compound target more precisely and to determine compound effects in tumor tissues
